Filamin-A cross-links actin filaments into dynamic orthogonal networks, and interacts with an array of proteins of diverse cellular functions. Because several filamin-A interaction partners are implicated in signaling of cell mobility regulation, we tested the hypothesis that filamin-A plays a role in cancer metastasis. Using four pairs of filamin-A proficient and deficient isogenic cell lines, we found that filamin-A deficiency in cancer cells significantly reduces their migration and invasion. Using a xenograft tumor model with subcutaneous and intracardiac injections of tumor cells, we found that the filamin-A deficiency causes significant reduction of lung, splenic and systemic metastasis in nude mice. We evaluated the expression of filamin-A in breast cancer tissues by immunohistochemical staining, and found that low levels of filamin-A expression in cancer cells of the tumor tissues are associated with a better distant metastasis-free survival than those with normal levels of filamin-A. These data not only validate filamin-A as a prognostic marker for cancer metastasis, but also suggest that inhibition of filamin-A in cancer cells may reduce metastasis and that filamin-A can be used as a therapeutic target for filamin-A positive cancer. 相似文献

入侵植物三叶鬼针草(Bidens pilosa)对我国农牧业生产造成了重大的损失。本文主要研究三叶鬼针草入侵与不同本地植物竞争对土壤微生物群落结构和土壤养分的影响。利用磷脂脂肪酸方法(phospholipid fatty acids, PLFAs)测定土壤微生物群落组成, 同时测定土壤养分和酶活性, 并利用Canoco4.5软件分析了土壤微生物、土壤养分和土壤酶活性的相关性。结果表明: (1)三叶鬼针草对革兰氏阳性菌、革兰氏阴性菌、丛枝菌根真菌等土壤微生物具有较强的聚集能力, 且其根际土壤聚集的微生物类群与本地植物种类密切相关。(2)三叶鬼针草入侵显著增加了入侵地土壤的有机碳含量, 降低了铵态氮的含量; 土壤中的速效钾、速效磷和硝态氮的含量则与本地植物种类密切相关。(3)相关性分析表明, 16:00和16:1 ω5c对铵态氮的含量影响较大, 而三叶鬼针草入侵地16:00和16:1 ω5c的含量显著高于裸土对照, 进而推测这一状况导致了铵态氮含量的降低。(4) 15:1 anteiso A和18:1 ω5c与速效钾的含量呈显著正相关, 而其含量在狗尾草(Setaria viridis)中显著高于其他处理, 三叶鬼针草与狗尾草混种处理中土壤中速效钾的含量高于其他处理。以上结果说明, 三叶鬼针草通过改变土壤微生物群落结构影响了土壤酶活性和土壤养分, 且这种改变与入侵地本地植物种类有关。 相似文献

Microsatellites are widely distributed throughout nearly all genomes which have been extensively exploited as powerful genetic markers for diverse applications due to their high polymorphisms. Their length variations are involved in gene regulation and implicated in numerous genetic diseases even in cancers. Although much effort has been devoted in microsatellite database construction, the existing microsatellite databases still had some drawbacks, such as limited number of species, unfriendly export format, missing marker development, lack of compound microsatellites and absence of gene annotation, which seriously restricted researchers to perform downstream analysis. In order to overcome the above limitations, we developed PSMD (Pan‐Species Microsatellite Database, http://big.cdu.edu.cn/psmd/ ) as a web‐based database to facilitate researchers to easily identify microsatellites, exploit reliable molecular markers and compare microsatellite distribution pattern on genome‐wide scale. In current release, PSMD comprises 678,106,741 perfect microsatellites and 43,848,943 compound microsatellites from 18,408 organisms, which covered almost all species with available genomic data. In addition to interactive browse interface, PSMD also offers a flexible filter function for users to quickly gain desired microsatellites from large data sets. PSMD allows users to export GFF3 formatted file and CSV formatted statistical file for downstream analysis. We also implemented an online tool for analysing occurrence of microsatellites with user‐defined parameters. Furthermore, Primer3 was embedded to help users to design high‐quality primers with customizable settings. To our knowledge, PSMD is the most extensive resource which is likely to be adopted by scientists engaged in biological, medical, environmental and agricultural research. 相似文献

In order to investigate the mitochondrial genome of Panthera tigris amoyensis, two South China tigers (P25 and P27) were analyzed following 15 cymt-specific primer sets. The entire mtDNA sequence was found to be 16,957 bp and 17,001 bp long for P25 and P27 respectively, and this difference in length between P25 and P27 occurred in the number of tandem repeats in the RS-3 segment of the control region. The structural characteristics of complete P. t. amoyensis mitochondrial genomes were also highly similar to those of P. uncia. Additionally, the rate of point mutation was only 0.3% and a total of 59 variable sites between P25 and P27 were found. Out of the 59 variable sites, 6 were located in 6 different tRNA genes, 6 in the 2 rRNA genes, 7 in non-coding regions (one located between tRNA-Asn and tRNA-Tyr and six in the D-loop), and 40 in 10 protein-coding genes. COI held the largest amount of variable sites (9 sites) and Cytb contained the highest variable rate (0.7%) in the complete sequences. 细胞因子假说是关于抑郁症发病机理的重要假说,为探讨抑郁症的发病机理和临床治疗方法提供了新方向.细胞因子分为前炎性细胞因子和抗炎性细胞因子.前炎性细胞因子与抑郁症的发病密切相关,而抗炎性细胞因子可能具有抗抑郁的作用.本文着重综述抗炎性细胞因子与抑郁症的关系.抗炎性细胞因子如白介素10、白介素1受体拮抗剂、白介素4、白介素13、转化生长因子β和脂联素等,在抑郁症中表达下降;补充外源抗炎性细胞因子则具有一定的抗抑郁作用.抗炎性细胞因子可通过拮抗前炎性细胞因子的作用,并与MAPK信号通路、神经递质和糖皮质激素相互作用而参与到抑郁症中.抗抑郁药能使抗炎性细胞因子的表达上升,这可能是药物起效的机制之一.抗炎策略在抑郁症的治疗中有重要应用前景. 相似文献

通过RACE技术,克隆了云南红豆杉紫杉烷2α-O-苯甲酰转运酶基因(TyTBT),该酶催化2-去苯甲酰-7,13-二乙酰巴卡亭Ⅲ生成7,13-二乙酰巴卡亭Ⅲ,是紫杉醇合成途径中的关键酶之一.TyTBT基因cDNA全长1481 bp,含有1 320 bp的开放读码框,编码440个氨基酸的多肽,分子量为50 050 Da,等电点为6.17.氨基酸序列比对表明TyTBT同植物酰化酶家族的其它成员有较高的相似性,超过67%,同东北红豆杉和曼地亚红豆杉的紫杉烷2α-O-苯甲酰转运酶氨基酸序列的一致性和相似性达到最高,分别为95%和96%.广泛地比对分析证明这种较高的相似性在红豆杉属的其它酶家族中具有普遍性,进化树分析表明同东北红豆杉和曼地亚红豆杉的紫杉烷2α-O-苯甲酰转运酶(TBT)的相似性高于紫杉醇合成途径中的其它酰化酶. 相似文献

基于最大熵模型的不同尺度物种分布概率优化热点分析: 以红色木莲为例 总被引:1,自引:0,他引:1
单一空间尺度构建的最大熵(maximum entropy, MaxEnt)模型是否具有代表性, 是MaxEnt模型应用与发展中面临的重要问题。本研究基于有效的地理分布位点数据, 利用最小凸多边形法(the minimum convex polygon method)在三江并流、云南省及全国3个空间尺度下分别识别了红色木莲(Manglietia insignis)的建模区域, 并进一步建立MaxEnt模型: 使用ROC曲线分析法与遗漏率(omission rate, OR)检验评估MaxEnt模型预测精度; 基于ArcGIS分析分布概率及其热点区域的分布趋势, 并通过分区统计工具Zonal识别潜在适宜分布区域的质心位置; 采用刀切法检验环境因子贡献率。结果表明: (1)不同尺度下红色木莲的MaxEnt模型都有良好的预测效果, 三江并流、云南省及全国尺度下的AUC值分别为0.936、0.887和0.930, OR值分别为0.18、0.15和0.20; (2)各尺度红色木莲的适生区格局呈现一致性分布趋势, 集中在独龙江、怒江和澜沧江3个流域; (3) 3个空间尺度下红色木莲的地理分布受不同环境因子影响, 存在着尺度依赖效应。由此可见, 红色木莲在不同空间尺度下的预测模型有着稳定的性能表现与良好的预测效果。此外, 我们建议在野外实地调查与野生生物资源保护中加强对普通物种的关注, 在预测物种地理分布的研究中将MaxEnt模型与热点分析结合使用。 相似文献

Lung cancer is the leading cause of cancer-related mortality all over the world. In recent years, pulmonary adenocarcinoma has surpassed squamous cell carcinoma in frequency and is the predominant form of lung cancer in many countries. Epidemiological investigations have shown an inverse relationship between garlic (Allium sativum) consumption and death rate from many cancers. Diallyl trisulfide (DATS) is one of the garlic-derived compounds (also known as: organosulfer compounds, OSC). DATS can induce apoptosis and inhibit the growth of many cancer cell lines. Our study demonstrated that the apoptotic incidents induced by DATS were a mitochondria-dependent caspase cascade through a significant decrease of the anti-apoptotic Bcl-2 that resulted in up-regulation of the ratio of Bax/Bcl-2 and the activity of caspase-3, -8, and -9. Eventually, DATS induced the apoptosis and inhibited the proliferation in a concentration- and time-dependent manner. Furthermore, by establishing an animal model of female BALB/c nude mice with A549 xenografts, we found that oral gavage of DATS significantly retarded growth of A549 xenografts in nude mice without causing weight loss or any other side effects compared with the control group. All the evidence both in vitro and in vivo suggested that DATS could be an ideal anti-cancer drug. 相似文献
