In previous studies, we have shown that the inactivation of the adenosine A2A receptor exacerbates chronic cerebral hypoperfusion-induced white matter lesions (WMLs) by enhancing neuroinflammatory responses. However, the molecular mechanism underlying the effect of the adenosine A2A receptor remains unknown. Recent studies have demonstrated that cystatin F, a potent endogenous cysteine protease inhibitor, is selectively expressed in immune cells in association with inflammatory demyelination in central nervous system diseases. To understand the expression of cystatin F and its potential role in the effect of A2A receptor on WMLs induced through chronic cerebral hypoperfusion, we investigated cystatin F expression in the WMLs of A2A receptor gene knockout mice, the littermate wild-type mice and wild-type mice treated daily with the A2A receptor agonist or both CGS21680 and A2A receptor antagonist CGS21680 after chronic cerebral hypoperfusion. The results of quantitative-PCR and western blot analysis revealed that cystatin F mRNA and protein expression were significantly up-regulated in the WMLs after chronic cerebral hypoperfusion. In addition, cystatin F expression in the corpus callosum was significantly increased in A2A receptor gene knockout mice and markedly decreased in mice treated with SCH58261 on both the mRNA and protein levels. Additionally, CGS21680 counteracted the attenuation of cystatin F expression produced by SCH58261 after chronic cerebral hypoperfusion. Moreover, double immunofluorescence staining revealed that cystatin F was co-localized with the activated microglia marker CD11b. In conclusion, the cystatin F expression in the activated microglia is closely associated with the effect of the A2A receptors, which may be related to the neuroinflammatory responses occurring during the pathological process. Adiantum reniforme var. sinensis (Adiantaceae) is an endangered fern endemic to the Three Gorges region in southwest China. To elucidate possible ecophysiological mechanisms restricting its distribution, effects of the availability of light (28%, 14% and 7% of open field) and soil moisture (60% and 40% of field capacity) on dry matter production and allocation, leaf morphology and water use efficiency (WUE) were examined in A. reniforme var. sinensis and its widespread congener A. capillus‐veneris. Both species had lower root/shoot ratio (R/S) and higher specific leaf area (SLA) when grown at low light. However, A. reniforme var. sinensis showed less plasticity for total leaf area (LA) and leaf area ratio (LAR) than A. capillus‐veneris, and its root mass, total mass and WUE decreased as light availability decreased. Under water stress, all traits of both species except WUE were significantly affected. However, drought stress decreased total mass of A. capillus‐veneris but did not have a significant effect on A. reniforme var. sinensis. Compared with A. capillus‐veneris, A. reniforme var. sinensis had significantly higher R/S but lower values for other analyzed traits. These results suggest that A. reniforme var. sinensis is relatively superior in drought tolerance but inferior at low light, allowing it to persist in habitats with low soil moisture and high light availability but with few coexisting species present. 相似文献

物种分类与识别是生物多样性监测的基础, 明确物种的类别及其分布是解决几乎所有生态学问题的前提。为深入了解基于多源遥感数据的植物物种分类与识别相关研究的发展现状和存在的问题, 本文对2000年以来该领域的研究进行了总结分析, 发现: 当前大多数研究集中在欧洲和北美地区的温带或北方森林以及南非的热带稀树草原; 使用最多的遥感数据是机载高光谱数据, 而激光雷达作为补充数据, 通过单木分割及提供单木的三维垂直结构信息, 显著提高了分类精度; 支持向量机和随机森林作为应用最广的非参数分类算法, 平均分类精度达80%; 随着计算机技术及机器学习领域的不断成熟, 人工神经网络在物种识别领域得以迅速发展。基于此, 本文对目前基于遥感数据的植物物种分类与识别中在分类对象复杂性、多源遥感数据整合、植物物候与纹理特征整合和分类算法技术等方面面临的挑战进行了总结, 并建议通过整合多时相监测数据、高光谱和激光雷达数据、短波红外等特定波谱信息、采用深度学习等方法来提高分类精度。 相似文献

Glioma is one of the major global health problems, including in China. Circular RNAs (circRNAs) have been increasingly identified and characterized in almost every aspect of biology, especially in cancer biology. This research desires to explore the functions and mechanism of a novel circRNA, circ_0079593, on regulating glioma progression. Quantitative real-time polymerase chain reaction (qRT-PCR) was carried out to measure the relative expression of circ_0079593, which was upregulated in matched cancerous tissues from 60 patients and four cell lines of glioma. A higher level of circ_0079593 in glioma specimens was linked to larger tumor size, higher WHO grade, and worse survival rate for patients with glioma. Moreover, circ_0079593 can be deemed as an independent prognostic predictor for glioma patients analyzed by multivariate method. Cell counting kit-8, flow cytometric, wound healing, and transwell experiments were used to evaluate cell growth, apoptosis, migration, and invasion influenced by circ_0079593 knockdown/overexpression. Exogenous downregulation of circ_0079593 expression significantly suppressed glioma cell proliferation by increasing cell apoptosis in vitro, and retarded the migratory and invasive potential. Ectopic expressed circ_0079593 could induce the opposite effects. Mechanistically, bioinformatics analysis, qRT-PCR, and dual-luciferase reporter assays showed that microRNA 182 (miR-182) and miR-433 could be sponged and negatively regulated by circ_0079593. Further, rescue assays demonstrated that the biological functions of circ_0079593 are dependent on its inhibition of miR-182 and miR-433. Collectively, the present work indicates that circ_0079593 may be used as an effective prognostic marker and therapeutic target for glioma. 相似文献

Adipocyte growth and development are complex and precisely orchestrated processes. Several microRNAs have been identified as critical regulators of the adipocyte growth and development. Recently, bta-miR-204 was found to be involved in adipogenesis; however, the underlying molecular mechanism involved in bta-miR-204-mediated regulation of proliferation, differentiation, and apoptosis of adipocytes is not fully understood or elucidated. In this study, quantitative real-time polymerase chain reaction (qRT-PCR), Cell Counting Kit-8, EdU, flow cytometer, Oil Red O staining, and the western blot assays were used to assess the role of bta-miR-204 in adipocyte growth and development. Overexpression of bta-miR-204 had no significant effect on 3T3-L1 cell proliferation. The forced expression of bta-miR-204 promoted 3T3-L1 cell differentiation. Meanwhile, overexpression of bta-miR-204 upregulated the expression of Bax and downregulated the expression of Bcl-2 both at messenger RNA and protein levels, which suggested that bta-miR-204 can promote 3T3-L1 cell apoptosis. Using bioinformatic analysis, dual-luciferase reporter system and qRT-PCR, TGFBR2, and ELOVL6 were identified as the direct target genes of bta-miR-204. Therefore, our study provides a novel insight into the role of bta-miR-204 in the regulation of adipocyte growth and development, which may provide a novel therapeutic alternative against obesity. 相似文献
