Osteosarcoma is one of the most common primary malignancies in bones and is characterized by high metastatic rates. Circulating tumor cells (CTCs) derived from solid tumors can give rise to metastatic lesions, increasing the risk of death in patients with cancer. Here, we used bioinformatics tools to compare the gene expression between CTCs and metastatic lesions in osteosarcoma to identify novel molecular mechanisms underlying osteosarcoma metastasis. We identified TRAIP as a key differentially expressed gene with prognostic significance in osteosarcoma. We demonstrated that TRAIP regulated the proliferation and invasion of osteosarcoma cells. In addition, we found that TRAIP promoted KANK1 polyubiquitination and subsequent degradation, downregulating IGFBP3 and activating the AKT pathway in osteosarcoma cells. These results support the critical role of the TRAIP/KANK1/IGFBP3/AKT signaling axis in osteosarcoma progression and suggest that TRAIP may represent a promising therapeutic target for osteosarcoma.Subject terms: Bone cancer, Oncogenes 相似文献

Androgen receptor splice variant 7 (AR-V7), a form of ligand-independent and constitutively activating variant of androgen receptor (AR), is considered as the key driver to initiate castration-resistant prostate cancer (CRPC). Because AR-V7 lacks ligand-binding domain, the AR-targeted therapies that aim to inactivate AR signaling through disrupting the interaction between AR and androgen are limited in CRPC. Thus, the emergence of AR-V7 has become the greatest challenge for treating CRPC. Targeting protein degradation is a recently proposed novel avenue for cancer treatment. Our previous studies have been shown that the oncoprotein AR-V7 is a substrate of the proteasome. Identifying novel drugs that can trigger the degradation of AR-V7 is therefore critical to cure CRPC. Here we show that nobiletin, a polymethoxylated flavonoid derived from the peel of Citrus fruits, exerts a potent anticancer activity via inducing G0/G1 phase arrest and enhancing the sensitivity of cells to enzalutamide in AR-V7 positive PC cells. Mechanically, we unravel that nobiletin selectively induces proteasomal degradation of AR-V7 (but not AR). This effect relies on its selective inhibition of the interactions between AR-V7 and two deubiquitinases USP14 and USP22. These findings not only enrich our understanding on the mechanism of AR-V7 degradation, but also provide an efficient and druggable target for overcoming CRPC through interfering the stability of AR-V7 mediated by the interaction between AR-V7 and deubiquitinase.Subject terms: Drug development, Translational research 相似文献

An ecosystem-level study was conducted in the Guandu wetlands insubtropical coastal Taiwan to examine how salinity influences the abundance,diversity, and structure of biotic communities. We surveyed eight permanentstudy sites, spanning freshwater marshes, to the gate on the dyke, andmesohaline mangroves representing a gradient of the extent of saltwaterincursions. Analyses of abiotic variables showed that salinity was the primarydetermining factor for discriminating habitat types in the wetlands, butcommunities differed in their sensitivity to salinity. The composition of plantand insect communities was most affected by the salinity gradient, suggestingthe utility of these communities for ecological monitoring of saltwaterincursions. However, spatial changes in communities at higher trophic levels,including macrobenthos, mollusks, fish, and birds, could not be explained simplyby the salinity gradient. Instead, changes in these communities were morerelevant to the composition of other biotic communities. Our results show thatspecies richness and diversity of plant communities were higher in the marshesthan in the mangroves. Nevertheless, insect communities censused in themangroves had higher diversity, despite lower abundance and species richness.Macrobenthos surveyed in the mangroves showed higher biomass and number of taxa.Mollusks and fish were also more abundant at sites near the gate compared to themarsh sites. This suggests that maintaining a tidal flux by means of gateregulation is necessary for conserving the spatial heterogeneity andbiodiversity of coastal wetlands. 相似文献

This report examines plasma amyloid proteins A40 and A42 and apolipoprotein E (apoE) levels and their relationships with age in non-demented older adults with (N = 32) or without the apoE-4 allele (N = 94). A levels did not differ between the groups whereas the 4 allele was associated with a significant reduction in plasma apoE. In subjects with the 4 allele, increasing age was associated with significant reduction in plasma A40. Subjects without the 4 allele showed a significant positive correlation between A40 and A42 levels. There was also a significant correlation between plasma A40 and apoE levels in all subjects. 相似文献

A transformation protocol, based on co-inoculation with two strains of Agrobacterium, Agrobacterium tumefaciens LBA4404 and A. rhizogenes 15834 containing a binary vector with the GUS gene, was established for the induction of transgenic hairy roots from sugar
beet (Beta vulgaris L.) explants. It resulted in marked improvement in the formation of hairy roots and the integration of the binary vector
T-DNA into the host genome. Of 250 inoculated sugar beet hypocotyls, 84% yielded hairy roots 5–7 days after inoculation, of
which 70% were co-transformed with the binary vector T-DNA. To determine stable expression of alien genes in hairy roots,
the nematode resistance gene Hs1

was used as a reporter gene. In addition, molecular marker analysis was applied to monitor stable incorporation of a translocation
from the wild beet B. procumbens. The molecular analysis and the nematode (Heterodera schachtii) resistance test in vitro demonstrated that the genomic structure and the expression of the Hs1

-mediated nematode resistance were well-maintained in all hairy root cultures even after repeated sub-culture.

491份宫颈拭子病毒分离结果表明:糜烂宫颈单纯疱疹病毒(HSV)分离阳性率(30.8%)是正常宫颈(2.6%)的11.8倍,用人干扰素治疗一个疗程后,病毒分离率下降至疗前的1/4.36例糜烂宫颈活体组织DNA分子杂交表明,乳头瘤病毒16型(HPV-16)阳性者占52.8和,HPV-18占17.9%,HPV-6B占28.1%,HPV-11占7.7%,251例宫颈糜烂患者经人(?)D型基因工程干扰素双盲对比治疗后,总有效率达93.8%,显效率达60%,分析临床疗效与HSV分离率的变化表明,临床有效病例中有35%(49/140)在治疗后病毒阴转,有57%在疗前疗后均未分离出HSV,有5%在疗前疗后保持阳性不变,有2.9%疗前阴性,疗后阳性,上述结果表明,HSV和HPV与慢性宫颈炎有一定关系。 相似文献

Ku70 plays an important role in the DSBR (DNA double-strand breaks repair) and maintenance of genomic integrity. Genetic variations
within human Ku70 have been demonstrated to be associated with increased risk of several types of cancers. In this hospital-based
case–control study, we aimed to investigate whether a single nucleotide polymorphism (SNP) in the promoter region (rs2267437)
of Ku70 gene is associated with susceptibility to breast cancer in Chinese Han population. A total of 293 patients with breast
cancer and 301 age-matched healthy controls were enrolled in this study. The Ku70 −1310C/G polymorphism was determined by
polymerase chain reaction-restriction fragment length polymorphism (PCR–RFLP) analysis. A significant difference in genotype
distribution and allele frequency was observed between patients and controls. The CG or GG carries were at higher risk of
breast cancer compared with the CC homozygotes (OR = 1.43, 95% CI = 1.02–2.00, P = 0.038 and OR = 3.53, 95% CI = 1.60–7.80, P = 0.002, respectively). Further stratification analysis revealed that G allele was associated with an increased risk of breast
cancer among premenopausal women (OR = 1.68, 95% CI = 1.21–2.33, P = 0.002), but not in postmenopausal women (OR = 1.33, 5% CI = 0.85–2.10, P = 0.216). Our study suggests that the Ku70 −1310C/G promoter polymorphism may be a susceptibility factor for breast cancer

The era of personalized medicine for cancer therapeutics has taken an important step forward in making accurate prognoses for individual patients with the adoption of high-throughput microarray technology. However, microarray technology in cancer diagnosis or prognosis has been primarily used for the statistical evaluation of patient populations, and thus excludes inter-individual variability and patient-specific predictions. Here we propose a metric called clinical confidence that serves as a measure of prognostic reliability to facilitate the shift from population-wide to personalized cancer prognosis using microarray-based predictive models. The performance of sample-based models predicted with different clinical confidences was evaluated and compared systematically using three large clinical datasets studying the following cancers: breast cancer, multiple myeloma, and neuroblastoma. Survival curves for patients, with different confidences, were also delineated. The results show that the clinical confidence metric separates patients with different prediction accuracies and survival times. Samples with high clinical confidence were likely to have accurate prognoses from predictive models. Moreover, patients with high clinical confidence would be expected to live for a notably longer or shorter time if their prognosis was good or grim based on the models, respectively. We conclude that clinical confidence could serve as a beneficial metric for personalized cancer prognosis prediction utilizing microarrays. Ascribing a confidence level to prognosis with the clinical confidence metric provides the clinician an objective, personalized basis for decisions, such as choosing the severity of the treatment. 相似文献

Soil organic carbon (SOC) plays an important role in improving soil properties and the C global cycle. Limited attention, though, has been given to assessing the spatial patterns and stocks of SOC in desert ecosystems. In this study, we quantitatively evaluated the spatial variability of SOC and its influencing factors and estimated SOC storage in a region (40 km2) of the Gobi desert. SOC exhibited a log-normal depth distribution with means of 1.6, 1.5, 1.4, and 1.4 g kg−1 for the 0–10, 10–20, 20–30, and 30–40 cm layers, respectively, and was moderately variable according to the coefficients of variation (37–42%). Variability of SOC increased as the sampling area expanded and could be well parameterized as a power function of the sampling area. Significant correlations were detected between SOC and soil physical properties, i.e. stone, sand, silt, and clay contents and soil bulk density. The relatively coarse fractions, i.e. sand, silt, and stone contents, had the largest effects on SOC variability. Experimental semivariograms of SOC were best fitted by exponential models. Nugget-to-sill ratios indicated a strong spatial dependence for SOC concentrations at all depths in the study area. The surface layer (0–10 cm) had the largest spatial dependency compared with the other layers. The mapping revealed a decreasing trend of SOC concentrations from south to north across this region of the Gobi desert, with higher levels close to an oasis and lower levels surrounded by mountains and near the desert. SOC density to depths of 20 and 40 cm for this 40 km2 area was estimated at 0.42 and 0.68 kg C m−2, respectively.
