Solid-phase microextraction and cuticular hydrocarbon differences related to reproductive activity in juniper bark borer Semanotus bifasciatus Motschulsky

Cuticular hydrocarbons of Cerambycidae species can function as signals for sex recognition. Little is known about the copulatory signals of the juniper bark borer Semanotus bifasciatus, a major economic threat to Platycladus orientalis Franco in China. Here, we investigated the cuticular hydrocarbons of both sexes of S. bifasciatus to determine the chemically mediated mating signals using the solid-phase microextraction (SPME) technique with carbowax/divinylbenzene fibers (CAR/DVB) and then analyzed by coupled gas chromatography-mass spectrometry (GC-MS). A series of aliphatic saturated straight-chain n-alkanes (n-C₂₃ to n-C₂₈), internally branched monomethylalkanes at carbons 3, 11, or 13, and dimethylalkanes were identified, which showed no qualitative differences in either sex and were similar in the samples with SPME fiber extraction and those with hexane extraction. The bioassay showed that 11-methylpentacosane (11-MeC₂₅), 11-methylhexacosane (11-MeC₂₆), and 11-methylheptacosane (11-MeC₂₇) have sex-specific recognition functions that triggered more mating attempts at a female-specific ratio of 100:4:60 than at a male-specific ratio of 100:85:50. In addition, the female-specific ratio of 11-methylalkanes can elicit about 70% of male mating attempts within about 60 s, whereas live females elicit about 98% of male mating attempts within 25 s. The discrepancy in the initiation of mating attempts by synthetic mixtures and live females suggests that the methyl isomers 3-MeC₂₅, 3-MeC₂₇, and/or 11,15-diMeC₂₇ may also be involved in the mating behavior of S. bifasciatus. These results suggest that 11-MeC₂₅, 11-MeC₂₆, and 11-MeC₂₇ constitute the contact sex pheromone of S. bifasciatus, with the presence or absence of 11-MeC₂₆ in particular playing an important role in mate recognition by males.     

生物成矿作用与生物矿化作用

列举５５种生物矿物，其中包括氧化物，含氧盐，水化物，羟基化物，钙化合物，指出它们通常经历非晶质、过渡、成熟和石化等４个相阶段。根据生物矿物体的元素丰度顺序与岩石圈和生物圈对比，将生矿元素分为６类。提出生矿元素保留于生物体内８种形式，即可溶无机化合物、有机络合物、生矿粒、生矿簇、骨针、骨片、骨壳和骨骼，并以磷为例阐明了生矿元素的富集过程。按照生物作用型式，建议划分生物矿床为堆积、粘结、吸附和储集４种成因类型，分别讨论它们成矿的生物、矿物和矿床，分析形成的环境、过程和阶段，并介绍中国某些著名生物矿床实例。最后总结了生物矿床的地史演化趋向：金属硫化物矿床→条带状硅铁（锰）粘结矿床→叠层状白云岩粘结矿床→磷酸盐和碳酸锰粘结矿床→铅锌和油气储集矿床→煤和油气。     

玉米根微粒体ABA结合蛋白的性质及逆境胁迫的影响

以玉米根微粒体为材料进行的微量放射配体结合（ＭＲＬＢ）实验表明玉米根微粒体膜上存在着ＡＢＡ结合位点,ＡＢＡ与ＡＢＡ结合蛋白（ＡＢＡ－ＢＰ）结合的最适ｐＨ为６．５,结合反应对温度（０℃和２５℃）不太敏感,ＡＢＡ与ＡＢＡ－ＢＰ的结合反应是一个动态平衡过程,５ｍｉｎ即可达最大结合（Ｂｍａｘ）的５０％,３０ｍｉｎ达到最大结合,１ｈ内基本保持不变。胰蛋白酶处理表明此结合位点为蛋白质,冻融实验则表明此蛋白与ＡＢＡ的结合不仅要求其自身具有特定构象而且需要有一定的膜脂环境,ＤＴＴ处理实验结果显示ＡＢＡ－ＢＰ中可能存在着二硫键。逆境处理可以提高玉米根微粒体膜对ＡＢＡ的结合活性,盐胁迫、渗透胁迫、干旱胁迫和热冲激处理分别使结合活性上升３４．９％、１７．８％、２３．１％和１３．３％。     

一例智力低下患者7q~ 标记染色体的来源鉴定

以人类染色体显微切割、PCR技术构建的现有人类染色体特异性和染色体区带特异性探针池作为绘画探针,采用正向染色体绘画技术,结合染色体筛查方法,查明了一例7q~ 标记染色体患者的染色体附加片段来源于3q26→3qter。确定该患者的核型为46,XX,-7, der(7)t(7;3)(7pter→7q32::3q26→3qter)。应用这个策略,能够快速有效地鉴定标记染色体的来源。     

应用FIA型微生物传感器测定谷氨酸含量的研究

目前应用酶或微生物细胞作为分子识别元件构建生物传感器测定谷氨酸含量的研究引起了广泛的兴趣,并陆续有实例报道。在这些报道中人们依据不同的酶催反应选择相应的离子选择性电极,如CO₂电极、NH₄⁺电极等,而测量对象有直接的测定谷氨酸,也有测定谷氨酸单钠的间接测定法。本文选用了可固定大量细胞的固定化细胞柱与流动注入法相结合的测量方法,并根据细胞柱的动力学模型分析动态响应曲线,计算测量结果,提高了测量精度,扩大了测量范围。     

Modulation of membrane dynamics and cell motility by membrane tension

The plasma membrane of most cells is drawn tightly over the cytoskeleton of the cell, resulting in a significant tension being developed in the membrane. The tension in the membrane can be calculated from the force required to separate it from the cytoskeleton; and the force itself can be measured rapidly by using laser tweezers. Recent observations indicate that decreasing membrane tension stimulates endocytosis and increasing tension stimulates secretion. Thus, membrane tension provides a simple physical mechanism to control the area of the plasma membrane. Here, we speculate that tension is a global parameter that the cell uses to control physically plasma membrane dynamics, cell shape and cell motility.     

动物细胞在鼓泡式生物反应器中的死亡速率

通过实验测定,证明生物反应器中细胞死亡速率与气体鼓泡速率成正比而与反应器体积成反比。实验发现气泡大小对细胞死亡速率具有两种作用,一种作用在于影响气泡表面积生成速率;另一种作用则在于影响细胞在气泡表面的吸附程度,其最佳直径为5mm左右。血清和Pluronic F68能显著降低细胞死亡速率,当Pluronic F68浓度达到0．1％时,kd趋于零。所有这些实验结果均与前文提出的生物反应器设计模型具有很好的一致性。     

苯甲醛对果蝇视觉联想记忆的阻断

采用飞行模拟系统,以视觉模式为线索、热惩罚为负强化因子,对于在不同发育时期经受苯甲醛处理过的果蝇的视觉飞行定向条件化进行了检验。苯甲醛气味分别作用于果蝇幼虫和成虫阶段,将阻断果蝇成虫建立视觉联想记忆的能力;雌性果蝇在处女期对苯甲醛气味的接触,会阻断其子代建立视觉联想记忆,这种视觉联想记忆的能力可以通过对其子代连续３代的正常饲养而逐渐得到恢复。
