A group of 14 9-substituted derivatives of 6-benzylaminopurine (BA), including the alanine conjugate, oxygen heterocyclic and alkyl derivatives, and compounds with a modified 9-ribose moiety, were assessed for their ability to retard soybean leaf senescence. The 9-alanine conjugate was very weakly active, and only two compounds, 9-(2-tetrahydropyranyl)-BA (9THP-BA) and 9-(2-tetrahydrofuranyl)-BA (9THF-BA), proved to be considerably more effective than BA. The metabolism of these three BA derivatives was determined to rationalize their differing activity. The alanine conjugate of BA was largely unmetabolized in leaf discs, but 9THP-BA and 9THF-BA released free BA and were also debenzylated to 9THP-adenine and 9THF-adenine, respectively. The three products of metabolism were identified by mass spectrometry. The enhanced activity of 9THP-BA and 9THF-BA, relative to that of BA, is attributed to their greater stability and their ability to gradually release free BA. This released BA was less susceptible to inactivation by alanine conjugate formation than was exogenous BA. The novel BA analogue 7-benzylaminooxazolo[5,4-d]pyrimidine, in which the 9-NH is replaced by oxygen, was inactive at 100 μM. For part II, see Zhang et al. 1987  相似文献

本文总结了武汉东湖水体中磷含量(均以PO4计算)的周年逐月季节和年际变动及其分布上的差异(1973-1985年)。按面积加权法计算总磷的平均含量为0.244毫克/升(1983-1985年),总溶解磷和溶解活性磷的平均含量分别为0.121毫克/升和0.051毫克/升(1981-1984年),总磷和总溶解磷周年中出现两次高峰含量,即春季(3-5月)和夏末秋初(8-9月)。低含量出现在水温最低的冬季(12-2月),周年中溶解活性磷高峰含量出现在冬末春初(1-3月),低含量多数出现在春天夏初(5-7月)。东湖水体中磷含量平面分布有明显的差异,而垂直分布表层和底层差异小,各种形态磷的组成中颗粒磷所占比较最大(1983-1984年平均值),平均占总磷63.4%,溶解非活性磷所占比较最小,平均占总磷12.0%。  相似文献

Cyclic AMP accumulation in response to forskolin, cholera toxin, or isoproterenol is dramatically increased in HIT T-15 cells, a clonal cell line of Syrian hamster pancreatic islet beta cells, as a function of passage number. Forskolin and cholera toxin elevate cyclic AMP levels 5- to 10-fold higher in later passages (87-100) than in earlier passages (70-80). A similar phenomenon is observed with isoproterenol (10 microM) which increases cyclic AMP levels 56-fold in older HIT cells (passage 94), whereas only marginally stimulating cyclic AMP production in younger cells (passage 70-82). To determine whether a change in the stimulatory or inhibitory guanine nucleotide regulatory proteins, Gs or Gi, was responsible for these observations, ADP-ribosylation of HIT cell membranes with cholera toxin and pertussis toxin was examined. All passages contained two cholera toxin substrates at 52 and 45 kDa. The amount of 52 kDa did not appear to change with passage number, but the amount of 45 kDa increased in the later passages (89 and 94). The ratio of 45 to 52 kDa cholera toxin substrate, as determined by densitometric analysis, increased from 0.1 in passages 70, 75, and 82 to 0.45 at passage 89. No passage related changes in a 40-kDa pertussis toxin substrate were observed. An increase in the amount of the 45-kDa alpha-subunit of Gs was confirmed on immunoblots using antisera specific for the alpha-subunits of Gs. The amount of functional Gs present in various HIT cell passages was examined by determining the extent to which extracts from HIT cell membranes reconstituted guanine nucleotide-sensitive adenylyl cyclase in S49 cyc- membranes. Extracts derived from passage 94 reconstituted three to four times more adenylyl cyclase activity in cyc- membranes than extracts from passages 70, 75, and 82. These data indicate that an increase in functional Gs in later passages may be the underlying cause for the increased responsiveness to isoproterenol and forskolin in later passages. These data also suggest that functional differences exist between the Gs alpha-subunits, with the smaller 45-kDa subunit being more efficacious in coupling to cyclic AMP synthesis than the larger 52-kDa subunit. This is a departure from the commonly held view that the two subunits have similar efficacies in stimulating adenylyl cyclase.  相似文献

We report that both Na+ and Ca2+ currents are involved in the action potentials and in the hormone release from rat somatotrophs in primary culture. Single somatotrophs were identified by reverse hemolytic plaque assay (RHPA) and transmembrane voltage and currents were recorded using the whole-cell mode of the patch-clamp technique. Somatotrophs displayed a mean resting potential of -80mV and an average input resistance of 5.7G omega. Most of the cells showed spontaneous or evoked action potentials. Single action potentials or the initial spike in a burst were characterized by their high amplitude and short duration. Tetrodotoxin (TTX, 1 microM) blocked single action potentials and the initial spikes in a burst, whereas action potentials of long duration and low amplitude persisted. Cobalt (2 mM) plus TTX (1 microM) blocked all the action potentials. Voltage-clamp experiments confirmed the presence of both a TTX-sensitive Na+ current and Co2(+)-sensitive Ca2+ currents. TTX or Na(+)-free medium slightly decreased the basal release of GH but did not markedly modify hGRF-stimulated GH release. However, Co2+ (2 mM), which partially decreased the basal release, totally blocked hGRF-stimulated release. We conclude that (1) Na+ currents which initiate rapid action potentials may participate in spontaneous GH release; (2) Ca2+ currents, which give rise to long duration action potentials and membrane voltage fluctuation, are probably involved in both basal and hGRF-stimulated GH releases.  相似文献

The spontaneous activity of neurons in the pars reticulata of substantia nigra (SNpr) was studied in chloral hydrate anesthetized rats. As a function of dose, intravenous diazepam decreased, and methyl-beta-carboline-3-carboxylate (beta CCM) increased discharge frequency. Two days after terminating a one week treatment with flurazepam (FZP), both diazepam and beta CCM showed decreased ability to alter SNpr neuronal activity. Neither residual FZP nor down-regulation of benzodiazepine receptors can account for these results. In contrast, behavioral testing revealed no change in the ability of i.v. beta CCM to cause convulsions, suggesting that sites other than the SNpr are of prime importance in expressing the convulsant actions of systemically injected beta CCM.  相似文献
