Endothermic mammals have a high energy cost to maintain a stable and high body temperature (Tb, around 37°C). Thyroid hormones are a major regulator for energy metabolism and Tb. The gut microbiota is involved in modulating host energy metabolism. However, whether the interaction between the gut microbiota and thyroid hormones is involved in metabolic and thermal regulations is unclear. We hypothesized that thyroid hormones via an interaction with gut microbiota orchestrate host thermogenesis and Tb. l -thyroxine-induced hyperthyroid Mongolian gerbils (Meriones unguiculatus) increased resting metabolic rate (RMR) and Tb, whereas Methimazole-induced hypothyroid animals decreased RMR. Both hypothyroid and hyperthyroid animals differed significantly in faecal bacterial community. Hyperthyroidism increased the relative abundance of pathogenic bacteria, such as Helicobacter and Rikenella, and decreased abundance of beneficial bacteria Butyricimonas and Parabacteroides, accompanied by reduced total bile acids and short-chain fatty acids. Furthermore, the hyperthyroid gerbils transplanted with the microbiota from control donors increased type 2 deiodinase (DIO2) expression in the liver and showed a greater rate of decline of both serum T3 and T4 levels and, consequently, a more rapid recovery of normal RMR and Tb. These findings indicate that thyroid hormones regulate thermogenesis depending on gut microbiota and colonization with normal microbiota by caecal microbial transplantation attenuates hyperthyroid-induced thermogenesis. This work reveals the functional consequences of the gut microbiota-thyroid axis in controlling host metabolic physiology and Tb in endotherms. 相似文献

Nicotine, one of the most commonly used drugs, has become a major concern because tobacco serves as a gateway drug and is linked to illicit drug abuse, such as cocaine and marijuana. However, previous studies mainly focused on certain genes or neurotransmitters which have already been known to participate in drug addiction, lacking endogenous metabolic profiling in a global view. To further explore the mechanism by which nicotine modifies the response to cocaine, we developed two conditioned place preference (CPP) models in mice. In threshold dose model, mice were pretreated with nicotine, followed by cocaine treatment at the dose of 2 mg/kg, a threshold dose of cocaine to induce CPP in mice. In high-dose model, mice were only treated with 20 mg/kg cocaine, which induced a significant CPP. 1H nuclear magnetic resonance based on metabonomics was used to investigate metabolic profiles of the nucleus accumbens (NAc) and striatum. We found that nicotine pretreatment dramatically increased CPP induced by 2 mg/kg cocaine, which was similar to 20 mg/kg cocaine-induced CPP. Interestingly, metabolic profiles showed considerable overlap between these two models. These overlapped metabolites mainly included neurotransmitters as well as the molecules participating in energy homeostasis and cellular metabolism. Our results show that the reinforcing effect of nicotine on behavioral response to cocaine may attribute to the modification of some specific metabolites in NAc and striatum, thus creating a favorable metabolic environment for enhancing conditioned rewarding effect of cocaine. Our findings provide an insight into the effect of cigarette smoking on cocaine dependence and the underlying mechanism. 相似文献

Infections with intestinal helminth and bacterial pathogens, such as enteropathogenic Escherichia coli, continue to be a major global health threat for children. To determine whether and how an intestinal helminth parasite, Heligomosomoides polygyrus, might impact the TLR signaling pathway during the response to a bacterial enteropathogen, MyD88 knockout and wild-type C57BL/6 mice were infected with H. polygyrus, the bacterial enteropathogen Citrobacter rodentium, or both. We found that MyD88 knockout mice co-infected with H. polygyrus and C. rodentium developed more severe intestinal inflammation and elevated mortality compared to the wild-type mice. The enhanced susceptibility to C. rodentium, intestinal injury and mortality of the co-infected MyD88 knockout mice were found to be associated with markedly reduced intestinal phagocyte recruitment, decreased expression of the chemoattractant KC, and a significant increase in bacterial translocation. Moreover, the increase in bacterial infection and disease severity were found to be correlated with a significant downregulation of antimicrobial peptide expression in the intestinal tissue in co-infected MyD88 knockout mice. Our results suggest that the MyD88 signaling pathway plays a critical role for host defense and survival during helminth and enteric bacterial co-infection. 相似文献

In the untransformed rice (WT) and transgenic rice with the PEPC and PPDK genes (CK) we determined activities of C4 photosynthetic enzymes, photosynthetic response to irradiance and temperature, the metabolic index of active oxygen, and
the yield component factors. The activities of C4 photosynthetic enzymes in WT were very low, while those of corresponding enzymes in CK were highly observable. Moreover,
after adenosine triphosphate (ATP) treatment, and under high irradiance and high temperature, the net photosynthetic rate
of CK increased by 17 and 12 %, respectively, as compared to that achieved without ATP treatment. The resistance of CK against
photo-oxidation was enhanced under these conditions, and CK yield increased by 15 %. ATP treatment enhanced the photosynthetic
productivity of CK, thereby proving that ATP is the key factor in enhancing the photosynthetic capacity of transgenic rice

以定虫隆汰选的抗性指数为23.78倍抗性种群(CH-R),并以上海田间种群(SH-R,对定虫隆的抗性指数为9.16倍)作为参比种群,研究小菜蛾Plutella xylostlla(L.)对定虫隆的抗性机制,结果表明:CH—R种群多功能氧化酶O-脱甲基活力比相对敏感种群(SZ-S)提高1倍,而SH-R种群多功能氧化酶O-脱甲基活力虽有所提高,但与SZ-S种群相比差异并不显著;CH-R和SH-R种群羧酸酯酶比活力均明显高于Sz-S种群,分别为SZ-S种群的4.61和2.18倍,且CH-R种群的Km仅为SZ-S种群的1/20;酸性磷酸酯酶和碱性磷酸酯酶比活力种群间无明显差异;CH-R种群的几丁质酶活力降低48%,酚氧化酶活力降低60%。说明小菜蛾对定虫隆的抗性机制具有多因子性,多功能氧化酶解毒代谢能力的提高可能是主导抗性机制之一,羧酸酯酶、几丁质酶和酚氧化酶也参与了小菜蛾对定虫隆的抗性。 相似文献
