Organocatalysts bearing sulfide or sulfone functions (1a-d) were studied for the direct asymmetric Michael addition of ketones and alkylidene malonates. The organocatalyst (S)-2-((naphthalen-2-ylthio)methyl)pyrrolidine, bearing a pyrrolidine and a sulfide moiety, showed a very high catalytic activity in the absence of additives. The reaction condition is mild, and the Michael adducts were obtained in very good enantioselectivities (up to 96%), diastereoselectivities (up to 95:5), and chemical yields (up to 95%).  相似文献

Conventional radiography is insensitive for early and accurate estimation of the mal-alignment and wear of knee prostheses. The two-staged (rough and fine) registration of the model-based RSA technique has recently been developed to in vivo estimate the prosthetic pose (i.e, location and orientation). In the literature, rough registration often uses template match or manual adjustment of the roentgen images. Additionally, possible error induced by the nonorthogonality of taking two roentgen images neither examined nor calibrated prior to fine registration. This study developed two RSA methods for automate the estimation of the prosthetic pose and decrease the nonorthogonality-induced error. The predicted results were validated by both simulative and experimental tests and compared with reported findings in the literature. The outcome revealed that the feature-recognized method automates pose estimation and significantly increases the execution efficiency up to about 50 times in comparison with the literature counterparts. Although the nonorthogonal images resulted in undesirable errors, the outline-optimized method can effectively compensate for the induced errors prior to fine registration. The superiority in automation, efficiency, and accuracy demonstrated the clinical practicability of the two proposed methods especially for the numerous fluoroscopic images of dynamic motion.  相似文献

Understanding the determinants of protein stability remains one of protein science's greatest challenges. There are still no computational solutions that calculate the stability effects of even point mutations with sufficient reliability for practical use. Amino acid substitutions rarely increase the stability of native proteins; hence, large libraries and high-throughput screens or selections are needed to stabilize proteins using directed evolution. Consensus mutations have proven effective for increasing stability, but these mutations are successful only about half the time. We set out to understand why some consensus mutations fail to stabilize, and what criteria might be useful to predict stabilization more accurately. Overall, consensus mutations at more conserved positions were more likely to be stabilizing in our model, triosephosphate isomerase (TIM) from Saccharomyces cerevisiae. However, positions coupled to other sites were more likely not to stabilize upon mutation. Destabilizing mutations could be removed both by removing sites with high statistical correlations to other positions and by removing nearly invariant positions at which "hidden correlations" can occur. Application of these rules resulted in identification of stabilizing mutations in 9 out of 10 positions, and amalgamation of all predicted stabilizing positions resulted in the most stable yeast TIM variant we produced (+8 °C). In contrast, a multimutant with 14 mutations each found to stabilize TIM independently was destabilized by 2 °C. Our results are a practical extension to the consensus concept of protein stabilization, and they further suggest the importance of positional independence in the mechanism of consensus stabilization.  相似文献

Activated sludge has been utilized for the treatment of volatile organic compounds (VOCs) which are emitted from industrial processes. Nevertheless, activated sludge systems often suffer from the problem caused by concentration gradients as well as pressure drops. Channeling is also a major problem in the treatment process. As the bed height of the packed activated sludge system increases, the pressure drop increases accordingly. To solve these problems, we proposed immobilized activated sludge column reactors for treating VOCs in air. The immobilization material used to mix with activated sludge was properly selected in this work. Elemental compositions of these materials were analyzed. In this study, we also proposed a VOC feed system so that more stable inlet concentrations could be achieved. Hence, the equipment and operating costs were reduced and the problem of VOCs leaking from peristaltic pumps was avoided. The moisture content of the system was well maintained and better VOC removal efficiency was achieved. With an operation condition of progressive VOC inlet concentrations, better removal efficiency of benzene and toluene was then obtained. In conclusion, by the utilization of immobilization materials selected from wastes as well as immobilized activated sludge column reactors, significant removal efficiency for both benzene and toluene was demonstrated.  相似文献

Infections caused by enterohemorrhagic Escherichia coli (EHEC) can lead to diarrhea with abdominal cramps and sometimes are complicated by severe hemolytic uremic syndrome. EHEC secretes effector proteins into host cells through a type III secretion system that is composed of proteins encoded by a chromosomal island, locus for the enterocyte effacement (LEE). EspA is the major component of the filamentous structure connecting the bacteria and the host''s cells. Synthesis and secretion of EspA must be carefully controlled since the protein is prone to polymerize. CesAB, CesA2, and EscL have been identified as being able to interact with EspA. Furthermore, the intracellular level of EspA declines when cesAB, cesA2, and escL are individually deleted. Here, we report a LEE gene named l0033, which also affects the intracellular level of EspA. We renamed l0033 as escA since its counterpart in enteropathogenic E. coli has been recently described. Similar to CesAB, EscL, and CesA2, EscA interacts with EspA and enhances the protein stability of EspA. However, EscA is also able to interact with inner membrane-associated EscL, CesA2, and EscN, but not with cytoplasmic CesAB. In terms of gene organizations, escA locates in LEE3. Expression of EscA is faithfully regulated via Mpc, the first gene product of LEE3. Since Mpc is tightly regulated to low level, we suggest that EscA is highly synchronized and critical to the process of escorting EspA to its final destination.  相似文献

There is considerable need for accurate suicide risk assessment for clinical, screening, and research purposes. This study applied the tripartite affect-behavior-cognition theory, the suicidal barometer model, classical test theory, and item response theory (IRT), to develop a brief self-report measure of suicide risk that is theoretically-grounded, reliable and valid. An initial survey (n = 359) employed an iterative process to an item pool, resulting in the six-item Suicidal Affect-Behavior-Cognition Scale (SABCS). Three additional studies tested the SABCS and a highly endorsed comparison measure. Studies included two online surveys (Ns = 1007, and 713), and one prospective clinical survey (n = 72; Time 2, n = 54). Factor analyses demonstrated SABCS construct validity through unidimensionality. Internal reliability was high (α = .86-.93, split-half = .90-.94)). The scale was predictive of future suicidal behaviors and suicidality (r = .68, .73, respectively), showed convergent validity, and the SABCS-4 demonstrated clinically relevant sensitivity to change. IRT analyses revealed the SABCS captured more information than the comparison measure, and better defined participants at low, moderate, and high risk. The SABCS is the first suicide risk measure to demonstrate no differential item functioning by sex, age, or ethnicity. In all comparisons, the SABCS showed incremental improvements over a highly endorsed scale through stronger predictive ability, reliability, and other properties. The SABCS is in the public domain, with this publication, and is suitable for clinical evaluations, public screening, and research.  相似文献

This study presents 1 use of optical coherence tomography (OCT) angiography technique to examine neurovascular coupling effect. Repeated B‐scans OCT recording is performed on the rat somatosensory cortex with cranial window preparation while its contralateral forepaw is electrically stimulated to activate the neurons in rest. We use an intensity‐based Doppler variance (IBDV) algorithm mapped cerebral blood vessels in the cortex, and the temporal alteration in blood perfusion during neurovascular activation is analyzed using the proposed IBDV quantitative parameters. By using principal component analysis‐based Fuzzy C Means clustering method, the stimulus‐evoked vasomotion patterns were classified into 3 categories. We found that the response time of small vessels (resting diameter 14.9 ±6.6 μm), middle vessels (resting diameter 21.1 ±7.9 μm) and large vessels (resting diameter 50.7 ±6.5 μm) to achieve 5% change of vascular dilation after stimulation was 1.5, 2 and 5.5 seconds, respectively. Approximately 5% peak change of relative blood flow (RBF) in both small and middle vessels was observed. The large vessels react slowly and their responses nearly 4 seconds delayed, but no significant change in RBF of the large vessels was seen.   相似文献

Recombination between RNA sequences plays a role in the fast evolution of a few viruses. There has been no report on hepatitis D virus (HDV) recombination. In this study, we analyzed genetic recombination of HDV and its possible impact on evolution and clinical course. The aligned HDV sequences allowed us to construct a phylogenetic tree which supported the notion of distinct lineages of HDV. The tree was also used in the analysis of recombination using partial likelihoods assessed through optimization. Nine segments of the HDV genome with significant levels of genetic recombination were detected. Five segments were in the hypervariable region, and four were in the delta-antigen- coding region. None could be found in the well-conserved autocleavage region that is essential for replication. Recombination occurred both between and within types. The results of this study indicated that the remarkable variation in HDV genomic sequences, particularly in the hypervariable region, among different genotypes may at least partly result from multiple episodes of genetic recombination during evolution. Genetic recombination may play a significant role in increasing genetic diversity. Importantly, a genetic recombination (nt 1082-1093) occurred in one of the immunogenic domains of hepatitis delta virus antigen recognized by human and woodchuck antibodies (amino acids 174-195). Genetic recombination also occurred at another segment between nt 1517 and 1535, which was close to one of the predicted T-cell epitopes (amino acids 26-41). In longitudinal analysis of HDV genomes at different time points during chronic infection, novel dominant HDV strains with amino acid changes at these epitopes usually emerged after severe hepatitis attacks. In the comparison of HDV clones during or shortly after flare-up of liver disease, Ka/Ks ratios of > 1 were frequently found, suggesting Darwinian positive selection. Therefore, recombination in these two segments may play an important role for HDV in the evasion of immunity.  相似文献

Introduction: Schwannoma of the male genital system is very uncommon and is mostly treated by surgery. However, prostatic schwannoma presenting with elevated prostate-specific antigen (PSA) level and treated conservatively are extremely rare. Case presentation: Herein, we present a rare case of a prostatic schwannoma in a 65-year-old man who initially presented with an elevated PSA level. Digital rectal examination revealed an enlarged prostate with a palpable hard nodule on the left side. Transrectal ultrasonography revealed an enlarged prostate with a well-defined homogeneously hypoechoic nodule in the left peripheral lobe. Biopsy was done, and histopathology revealed a prostatic schwannoma. Conservative treatment with regular image follow-up was done per the patient’s preference. Mild PSA progression but no worsening of symptoms was found in 6 years of follow-up. Conclusions: PSA elevation could be a rare presentation of prostatic schwannoma. Treatment options other than surgery, such as conservative treatment with close observation, could be feasible for these rare tumors and long-term survivorship can be achieved.  相似文献

Background and aims

Limited information is available on comparing the iron plaque formation capabilities and their effect on arsenic (As) uptake by different rice plant genotypes grown in As-contaminated soils. This study investigates the effect of iron plaque on As uptake in different rice genotypes grown in As-contaminated soils from the Guandu Plain of northern Taiwan.

Methods

Twenty-eight rice genotypes including 14 japonica and 14 indica genotypes were used in this study. Rice seedlings were grown in As-contaminated soils for 38 days. The iron plaque formed on the rice roots were extracted using dithionite–citrate–bicarbonate. The concentrations of As, Fe, and P in soil solutions, iron plaque, and plants were measured. The speciation of As in the root’s iron plaque was determined by As K-edge X-ray absorption near-edge structure spectroscopy (XANES).

Results

The amounts of iron plaque formation on roots were significantly different among 28 tested rice genotypes, and 75.7–92.8 % of As uptake from soils could be sequestered in iron plaque. However, there were no significant negative correlations between the amounts of Fe or As in the iron plaque and the content of As accumulated in rice plants of tested genotypes. XANES data showed that arsenate was the predominant As species in iron plaque, and there were difference in the distribution of As species among different rice genotypes.

Conclusions

The iron plaque can sequester most of As uptake from soils no matter what rice genotypes used in this study. However, the iron plaque alone did not control the extent of As accumulation in rice plants from As-contaminated soils among 28 tested rice genotypes. Low As uptake genotypes of rice selected from this study can be recommended to be grown in the As-contaminated soils.  相似文献
