In seasonally dry environments, woody species have different survival strategies. However, little is known about how environmental variables affect the phenology and water dynamics of these species. We aim to understand which variables initiate the vegetative phenophases of species in a tropical semiarid climate at 3°S latitude, where variation in photoperiod is minimal and rainfall is seasonal. We hypothesize that groups of species with similar vegetative phenologies, under similar conditions, are functionally similar in terms of water storage and use. We analyse the relationship between functional characteristics related to the acquisition and utilization of water, such as wood density, water storage capacity, water potential and vegetative phenology. The attributes were ordered by multidimensional scaling, and a multiple response permutation procedure was used to test consistency of the groups. Canonical correspondence analysis and Mantel tests were used to evaluate the phenophase response to environmental variables. We found four functional groups: (i) deciduous low wood density, which lose 75% of their leaves one month before the end of the rains; (ii) evergreen high wood density; (iii) early deciduous high wood density, which lose 75% of their leaves one month after the end of the rains; and (iv) late deciduous high wood density, which lose 75% of their leaves two months after the end of the rains. As expected, the vegetative phenodynamics of the deciduous high wood density group were mainly influenced by water availability. The evergreens did not show a correlation with rainfall. Only leaf shedding of the late deciduous, and the vegetative phenophases of the evergreens, responded to an increase in temperature and photoperiod. Bud‐break responded to increased photoperiod and soil humidity in the deciduous low wood density group. The foliar periodicity groups can be explained by the presence of species that differ mainly in their mechanisms of water acquisition and use. 相似文献
Peptoids are versatile peptidomimetic molecules with wide-ranging applications from drug discovery to materials science. An understanding of peptoid sequence features that contribute to both their three-dimensional structures and their interactions with lipids will expand functions of peptoids in varied fields. Furthermore, these topics capture the enthusiasm of undergraduate students who prepare and study diverse peptoids in laboratory coursework and/or in faculty led research. Here, we present the synthesis and study of 21 peptoids with varied functionality, including 19 tripeptoids and 2 longer oligomers. We observed differences in fluorescence spectral features for 10 of the tripeptoids that correlated with peptoid flexibility and relative positioning of chromophores. Interactions of representative peptoids with sonicated glycerophospholipid vesicles were also evaluated using fluorescence spectroscopy. We observed evidence of conformational changes effected by lipids for select peptoids. We also summarize our experiences engaging students in peptoid-based projects to advance both research and undergraduate educational objectives in parallel. 相似文献
The levels of organic pollutants, such as optical brightener (OB) compounds, in the global environment have been increasing in recent years. The toxicological effects and signal transduction systems associated with OB toxicity have not been thoroughly studied. The ubiquitin-proteasome system (UPS) plays a crucial role in regulating multiple essential cellular processes, and proteasome-associated cysteine deubiquitinases (DUBs), ubiquitin C-terminal hydrolase L5 (UCHL5) and USP14, are two major regulators for (de)ubiquitination and stability of many important target proteins. Therefore, potential inhibition of UCHL5 and USP14 activities by some environmental chemicals might cause in vivo toxicity. In the current study we hypothesize that electrophilic OB compounds, such as 4,4′-diamino-2,2′-stilbenedisulfonic acid(DAST), fluorescent brightener 28 (FB-28) and FB-71, can interact with the catalytic triads (CYS, HIS, and ASP) of UCHL5 and USP14 and inhibit their enzymatic activities, leading to cell growth suppression. This hypothesis is supported by our findings presented in this study. Results from in silico computational docking and ubiquitin vinyl sulfone assay confirmed the UCHL5/USP14-inhibitory activities of these OB compounds that have potencies in an order of: FB-71 > FB-28 > DAST. Furthermore, inhibition of these two proteasomal DUBs by OBs resulted in cell growth inhibition and apoptosis induction in two human breast cancer cell models. In addition, we found that OB-mediated DUB inhibition triggers a feedback reaction in which expression of UCHL5 and USP14 proteins is increased to compromise the suppressed activities. Our study suggests that these commonly used OB compounds may target and inhibit proteasomal cysteine DUBs, which should contribute to their toxicological effects in vivo. 相似文献
Gonad development of the silver therapon Leiopotherapon plumbeus in two volcanic crater lake habitats (Sampaloc Lake, Taal Lake) in south Luzon, Philippines was examined during the annual reproductive cycle. The minimum body size‐at‐maturity of fish in these two lake habitats was also compared. Four gonad development stages were characterized as basis for the classification of ovarian (immature, maturing, mature, spawned) and testicular maturation (immature, maturing, mature) phases. The occurrence of all development stages in individual gonads suggest an asynchronous development whereby advanced stages are recruited continuously from a pool of younger stage germ cells to result in elevated female and male GSI throughout the annual cycle due to active gonadogenesis. Together with the increasing occurrence of advanced stage oocytes and spermatozoa from March until October, the elevated GSI of fish may indicate peak gonadal growth during the onset of the dry season (December–January) for eventual spawning from the beginning (May–June) until the end of the wet season (October–November). In both lake habitats, male fish were smaller than females but, regardless of sex, the minimum size‐at‐maturity of fish in Sampaloc Lake was significantly smaller than fish in Taal Lake. Overall, asynchronous development during oogenesis and spermatogenesis allows for year‐round reproduction of silver therapon, with elevated gonad growth in the dry season in preparation for spawning during the wet season. Compared with fish in Taal Lake, a smaller size‐at‐maturity of fish in Sampaloc Lake may be a response of the wild fishery stock to long‐term high fishing mortality and degradation of the lake habitat. 相似文献
Several toxic effects arise from Al's presence in living systems, one of them being the alteration of the natural role of enzymes and non-enzyme proteins. Al(III) is capable of entering protein active sites that in normal conditions should be occupied by other metals. Even if Mg(II) is an ubiquitous metal in biological systems, the interference of aluminium in magnesium metabolism is not clear yet. In this work, a systematic study of the affinity of Al(III) for different protein binding sites is presented, with special attention on structural parameters, the role of the charge and the presence of different ligands in the protein cavity. The specificity of the binding site for Al(III) against Mg(II) has been studied, and also the thermodynamical propensity of a Mg(II)/Al(III) exchange. Stevioside is a natural sweetener obtained from the leaves of Stevia rebaudiana. It is a glycoside of steviol and other glycosides of the same aglycone are also found in the plant. Stevioside is usually the major component in commercial products, but it is not the one with the best organoleptic properties. It has a bitter aftertaste and, for this reason, attempts have been made in order to modify its molecule. In this work, the commercial and purified stevioside were modified by hydrolytic enzymes from Gibberella fujikuroi. A screening was carried out on six strains of the fungus in order to select the most active. The production of the enzymes by the fungi was induced by its culture in a medium containing stevioside as the sole carbon source and the enzymatic extract was then used in the experiments. The products obtained were analyzed by HPLC-UV and HPLC-MS/MS. The results showed a significant increase in the concentration of rebaudioside A in the final product, which has better organoleptic properties than stevioside. 相似文献
Olfactory ensheathing cells (OECs) are the main glial cell type that populates mammalian olfactory nerves. These cells have a great capacity to promote the regeneration of axons when transplanted into the injured adult mammalian CNS. However, little is still known about the molecular mechanisms they employ in mediating such a task. Brain-derived neurotrophic factor (BDNF) was identified as a candidate molecule in a genomic study that compared three functionally different OEC populations: Early passage OECs (OEC Ep), Late passage OECs (OEC Lp) and the OEC cell line TEG3 [Pastrana, E., Moreno-Flores, M.T., Gurzov, E.N., Avila, J., Wandosell, F., Diaz-Nido, J., 2006. Genes associated with adult axon regeneration promoted by olfactory ensheathing cells: a new role for matrix metalloproteinase 2. J. Neurosci. 26, 5347-5359]. We have here set out to determine the role played by BDNF in the stimulation of axon outgrowth by OECs. We compared the extracellular BDNF levels in the three OEC populations and show that it is produced in significant amounts by the OECs that can stimulate axon regeneration in adult retinal neurons (OEC Ep and TEG3) but it is absent from the extracellular medium of OEC Lp cells which lack this capacity. Blocking BDNF signalling impaired axonal regeneration of adult retinal neurons co-cultured with TEG3 cells and adding BDNF increased the proportion of adult neurons that regenerate their axons on OEC Lp monolayers. Combining BDNF with other extracellular proteins such as Matrix Metalloproteinase 2 (MMP2) further augmented this effect. This study shows that BDNF production by OECs plays a direct role in the promotion of axon regeneration of adult CNS neurons. 相似文献

Palm fruits show great structural complexity, and in-depth studies of their development are still scarce. This work aimed to define the developmental stages of the fruit of the neotropical palm Butia capitata and to characterize the ontogenesis of its pericarp. Biometric, anatomical, and histochemical evaluations were performed on pistillate flowers and developing fruits. The whole fruit develops in three phases: (I) histogenesis (up to 42 days after anthesis – DAA), when the topographic regions of the pericarp are defined; (II) pyrene maturation (42 to 70 DAA), when the sclerified zone of the pericarp is established; and (III) mesocarp maturation (70 to 84 DAA), when reserve deposition is completed. During pericarp ontogenesis (i) the outer epidermis and the outer mesophyll of the ovary give origin to the exocarp (secretory epidermis, collenchyma, parenchyma, sclerenchyma, and vascular bundles); (ii) the median ovarian mesophyll develops into the mesocarp, with two distinct topographical regions; (iii) the inner ovarian epidermis originates the endocarp; and in the micropylar region, it differentiates into the germination pore plate, a structure that protects the embryo and controls germination. (iv) Most of the inner region of the mesocarp fuses with the endocarp and, both lignified, give rise to the stony pyrene; (v) in the other regions of the mesocarp, carbohydrates and lipids are accumulated in a parenchyma permeated with fiber and vascular bundles. The development of the B. capitata pericarp presents high complexity and a pattern not yet reported for Arecaceae, which supports the adoption of the Butia-type pyrenarium fruit class.
