Matriptase is a member of the family of type II transmembrane serine proteases that is essential for development and maintenance of several epithelial tissues. Matriptase is synthesized as a single-chain zymogen precursor that is processed into a two-chain disulfide-linked form dependent on its own catalytic activity leading to the hypothesis that matriptase functions at the pinnacle of several protease induced signal cascades. Matriptase is usually found in either its zymogen form or in a complex with its cognate inhibitor hepatocyte growth factor activator inhibitor 1 (HAI-1), whereas the active non-inhibited form has been difficult to detect. In this study, we have developed an assay to detect enzymatically active non-inhibitor-complexed matriptase by using a biotinylated peptide substrate-based chloromethyl ketone (CMK) inhibitor. Covalently CMK peptide-bound matriptase is detected by streptavidin pull-down and subsequent analysis by Western blotting. This study presents a novel assay for detection of enzymatically active matriptase in living human and murine cells. The assay can be applied to a variety of cell systems and species.  相似文献

The distribution of nitrification potential (NP) with depth in sediment and season was investigated in a shallow sandy sediment (0.5 m water) and a deeper muddy sediment (17m water). In both sediments, nitrifying bacteria were present in the anoxic strata (oxygen penetration was 5 mm below the surface). The NP at 6–8 cm depth in the sediment was 50% and 10% of the surface NP at the sandy and muddy sediment, respectively. It is suggested that bioturbation and physical disturbance of the sediment were the most likely reasons for this distribution. The NP increased as sediment temperature decreased. This effect was less marked in the muddy sediment. It is concluded that during the summer, the numbers or specific activity of nitrifying bacteria diminished for the following reasons: There was decreased O2 penetration into the sediment and increased competition for O2 by heterotrophs; there was increased competition for NH4 + and there was inhibition by H2S. These effects counteracted the potentially higher growth rates and increased rates of NH4 + production at the elevated summer temperatures. The potential nitrification rates in the upper 1 cm, which were measured at 22°C, were converted to calculated rates at the in situ temperature (Q10=2.5) and in situ oxygen penetration. These calculated rates were shown to closely resemble the measured in situ rates of nitrification. The relationship between the in situ rates of nitrification and the nitrification potential is discussed.  相似文献

Few developments in microbiological diagnostics have had such a rapid impact on species level identification of microorganisms as matrix-assisted laser desorption/ionization time of flight mass spectrometry (MALDI-TOF MS). Conventional differentiation methods rely on biochemical criteria and require additional pre-testing and lengthy incubation procedures. In comparison, MALDI-TOF MS can identify bacteria and yeast within minutes directly from colonies grown on culture plates. This radically new, methodically simple approach profoundly reduces the cost of consumables and time spent on diagnostics. The reliability and accuracy of the method have been demonstrated in numerous studies and different systems are already commercially available. Novel applications of the system besides microbial species level identification are also being explored. This includes identification of pathogens from positive blood cultures or directly from patient samples, such as urine. Currently, intriguing MALDI-TOF MS developments are being made regarding the phenotypic detection of certain antibiotic resistance mechanisms, e.g., β-lactamases and carbapenemases. This mini review provides an overview of the literature in the field and also includes our own data and experiences gathered from over 4 years of routine MALDI-TOF MS use in a university hospital’s microbiological diagnostics facility.  相似文献

The zebrafish (Danio rerio) has proven to be a powerful vertebrate model system for the genetic analysis of developmental pathways and is only beginning to be exploited as a model for human disease and clinical research. The attributes that have led to the emergence of the zebrafish as a preeminent embryological model, including its capacity for forward and reverse genetic analyses, provides a unique opportunity to uncover novel insights into the molecular genetics of cancer. Some of the advantages of the zebrafish animal model system include fecundity, with each female capable of laying 200-300 eggs per week, external fertilization that permits manipulation of embryos ex utero, and rapid development of optically clear embryos, which allows the direct observation of developing internal organs and tissues in vivo. The zebrafish is amenable to transgenic and both forward and reverse genetic strategies that can be used to identify or generate zebrafish models of different types of cancer and may also present significant advantages for the discovery of tumor suppressor genes that promote tumorigenesis when mutationally inactivated. Importantly, the transparency and accessibility of the zebrafish embryo allows the unprecedented direct analysis of pathologic processes in vivo, including neoplastic cell transformation and tumorigenic progression. Ultimately, high-throughput modifier screens based on zebrafish cancer models can lead to the identification of chemicals or genes involved in the suppression or prevention of the malignant phenotype. The identification of small molecules or gene products through such screens will serve as ideal entry points for novel drug development for the treatment of cancer. This review focuses on the current technology that takes advantage of the zebrafish model system to further our understanding of the genetic basis of cancer and its treatment.  相似文献

Sequence analysis of the Legionella micdadei groELS operon   总被引:4,自引:0,他引:4  
A 2.7 kb DNA fragment encoding the 60 kDa common antigen (CA) and a 13 kDa protein of Legionella micdadei was sequenced. Two open reading frames of 57,677 and 10,456 Da were identified, corresponding to the heat shock proteins GroEL and GroES, respectively. Typical -35, -10, and Shine-Dalgarno heat shock expression signals were identified upstream of the L. micdadei groEL gene. Further upstream, a poly-T region, also a feature of the sigma 32-regulated Escherichia coli groELS heat shock operon, was found. Despite the high degree of homology of the expression signals in E. coli and L. micdadei, Western blot analysis with an L. micdadei specific anti-groEL antibody did not reveal a significant increase in the amount of the GroEL protein during heat shock in L. micdadei or in the recombinant E. coli expressing L. micdadei GroEL.  相似文献

Objective

Childhood cancer is rare and symptoms tend to be unspecific and vague. Using the utilization of health care services as a proxy for symptoms, the present study seeks to determine when early symptoms of childhood cancer are seen in general practice.

Methods

A population-based matched comparative study was conducted using nationwide registry data. As cases, all children in Denmark below 16 years of age (N = 1,278) diagnosed with cancer (Jan 2002-Dec 2008) were included. As controls, 10 children per case matched on gender and date of birth (N = 12,780) were randomly selected. The utilization of primary health care services (daytime contacts, out-of-hours contacts and diagnostic procedures) during the year preceding diagnosis/index date was measured for cases and controls.

Results

During the six months before diagnosis, children with cancer used primary care more than the control cohort. This excess use grew consistently and steadily towards the time of diagnosis with an IRR = 3.19 (95%CI: 2.99–3.39) (p<0.0001) during the last three months before diagnosis. Children with Central Nervous System (CNS) tumours had more contacts than other children during the entire study period. The use of practice-based diagnostic tests and the number of out-of-hours contacts began to increase four to five months before cancer diagnosis.

Conclusions

The study shows that excess health care use, a proxy for symptoms of childhood cancer, occurs months before the diagnosis is established. Children with lymphoma, bone tumour or other solid tumours had higher consultation rates than the controls in the last five months before diagnosis, whereas children with CNS tumour had higher consultation rates in all twelve months before diagnosis. More knowledge about early symptoms and the diagnostic pathway for childhood cancer would be clinically relevant.  相似文献

Flower biology and pollination of Moneses uniflora, Orlhilia secunda, Pyrola minor, P. rotundifolia, P. chlorantha, and Chimaphila umbellata are described and discussed in relation to patterns in sexual traits and possible evolution of buzz-pollination within the group. The large number of pollen grains are packed into units of monads in Orthilia, tetrads in Moneses and Pyrola, or polyads in Chimaphila. Pollen is the sole reward to visiting insects except in the nectar-producing O. secunda and C. umbellata. Correlations are present between several sexual traits. Pollen: ovule ratios are very low, indicating efficient pollen transfer. Use of polyads may reduce the number of siring males. This trend may be counteracted by larger stigmas as in C. umbellata. Increasing pollen unit size may enhance relatedness of seeds, reduce abortion, and improve seed set. This may explain the evolution, within the Pyrolaceae and other families, of higher ovule numbers with larger pollen units. The plants are mainly pollinated by bumblebees. Chimaphila umbellata is visited by nectar-collectors; Moneses uniflora, Pyrola minor, P. rotundifolia, and P. chlorantha are visited exclusively by pollen-buzzers; and Orthilia secunda is visited by both nectar-collectors and pollen-buzzers. Nectar secretion is suggested to be an ancient attractant in the group with C. umbellata having the least specialized pollination system, and buzz-pollination in M. uniflora and in the studied Pyrola species regarded as an advanced system.  相似文献

In the present study we investigated the ability of high‐throughput FTIR spectroscopy in combination with multivariate data analysis to reveal if any combinatory effects of fatty acids in mixture are present in liver HepG2 cell cultures after three days of exposure. For this investigation we used an experimental mixture design containing three different octadecenoic acids (oleic acid: C18 : 1 cis‐ 9, elaidic acid: C18 : 1 trans‐ 9 and vaccenic acid: C18 : 1 trans‐ 11) of a total concentration of 100 μM. The results obtained revealed both additive and non‐additive effects of individual fatty acids when combined in mixtures. Furthermore, we demonstrate by use of scanning electron microscopy that cells are preserved as intact structures ensuring that FTIR measurements are obtained on whole cell keeping cell compounds in their natural surroundings during measurements.
