为丰富产紫杉醇植物内生真菌资源库,从曼地亚红豆杉Taxus media茎中分离得到一株产紫杉醇的内生真菌TMS-26。通过对TMS-26的发酵提取物进行高效液相色谱分析,发现其具有与紫杉醇标准品(4.545 min)相近的色谱特征峰。进一步通过液质联用仪检测发现,内生真菌TMS-26的发酵提取物中具有与紫杉醇标准品((M+Na)+=876)相近的质谱特征峰,表明内生真菌TMS-26能够产生紫杉醇。同时通过传统形态学分类鉴定方法和18S r DNA序列分析、Internal-transcribed spacer(ITS)序列分析等现代分子生物学分类鉴定方法,最终将内生真菌TMS-26鉴定为曲霉属烟曲霉Aspergillus fumigatus,并命名为"烟曲霉TMS-26"。  相似文献

山黧豆(Lathyrus sativus)种子在萌发过程中,根芽中β-N-草酰基-L-2,3-二氨基丙酸(β-ODAP)6d前逐渐增加,尔后不断下降;在水分胁迫条件下,β-ODAP和游离氨基酸含量随水分胁迫的增加而上升,子叶中β-ODAP在3d前高于根芽,然后下降并低于根芽,子叶中β-ODAP含量随着胁迫的增加而降低。  相似文献

Much attention has recently been devoted to the delimitation of species units in Cylindrocladium(Cy.). In this regard the present study focuses on the taxa within the unresolved Cy. floridanum and Cy. spathiphylli species complexes. Maximum parsimony analyses of DNA sequences of ITS, β-tubulin and histone regions of rRNA genes, and mating experiments revealed a geographically isolated species of Cylindrocladium in the Cy. spathiphylli(teleomorph:Calonectria spathiphylli) species complex.Cy. pseudospathiphylli sp. nov. (teleomorph:Ca. pseudospathiphylli sp. nov.) is described as a new phylogenetic, biological and morphological species. It is distinguished from Cy. spathiphylli by being homothallic, having smaller macroconidia, and distinct DNA sequences of β-tubulin and histone genes. Similarly, parsimony analysis of a combined data set also indicated several phylogenetic species to exist within Cy. floridanum(teleomorph:Ca. kyotensis). Based on differences in vesicle morphology and conidium dimensions, the Canadian population of Cy. floridanum, formerly known as Cy. floridanum Group 2, is described as Cy. canadense sp. nov., while a further collection from Hawaii is described as Cy. pacificum sp. nov.  相似文献

Crystal proteins from Bacillus thuringiensis subsp. thompsoni strain HnC are active against the codling moth, Cydia pomonella, a major pest of orchards. Inclusion bodies purified from strain HnC displayed an LC50 of 3.34 × 10−3μg/μl. HnC-purified crystals were tenfold more active than Cry2Aa and Cry1Aa toxins, and 100-fold more toxic than Cry1Ab. The 34-kDa and 40-kDa proteins contained in HnC inclusion bodies were shown to act synergistically. The toxicity of crystal proteins produced by the recombinant B. thuringiensis strain BT-OP expressing the full-length native operon was about tenfold higher than that of the 34-kDa protein. When the gene encoding the non-insecticidal 40-kDa protein, which is not active, was introduced into the recombinant strain producing only the 34-kDa protein, the toxicity was raised tenfold and was similar to that of the strain BT-OP. 目的:探讨规律性耐力运动对脑皮层增龄性老化适应性的作用与机制。方法:将三个不同年龄段的健康SPF级雄性Sprague-Dawley大鼠分为3月龄 (青年,n=20)、13月龄 (中年,n=24)和23月龄 (老年,n=24)组,每组又随机分为静息组和运动组;静息组三组静息,运动组三组实施10周递增负荷规律的中等强度耐力运动:运动方式为跑台运动(坡度0),运动强度从最大摄氧量(V·O2max) 60%~65%逐渐递增到70%~75%,运动时间为10周;取大鼠脑皮层,HE染色测试大鼠脑皮层增龄性形态学变化,检测BDNF和SOD的蛋白表达及突触素-1(SYN1)和CaMK IIα/AMPKα1/ mTOR通路等相关基因。结果:静息各组大鼠的脑皮层结构呈现年龄增龄性衰老变化,脑皮层SOD表达呈增龄性下降趋势,BDNF表达变化呈增龄性上升趋势,SYN1和CaMK IIα表达水平随增龄性趋势变化不大,AMPKα1和SirT2以及 IP3R、AKT1、mTOR mRNA表达水平随年龄变化呈现中年略上升而老年下降趋势;与静息各组大鼠相比,运动各组大鼠脑皮层神经细胞核排列紧密有序,显微镜下观察细胞核的数量明显增加,运动促进大鼠脑皮层SOD、BDNF和突触素SYN1表达水平增加,其中老年大鼠SOD、BDNF表达水平显著上调(P<0.01),青年和老年大鼠SYN1表达水平显著上调(P<0.05),运动上调中年和老年大鼠脑皮层CaMK IIα表达水平上调(P<0.01),而对青年大鼠CaMK IIα表达水平却是下调(P<0.01),运动可上调青年大鼠脑皮层的AMPKα1表达水平(P<0.05),而对中年和老年大鼠AMPKα1的影响不显著,运动均可上调各年龄大鼠脑皮层的SirT2表达水平(P<0.05),运动上调各年龄大鼠脑皮层的IP3R/AKT1/ mTOR表达水平,其中青年IP3R显著上调(P<0.01),青年和中年mTOR显著上调(P<0.01),老年mTOR也显著上调(P<0.05)。结论:耐力运动通过上调BDNF的表达水平,调控CaMK IIα信号、激活AMPK信号通路和IP3R/AKT1/mTOR信号通路,改善脑皮层的突触可塑性。  相似文献

The molecular characterization of symbionts is pivotal for understanding the cross-talk between symbionts and hosts. In addition to valuable knowledge obtained from symbiont genomic studies, the biochemical characterization of symbionts is important to fully understand symbiotic interactions. The bean bug (Riptortus pedestris) has been recognized as a useful experimental insect gut symbiosis model system because of its cultivatable Burkholderia symbionts. This system is greatly advantageous because it allows the acquisition of a large quantity of homogeneous symbionts from the host midgut. Using these naïve gut symbionts, it is possible to directly compare in vivo symbiotic cells with in vitro cultured cells using biochemical approaches. With the goal of understanding molecular changes that occur in Burkholderia cells as they adapt to the Riptortus gut environment, we first elucidated that symbiotic Burkholderia cells are highly susceptible to purified Riptortus antimicrobial peptides. In search of the mechanisms of the increased immunosusceptibility of symbionts, we found striking differences in cell envelope structures between cultured and symbiotic Burkholderia cells. The bacterial lipopolysaccharide O antigen was absent from symbiotic cells examined by gel electrophoretic and mass spectrometric analyses, and their membranes were more sensitive to detergent lysis. These changes in the cell envelope were responsible for the increased susceptibility of the Burkholderia symbionts to host innate immunity. Our results suggest that the symbiotic interactions between the Riptortus host and Burkholderia gut symbionts induce bacterial cell envelope changes to achieve successful gut symbiosis.  相似文献

Background

Human population movement across country borders presents a real challenge for malaria control and elimination efforts in Cambodia and its neighbouring countries. To quantify Plasmodium infection among the border-crossing population, including asymptomatic and artemisinin resistant (AR) parasites, three official border crossing points, one from each of Cambodia''s borders with Thailand, Laos and Vietnam, were selected for sampling.

Methods and Findings

A total of 3206 participants (of 4110 approached) were recruited as they crossed the border, tested for malaria and interviewed. By real-time polymerase chain reaction (RT-PCR), 5.4% of all screened individuals were found to harbour Plasmodium parasites. The proportion was highest at the Laos border (11.5%). Overall there were 97 P. vivax (55.7%), 55 P. falciparum (31.6%), two P. malariae (1.1%) and 20 mixed infections (11.5%). Of identified infections, only 20% were febrile at the time of screening. Of the 24 P. falciparum samples where a further PCR was possible to assess AR, 15 (62.5%) had mutations in the K13 propeller domain gene, all from participants at the Laos border point. Malaria rapid diagnostic test (RDT) pLDH/HRP-2 identified a positivity rate of 3.2% overall and sensitivity compared to RT-PCR was very low (43.1%). Main individual risk factors for infection included sex, fever, being a forest-goer, poor knowledge of malaria prevention methods and previous malaria infection. Occupation, day of the week and time of crossing (morning vs. afternoon) also appeared to play an important role in predicting positive cases.

Conclusions

This study offers a novel approach to identify asymptomatic infections and monitor AR parasite flow among mobile and migrant populations crossing the borders. Similar screening activities are recommended to identify other hot borders and characterise potential hot spots of AR. Targeted “customised” interventions and surveillance activities should be implemented in these sites to accelerate elimination efforts in the region.  相似文献

Bacillus thuringiensis is a widely used biopesticide that produced various insecticidal active substances during its life cycle. Separation and purification of numerous insecticide active substances have been difficult because of the relatively short half-life of such substances. On the other hand, substances can be synthetized at different times during development, so samples at different stages have to be studied, further complicating the analysis. A dual genomic and proteomic approach would enhance our ability to identify such substances, and particularily using mass spectrometry-based proteomic methods. The comparative analysis for genomic and proteomic data have showed that not all of the products deduced from the annotated genome could be identified among the proteomic data. For instance, genome annotation results showed that 39 coding sequences in the whole genome were related to insect pathogenicity, including five cry genes. However, Cry2Ab, Cry1Ia, Cytotoxin K, Bacteriocin, Exoenzyme C3 and Alveolysin could not be detected in the proteomic data obtained. The sporulation-related proteins were also compared analysis, results showed that the great majority sporulation-related proteins can be detected by mass spectrometry. This analysis revealed Spo0A~P, SigF, SigE(+), SigK(+) and SigG(+), all known to play an important role in the process of spore formation regulatory network, also were displayed in the proteomic data. Through the comparison of the two data sets, it was possible to infer that some genes were silenced or were expressed at very low levels. For instance, found that cry2Ab seems to lack a functional promoter while cry1Ia may not be expressed due to the presence of transposons. With this comparative study a relatively complete database can be constructed and used to transform hereditary material, thereby prompting the high expression of toxic proteins. A theoretical basis is provided for constructing highly virulent engineered bacteria and for promoting the application of proteogenomics in the life sciences.  相似文献

MgtC is required for intramacrophage replication of intracellular pathogens and growth in low Mg(2+) medium. A link between these two phenotypes has been proposed due to putative Mg(2+) deprivation inside phagosome. MgtC is part of a family of proteins that share a conserved N-terminal transmembrane domain and a variable C-terminal domain. A combination of predictive and experimental approaches indicates that the Salmonella MgtC C-terminal domain is cytoplasmic, adopts a fold also found in metal transporters and RNA interacting domain, and does not bind Mg(2+). MgtC homologues from diverse gamma-proteobacteria, including the extracellular pathogens Yersinia pestis, Photorhabdus luminescens and Pseudomonas aeruginosa, have been expressed in a SalmonellaDeltamgtC strain. The Y. pestis MgtC fully replaced the Salmonella MgtC whereas P. luminescens or P. aeruginosa MgtC complemented only in low Mg(2+) medium, thus dissociating for the first time the two MgtC-related phenotypes. In addition, we identified single amino acids changes that prevent or promote MgtC role in macrophages without affecting MgtC role in low Mg(2+) culture. A SalmonellaDeltamgtC strain showed elongated and autoaggregated bacteria in low Mg(2+) medium but not in macrophages. Taken together our results suggest that MgtC has a dual role when bacteria localize in macrophages or low Mg(2+) environment.  相似文献
