Root hair tip growth provides a unique model system for the study of plant cell polarity. Transgenic plants expressing constitutively active (CA) forms of ROP (Rho-of-plants) GTPases have been shown to cause the disruption of root hair polarity likely as a result of the alteration of actin filaments (AF) and microtubules (MT) organization. Towards understanding the mechanism by which ROP controls the cytoskeletal organization during root hair tip growth, we have screened for CA-rop2 suppressors or enhancers using CA1-1, a transgenic line that expresses CA-rop2 and shows only mild disruption of tip growth. Here, we report the characterization of a CA-rop2 enhancer (cae1-1 CA1-1) that exhibits bulbous root hairs. The cae1-1 mutation on its own caused a waving and branching root hair phenotype. CAE1 encodes the root hair growth-related, ARM domain-containing kinesin-like protein MRH2 (and thus cae1-1 was renamed to mrh2-3). Cortical MT displayed fragmentation and random orientation in mrh2 root hairs. Consistently, the MT-stabilizing drug taxol could partially rescue the wavy root hair phenotype of mrh2-3, and the MT-depolymerizing drug Oryzalin slightly enhanced the root hair tip growth defect in CA1-1. Interestingly, the addition of the actin-depolymerizing drug Latrunculin B further enhanced the Oryzalin effect. This indicates that the cross-talk of MT and AF organization is important for the mrh2-3 CA1-1 phenotype. Although we did not observe an apparent effect of the MRH2 mutation in AF organization, we found that mrh2-3 root hair growth was more sensitive to Latrunculin B. Moreover, an ARM domain-containing MRH2 fragment could bind to the polymerized actin in vitro. Therefore, our genetic analyses, together with cell biological and pharmacological evidence, suggest that the plant-specific kinesin-related protein MRH2 is an important component that controls MT organization and is likely involved in the ROP2 GTPase-controlled coordination of AF and MT during polarized growth of root hairs.  相似文献

Small ubiquitin-related protein modifiers (SUMO) modification is an important mechanism for posttranslational regulation of protein function. However, it is largely unknown how the sumoylation pathway is regulated. Here, we report that nitric oxide (NO) causes global hyposumoylation in mammalian cells. Both SUMO E2 conjugating enzyme Ubc9 and E3 ligase protein inhibitor of activated STAT3 (Pias3) were targets for S-nitrosation. S-nitrosation did not interfere with the SUMO conjugating activity of Ubc9, but promoted Pias3 degradation by facilitating its interaction with tripartite motif-containing 32 (Trim32), a ubiquitin E3 ligase. On the one hand, NO promoted Trim32-mediated Pias3 ubiquitination. On the other hand, NO enhanced the stimulatory effect of Pias3 on Trim32 autoubiquitination. The residue Cys459 of Pias3 was identified as a target site for S-nitrosation. Mutation of Cys459 abolished the stimulatory effect of NO on the Pias3-Trim32 interaction, indicating a requirement of S-nitrosation at Cys459 for positive regulation of the Pias3-Trim32 interplay. This study reveals a novel crosstalk between S-nitrosation, ubiquitination, and sumoylation, which may be crucial for NO-related physiological and pathological processes.  相似文献

Two new sesqulterpenoid glucosides, namely α-hydroxycostic acid 6-β-D-glucopyranoside (compound 1) and 11 βH-11,13-dlhydrodehydrocostuslactone 8α-O-(6'-acetyl)-β-D-glucopyranoside (compound 2), along with 11 known sesqulterpenoids (compounds 3-13) were isolated from the aerial parts of Saussurea involucrate (Kar. et Kir.) Sch.- BIp. The structures of the new sesquiterpenoid glucosides were established by one- and two-dimensional nuclear magnetic resonance and mass spectrometry analysis.  相似文献

We investigated the role of the "sieve tube-companion cell complex" lining the tube periphery, particularly the microfilament and microtubule, in assisting the pushing of phloem sap flow. We made a simple phloem transport system with a living radish plant, in which the conducting channel was exposed for local treatment with chemicals that are effective in modulating protoplasmic movement (acetylcholine, (ACh) a neurotransmitter in animals and insects; cytochalasin B, (CB) a specific inhibitor of many cellular responses that are mediated by microfilament systems and amiprophos-methyl, (APM) a specific inhibitor of many cellular responses that are mediated by microtubule systems). Their effects on phloem transport were estimated by two experimental devices: (i) a comparison of changes in the amount of assimilates in terms of carbohydrates and ^14C-labeled photosynthetic production that is left in the leaf blade of treated plants; and (ii) distribution patterns of ^14C-labeled leaf assimilates in the phloem transport system. The results indicate that CB and APM markedly inhibited the transfer of photosynthetic product from leaf to root via the leaf vein, while ACh enhanced the transfer of photosynthetic product in low concentrations (5.0×10^-4 mol/L) but inhibited it in higher concentrations (2.0×10^-3 mol/L) from leaf to root via the leaf vein. Autoradiograph imaging clearly reveals that ACh treatment is more effective than the control, and both CB and APM treatments effectively inhibit the passage of radioactive assimilates. All of the results support the postulation that the peripheral protoplasm in the sieve tube serves not only as a passive semi-permeable membrane, but is also directly involved in phloem transport.  相似文献

对采自中国4个省份的9个梨树腐烂病菌分离株和7个苹果树腐烂病菌分离株的ITS序列进行了测定和分析,并结合GenBank的有性型Valsa ceratosperma、V.ambiens和V.mali的ITS序列构建了系统发育树。结果表明梨和苹果树的各分离株在ITS核苷酸序列上分化较小(p-distance=1.55%),均在V.ceratosperma聚类组,但二者又分别处于两个独立小分支。其与V.ambiens和V.mali处在不同的聚类组中,且亲缘关系较远,表明供试梨树腐烂病菌并非V.ambiens。培养性状和生物学特点的研究结果还发现,梨树腐烂病菌各分离株无论在菌落颜色、产孢特点、还是37℃高温的生长情况都和苹果腐烂病菌有一定差别。前者菌落始在PDA终为乳白色,而后者菌落初为白色后期变褐色;在20%ABA上,前者形成的产孢体较大而数量较少,在37℃高温下能正常生长,后者则形成的产孢体较小而数量较多,在37℃高温下不能正常生长。并未发现二者在子实体上有稳定明显的差异。因而表明梨树腐烂病菌应为V.ceratosperma,但可用培养性状和生物学特点进行区分其和苹果树腐烂病菌。  相似文献

We investigated the effects of homocysteine (Hcy) and oxidized low density lipoprotein (ox-LDL) on DNA methylation in the promoter region of the estrogen receptor α (ERos) gene,and its potentialmechanism in the pathogenesis of atherosclerosis.Cultured smooth muscle cells (SMCs) of humans weretreated by Hcy and ox-LDL with different concentrations for different periods of time.The DNA methylationstatus was assayed by nested methylation-specific polymerase chain reaction,the lipids that accumulated inthe SMCs and foam cell formations were examined with Oil red O staining.The proliferation of SMCs wasassayed by the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide method.The results showedthat ox-LDL in moderate concentrations (10-40 mg/L) induced de novo methylation in the promoter regionof the ERα gene of SMCs.However,high concentrations (50 mg/L) of ox-LDL,resulted in demethylation ofERα.The Hcy treatment resulted in de novo methylation in the promoter region of the ERα gene with aconcentration- and treating time-dependent manner,and a dose-dependent promoting effect on SMCproliferation.These data indicated that the two risk factors for atherosclerosis had the function of inducingde novo methylation in the promoter region of the ERα gene of SMCs. However,high concentrations (50rag/L) of ox-LDL induced demethylation,indicating that different risk factors of atherosclerosis with differentpotency might cause different aberrant methylation patterns in the promoter region of the ERα gene.Theatherogenic mechanism of Hcy might involve the hypermethylation of the ERα gene,leading to the proliferationof SMCs in atherosclerotic lesions.  相似文献

Satellite RNAs(satRNAs)are molecular parasites that interfere with the pathogenesis of thehelper viruses.In this study,the relative accumulation of cucumber mosaic virus(CMV)-Fny genomicRNAs with or without satRNAs were quantitatively analyzed by real-time RT-PCR.The results showed thatsatRs apparently attenuated the symptoms of CMV-Fny on Nicotiana tabacum by depressing the accumu-lation of CMV-Fny genomic RNAs,tested as open reading frames.The accumulation of CMV-Fny 1a,2a,2b,3a,and CP genes was much higher than that of CMV-Fny with satRs added(CMV-Fsat),at differentinoculation times.CMV-FnyΔ2b,in which the complete 2b gene and 41 amino acids at the C-terminal of the2a gene were deleted,caused only a slight mosaic effect on N.tabacum seedlings,similar to that of CMV-Fsat,but the addition of satRs to CMV-FnyΔ2b showed further decrease in the accumulation of CMV-FnyΔ2b genomic RNAs.Our results indicated that the attenuation of CMV,by adding satRs or deleting the2b gene,was due to the low accumulation of CMV genomic RNAs,and that satRNA-mediated reduction ofCMV genomic RNAs accumulation in N.tabacum was possibly related to the 2b gene.  相似文献

应用组织学方法及免疫组织化学技术显示,黄鳝性逆转生殖发育过程中,生殖干细胞(GSCs)定位分布于生殖褶中,黄鳝雌性发育阶段的GSCs分散或成团存在,间性及雄性发育阶段GSCs均区分为A、B两种不同类型,雌性发育阶段GSCs与A、B两类GSCs在超微结构上存在差异。结果表明,生殖褶中GSCs是黄鳝分化生殖腺中唯一具有有丝分裂能力的生殖细胞群,雌性发育阶段GSCs表现出卵原干细胞特征,间性及雄性发育阶段GSCs为精原干细胞。CD49整合素是黄鳝雌性发育阶段GSCs和A类GSCs的表征分子。  相似文献

通过对国人Ⅰ型遗传性淋巴水肿一家系分子遗传学检测,报告VEGFR-3基因新突变。首先在Ⅰ型遗传性淋巴水肿对该家系进行致病基因的连锁分析,然后用DNA直接测序方法进行基因突变分析。连锁分析和单倍体分析确定该家系致病基因位于5q35.3,与Ⅰ型遗传性淋巴水肿连锁。VEGFR-3基因突变分析发现了一个新的错义突变D1055V,该错义突变在家系中共分离,且在100个正常对照组中未发现该序列改变。本研究首次报告了国内Ⅰ型遗传性淋巴水肿VEGFR-3基因新的错义突变D1055V,丰富了VEGFR-3基因基因突变谱,为今后开展遗传性淋巴水肿的基因诊断和遗传咨询奠定基础。  相似文献

Tropical forests continue to vanish rapidly,but few long-term studies have ever examined if and how the lost forests can be restored.Based on a 45-year restoration study in south China,we found that a tropical rain forest,once completely destroyed,could not recover naturally without deliberate restoration efforts.We identified two kinds of thresholds that must be overcome with human ameliorative measures before the ecosystem was able to recover.The first threshold was imposed primarily by extreme physical conditions such as exceedingly high surface temperature and impoverished soil,while the second was characterized by a critical level of biodiversity and a landscape context that accommodates dispersal and colonization processes.Our three treatment catchments(un-restored barren land,single-species plantation,and mixed-forest stand)exhibited dramatically different changes in biodiversity and ecosystem functioning over 4 decades.The mixed forest,having the highest level of biodiversity and ecosystem functioning,possesses several major properties of tropical rain forest.These findings may have important implications for the restoration of many severely degraded or lost tropical forest ecosystems.  相似文献
