Cochliomycin A is a compound with anti-barnacle settlement activity and low toxicity, but the molecular mechanism of the compound is unknown. Here, isobaric tags for the relative or absolute quantitation (iTRAQ) labeling proteomic method were applied to analyze changes in the proteome of Amphibalanus(=Balanus) amphitrite cyprids in response to cochliomycin A treatment. Cochliomycin A affected the cytochrome P450, glutathione S-transferase (GST) and NO/cGMP pathways, among which the NO/cGMP pathway was considered to play a key role in barnacle larval settlement, while the cytochrome P450 and the GST pathways are mainly for detoxification. The results of real-time PCR further suggested the NO/cGMP pathway was activated in response to cochliomycin A. Larval settlement assays revealed that S-methylisothiourea sulfate (SMIS) and 1H-(1,2,4)oxadiazolo[4,3-a]quinoxalin-1-one (ODQ) rescued cyprids from cochliomycin A-induced inhibition of larval settlement. The findings supported the hypothesis that cochliomycin A inhibited barnacle larval settlement by stimulating the NO/cGMP pathway. 相似文献
The sensory drive hypothesis proposes that environmental factors affect both signalling dynamics and the evolution of signals and receivers. Sound detection and equilibrium in marine fishes are senses dependent on the sagittae otoliths, whose morphological variability appears intrinsically linked to the environment. The aim of this study was to understand if and which environmental factors could be conditioning the evolution of this sensory structure, therefore lending support to the sensory drive hypothesis. Thus, we analysed the otolith shape of 42 rockfish species (Sebastes spp.) to test the potential associations with the phylogeny, biological (age), ecological (feeding habit and depth distribution) and biogeographical factors. The results showed strong differences in the otolith shapes of some species, noticeably influenced by ecological and biogeographical factors. Moreover, otolith shape was clearly conditioned by phylogeny, but with a strong environmental effect, cautioning about the use of this structure for the systematics of rockfishes or other marine fishes. However, our most relevant finding is that the data supported the sensory drive hypothesis as a force promoting the radiation of the genus Sebastes. This hypothesis holds that adaptive divergence in communication has significant influence relative to other life history traits. It has already been established in Sebastes for visual characters and organs; our results showed that it applies to otolith transformations as well (despite the clear influence of feeding and depth), expanding the scope of the hypothesis to other sensory structures. 相似文献
In addition to the known antitumour effects of ursolic acid (UA), increasing evidence indicates that this molecule plays a role in cardiac protection. In this study, the effects of ursolic acid on the heart in mice treated with doxorubicin (DOX) were assessed. The results showed that ursolic acid improved left ventrical fractional shortening (LVFS) and left ventrical ejection fraction (LVEF) of the heart, increased nitrogen oxide (NO) levels, inhibited reactive oxygen species (ROS) production and decreased cardiac apoptosis in mice treated with doxorubicin. Mechanistically, ursolic acid increased AKT and endothelial nitric‐oxide synthase (eNOS) phosphorylation levels, and enhanced eNOS expression, while inhibiting doxorubicin induced eNOS uncoupling through NADPH oxidase 4 (NOX4) down‐regulation. These effects of ursolic acid resulted in heart protection from doxorubicin‐induced injury. Therefore, ursolic acid may be considered a potential therapeutic agent for doxorubicin‐associated cardiac toxicity in clinical practice. 相似文献
Microplastics have been widely considered as contaminants for the environment and biota. Till now, most previous studies have focused on the identification and characterization of microplastics in freshwater, sea water, and the terrestrial environment. Although microplastics have been extensively detected in the wastewater, research in this area is still lacking and not thoroughly understood. To fill this knowledge gap, the current review article covers the analytical methods of microplastics originating from wastewater streams and describes their sources and occurrences in wastewater treatment plants (WWTPs). Studies indicated that microplastic pollution caused by domestic washing of synthetic fibers could be detected in the effluent; however, most microplastics from personal care and cosmetic products (PCCPs) can be efficiently removed during wastewater treatment. Moreover, various techniques for sampling and analyzing microplastics from wastewater systems are reviewed; while, the implementation of standardized protocols for microplastics is required. Finally, the fate of microplastics during wastewater treatments and the environmental contamination of effluent to environment are presented. Previous studies reported that the advanced wastewater treatment (e.g., membrane bioreactor) is needed for improving the removal efficiency of small-sized microplastics (<?100 µm). Although the role of microplastics as transport vectors for persistent organic pollutants (POPs) is still under debate, they have demonstrated abilities to absorb harmful agents like pharmaceuticals.
Trillions of microbes reside in the human body and participate in multiple physiological and pathophysiological processes that affect host health throughout the life cycle. The microbiome is hallmarked by distinctive compositional and functional features across different life periods.Accumulating evidence has shown that microbes residing in the human body may play fundamental roles in infant development and the maturation of the immune system. Gut microbes are thought to be essential for the facilitation of infantile and childhood development and immunity by assisting in breaking down food substances to liberate nutrients, protecting against pathogens, stimulating or modulating the immune system, and exerting control over the hypothalamic–pituitary–adrenal axis. This review aims to summarize the current understanding of the colonization and development of the gut microbiota in early life, highlighting the recent findings regarding the role of intestinal microbes in pediatric diseases. Furthermore, we also discuss the microbiota-mediated therapeutics that can reconfigure bacterial communities to treat dysbiosis. 相似文献
Evidence suggests that global maize yield declines with a warming climate, particularly with extreme heat events. However, the degree to which important maize processes such as biomass growth rate, growing season length (GSL) and grain formation are impacted by an increase in temperature is uncertain. Such knowledge is necessary to understand yield responses and develop crop adaptation strategies under warmer climate. Here crop models, satellite observations, survey, and field data were integrated to investigate how high temperature stress influences maize yield in the U.S. Midwest. We showed that both observational evidence and crop model ensemble mean (MEM) suggests the nonlinear sensitivity in yield was driven by the intensified sensitivity of harvest index (HI), but MEM underestimated the warming effects through HI and overstated the effects through GSL. Further analysis showed that the intensified sensitivity in HI mainly results from a greater sensitivity of yield to high temperature stress during the grain filling period, which explained more than half of the yield reduction. When warming effects were decomposed into direct heat stress and indirect water stress (WS), observational data suggest that yield is more reduced by direct heat stress (?4.6 ± 1.0%/°C) than by WS (?1.7 ± 0.65%/°C), whereas MEM gives opposite results. This discrepancy implies that yield reduction by heat stress is underestimated, whereas the yield benefit of increasing atmospheric CO2 might be overestimated in crop models, because elevated CO2 brings yield benefit through water conservation effect but produces limited benefit over heat stress. Our analysis through integrating data and crop models suggests that future adaptation strategies should be targeted at the heat stress during grain formation and changes in agricultural management need to be better accounted for to adequately estimate the effects of heat stress. 相似文献
