人肌肌酸激酶胍变性时的失活与构象变化的比较研究

应用二阶导数光谱、紫外差吸收光谱和荧光光谱等监测手段,研究了人肌肌酸激酶在盐酸胍溶液中的构象变化。二阶导数光谱结果表明,若以6M盐酸胍中肌酸激酶酪氨酸残基的暴露程度为100％,则天然酶酪氨酸残基的暴露程度只有2％。而紫外差吸收光谱和荧光光谱的变化与兔肌肌酸激酶的结果相似。比较不同胍浓度下人肌肌酸激酶的失活与构象变化,表明酶的失活先于构象变化。同时还测定了不同浓度胍溶液中人肌酶的失活与构象变化的速度常数。结果表明以几种方法测定的构象变化均为单相的一级过程,而酶的失活却呈现了由快慢两相组成的一级反应过程。比较同浓度胍溶液中的失活速度与构象变化速度,发现酶失活的快相反应速度常数比构象变化的速度常数大1—2个数量级,慢相速度常数与构象变化速度常数相近。上述结果进一步支持了酶的活性部位构象柔性的观点。     

中国奥甲螨属一新种(蜱螨亚纲:甲螨亚目:奥甲螨科)

本文记述了采自吉林省白城草原的奥甲螨科Oppiidae 奥甲螨属Oppia一新种——白城奥甲螨Oppia baichengensis sp.nov.。对新种的形态特征进行了描述.并对新种与属内近似种作了比较鉴别。文内所用量度单位均为微米(μm)。     

神农架拐棍竹林的初步研究

拐棍竹(Fargesia spathacea Franch.)是中国特有分布种类,主要分布川、滇、陕、甘等省。是极有开发价值的植物资源,又是大熊猫取食主要竹种之一。本文研究了神农架拐棍竹林的生态生物学特点,客观地估算了其蕴藏量,进行了营养成分分析。现报道如下:     

稻米垩白形成的气象生态基础研究

在全国13个点、19个品种多播期试验基础上,对稻米垩自形成的气象生态基础进行了分析。结果表明,水稻齐穗后15天的日均温是影响稻米垩白大小的主要气象因子。经对稻米垩白与齐穗后15天内均温关系的动态分析可知,稻米垩白随该时段温度提高而增大的拐点温度约为29℃(品种间略有差异),接近或超过该点温度,稻米垩白会突发性地增加。     

光钳操纵生物活体的动态监测技术的研究

采用摄像、录像和视屏监控系统及显色偏振装置与光钳系统耦合,从空间分辨、色分辨和时间分辨多方面改善系统品质,实现了光钳捕获与操纵生物活体的动态监测、实时记录、资料保存和屏幕再现的功能,并能测量光钳操纵细胞的位移量和由此计算操纵速度,提高了光钳的自我调整和光钳操纵细胞的精细度。本研究为激光光钳技术在细胞工程等方面的应用研究提供了行之有效的技术手段。     

人体急性缺氧性意识障碍

实验分为低压组(低压舱7,000米)与常压组(吸入7.6％低氧混合气),每组受试者各为12名,有7名参加了两组实验。每组各有4名平均经15分23秒发生了意识模糊或丧失(通称意识障碍)。障碍者的肺泡氧分压为29.9±0.8mmHg。障碍发生前,呈现严重无力,觉醒度降低,反应迟钝,脑波特征是高幅6波(100-300μV)占优势。障碍发生时,高幅δ波丧失节律,幅度衰减,波型不规则,额枕失同步;呼吸循环代偿功能增强程度明显高于障碍前。本工作对于预测和评价人体急性缺氧引起的意识障碍有意义。     

神农香菊干花净油成分的研究

神农香菊全草提取的香浸膏香气浓郁独特,可用于调配多种高档化妆用香精,也可直接用于饮料中,在香精香料工业中具有较大的实用价值。本文首次报告了我们用毛细管气相色谱仪和色谱/质谱/计算机联用分析仪,对神农香菊干花净油成分进行分析的初步结果。鉴定出的已知成分包括脂肪族类,含氧或非含氧的单萜及倍半萜类化合物32个。     

Autocrine role of estrogens in the augmentation of luteinizing hormone receptor formation in cultured rat granulosa cells

The effects of estrogens on gonadotropin-stimulated luteinizing hormone (LH) receptor formation were examined in primary cultures of rat granulosa cells. Granulosa cells were cultured for 3 days with increasing concentrations of follicle-stimulating hormone (FSH) in the presence or absence of native and synthetic estrogens. Follicle-stimulating hormone stimulated LH receptor formation in a dose-dependent fashion, and estrogens enhanced the FSH-stimulated LH receptor content by decreasing the apparent ED50 of FSH. At 6.25 ng/ml FSH, the enhancement in LH receptor was estrogen dose dependent, with an ED50 value of about 3 X 10(-9) M for 17 beta-estradiol. The increased LH receptor content seen in cells treated with FSH and estrogen was correlated with increased cAMP production by these cells in response to LH stimulation. Time course studies revealed enhancement of FSH-stimulated LH receptor induction at 48 and 72 h of culture. Granulosa cells were also cultured with FSH for 2 days to induce functional LH receptors, then further cultured for 3 days with LH in the presence or absence of estrogens. At 30 ng/ml LH, increasing concentrations of estrogens maintained LH receptor content in a dose-dependent fashion, with their relative estrogenic potencies in keeping with reported binding affinities to estrogen receptors. An autocrine role of estrogens on LH receptor formation was further tested in granulosa cells treated with FSH and an aromatase substrate (androstenedione) to increase estrogen biosynthesis. Cotreatment with semipurified estrogen antibodies partially blocked the FSH stimulation of LH receptors, whereas nonimmune serum was ineffective. Also, inclusion of diethylstilbestrol prevented the inhibitory effect of the estrogen antibodies. Thus, local estrogens in ovarian follicles may play an autocrine role in granulosa cells to enhance LH receptor formation and to increase granulosa cell responsiveness to the LH surge, with subsequent ovulation and adequate corpus luteum formation.     

草鱼血液学的研究 Ⅰ.九项血液常数的周年变化

用741尾健康草鱼种作红细胞、血红蛋白、红细胞比积、红细胞平均体积、红细胞平均血红蛋白量、红细胞平均血红蛋白浓度、红细胞沉降率、血浆总蛋白和血浆葡萄糖浓度等九项血液常数的测定。所得的年均值经变异系数分析,除红细胞沉降率和血浆葡萄糖浓度外,其余各项常数的可靠性基本一致。另从周年变动的规律分析,高值出现在冬夏季,低值出现的月份较为分散。水温仅对总蛋白含量的变动呈现显著的负相关(P<0.01)。而体重与血液常数存在着一定的相关因素,在10—90克之间的鱼种中,血红蛋白、红细胞比积、红细胞平均体积、红细胞平均血红蛋白量与体重呈现直线正相关;红细胞沉降率为负相关。     

Geographical variability of alpha-1-antitrypsin alleles in China: A study on six Chinese populations

Summary Alpha-1-antitrypsin phenotypes of six Chinese Han populations (20°–45°N. latitude) were determined. The frequency of allele M2 increases from North to South China. The north-south change of M2 appears to be mainly at the expense of alleles M1 and M3. Geographical variability of other variants was also observed. In the northern populations, the most common variants are M4 and Etokyo; in the southern populations, Pweishi, a variant which can not be distinguished electrophoretically from Pyasugi. These results form a distribution pattern of alpha-1-antitrypsin alleles in China.
