Isolation and expression in Escherichia coli of hepB and hepC, genes coding for the glycosaminoglycan-degrading enzymes heparinase II and heparinase III, respectively, from Flavobacterium heparinum.

Upon induction with heparin, Flavobacterium heparinum synthesizes and secretes into its periplasmic space heparinase I (EC 4.2.2.7), heparinase II, and heparinase III (heparitinase; EC 4.2.2.8). Heparinase I degrades heparin, and heparinase II degrades both heparin and heparan sulfate, while heparinase III degrades heparan sulfate predominantly. We isolated the genes encoding heparinases II and III (designated hepB and hepC, respectively). These genes are not contiguous with each other or with the heparinase I gene (designated hepA). hepB and hepC were found to contain open reading frames of 2,316 and 1,980 bp, respectively. Enzymatic removal of pyroglutamate groups permitted sequence analysis of the amino termini of both mature proteins. It was determined that the mature forms of heparinases II and III contain 746 and 635 amino acids, respectively, and have calculated molecular weights of 84,545 and 73,135, respectively. The preproteins have signal sequences consisting of 26 and 25 amino acids. Truncated hepB and hepC genes were used to produce active, mature heparinases II and III in the cytoplasm of Escherichia coli. When these enzymes were expressed at 37 degrees C, most of each recombinant enzyme was insoluble, and most of the heparinase III protein was degraded. When the two enzymes were expressed at 25 degrees C, they were both present predominantly in a soluble, active form.     

Genetic and clonal diversity in Korean populations ofVitex rotundifolia (Verbenaceae)

Vitex rotundifolia L.f. is a woody perennial and has sexual and asexual modes of reproduction. Allozyme study was conducted on 550 plants in 13 Korean populations. The levels of genetic variability and divergence within and among populations, respectively, are considerably lower and higher than the mean values for woody plants with similar life history tralts. Mean percentage of polymorphic loci (P _P), mean number of alleles per locus (A _P), and mean genetic diversity (He _P) within populations ofV. rotundifolia were: 16.7%, 1.21, and 0.047. On average, about 79% of the total variation inV. rotundifolia was common to all populations (meanG _ST=0.208). In addition, significant differences in allele frequencies among populations were found in all polymorphic loci examined (P<0.001). On the other hand, levels of genotypic diversity within and among populations were moderate. About 44% (18/41) of multilocus genotypes were “local genotypes” (genotypes occurring in only one population), whereas only one “widespread genotype” (genotypes occurring in more than 75% of the populations) were detected. The mean number of multilocus genotypes per population (G) and mean genotypic diversity index (D _G) were 8.4 and 0.74, respectively. Most common multilocus genotypes found in populations were homozygous for five polymorphic loci. The abundance of ramets of these genets is responsible for the low levels of expected heterozygosity within populations. The results indicate that clonal reproduction may act as an enhancer of genetic drift by reducing effective size of local populations ofV. rotundifolia.     

猪精子凝集素的纯化,性质及其作用

用胎球蛋白－Ｓｅｐｈａｒｏｓｅ亲和层析和凝胶过滤层析从精子和精浆中分离纯化了猪精子凝集素（简称ＢＳＬ）。ＢＳＬ的血凝活性只被若干糖蛋白和聚糖所抑制。ＢＳＬ的分子量为５６ｋｄ,由分子量分别为１３．６ｋｄ（β）和１６．０ｋｄ（α）的两个不同的亚基以α１β３所组成。ＢＳＬ为糖蛋白,含中性糖３．２％,不含唾液酸。用ＥＬＩＳＡ法测定猪精子中ＢＳＬ的含量及分布,表明７０％嵌入在精子膜中,２５％结合在精子表面,     

利用激光微束穿刺法将外源基因导入小麦的研究

用激光微束穿刺法将携带有新霉素磷酸转移酶（ＮＰＴ－Ⅱ）基因的质粒ｐＪＩＴ１０１导入京花１号小麦幼胚细胞。方法是将小麦幼胚细胞进行高渗缓冲液预处理,用微米级的激光微束处理,然后在卡那霉素培养基上筛选出具抗性的愈伤组织及绿色小植株。第一年,从１５０个小麦幼胚中,在卡那霉素培养基上筛选出４株绿苗,取两株进行ＮＰＴ－Ⅱ酶活性分析,测到了ＮＰＴ－Ⅱ酶的活性。第二年重复实验,从２４５个小麦幼胚中,经筛选获得１株绿苗,进行了叶片ＤＮＡＰＣＲ扩增检测,转化的绿色小苗扩增出所导入的ＮＰＴ－Ⅱ基因编码的片段。结果表明,外源ＮＰＴ－Ⅱ基因已导入了小麦,并实现了整合表达。

记述采自我国云南省勐腊的盖蛛属１新纪录种－鹤嘴盖蛛Ｎｅｒｉｅｎｅｍａｃｅｌｌａ（Ｔｈｏｒｅｌｌ．１８９８）。

以融合蛋白形式在大肠杆菌中表达人MCP－1

将编码人单核细胞趋化蛋白－１（ＭＣＰ－１）的基因亚克隆到大肠杆菌表达载体ｐＥＸ３１Ａ中,在大肠杆菌中表达出ＭＳ２／ＭＣＰ－１融合蛋０白,该表达产物约占菌体总蛋白的１５％左右,Ｗｅｓｔｅｒｎｂｌｏｔ检测表明,表达产物可与ＭＣＰ－１抗体特异反应。采用琼脂糖平板法进行活性测定表明,表达产物具有明显的单核细胞趋化活性,说明Ｎ端融合一段细菌蛋白对ＭＣＰ－１有无趋化活性可能没有影响。     

Effect of physostigmine on relative acetylcholine output induced by systemic treatment with scopolamine in in vivo microdialysis of rat frontal cortex

By means of an in vivo brain microdialysis, the effect of different concentrations of physostigmine on the acetylcholine level in the dialysate of rat frontal cortex was studied. Perfusion of the various degrees of physostigmine (eserine) concentration (10 nM−10 μM) into the cortex through the dialysis membrane increased the basal acetylcholine level in a dose-dependent manner. In the presence of 10 nM, 0.1 μM and 10 μM physostigmine in the perfusate, systemic treatment with scopolamine (0.5 mg/kg, i.p.) increased 200, 270 and 510%, respectively, the relative acetylcholine level in the dialysates in comparison with the corresponding basal levels, while in the absence of physostigmine the treatment increased it only 40%. From these results, it appears that perfusion of physostigmine at a variety of concentrations, changes not only the basal level of acetylcholine induced by the inhibition of acetylcholinesterase but also the relative acetylcholine output induced by systemic treatment with scopolamine.

Multiple genotypes of mitochondrial DNA within a horse population from a small region in Yunnan province of China

mtDNA genotypes of six domestic horses (three adult short horses whose heights are under 1 m and three common domestic horses) from a small region of 15 km² in Malipo county of Yunnan province of China were investigated by the technique of restriction fragment length polymorphism (RFLP) with 16 restriction endonucleases which recognize 6-bp sequences. An average of 56 fragments for an individual was obtained. Unlike other domestic animals, this population of horses exhibits high mtDNA genetic diversity. Each of the six horses has a specific mtDNA genotype showing a pattern of multiple maternal origins, as suggested by fossil and literature records. We think the population of horses is an amazing seed-resource pool of horses and hence deserves to be paid more attention from the view of conservation genetics. However, it is also remarkable that we did not find any typical mtDNA genetic markers which would discriminate between short horses and common domestic horses.
