Restriction-map variation with the yellow-achaete-scute region in five populations of Drosophila melanogaster 总被引:9,自引:0,他引:9
It has been proposed that the degree of recombination for a genomic region
will affect the level of both nucleotide heterozygosity and the density of
transposable elements. Both features of genomic diversity have been
examined in a number of recent reports for regions undergoing relatively
normal levels of recombination in Drosophila melanogaster. In this study
the genomic variation associated with yellow-achaete- scute loci located at
the tip of the X chromosome is examined by six- cutter restriction mapping.
In this region, as usual for regions adjacent to telomeres, crossing-over
is dramatically reduced, and published studies of visible mutants indicate
extremely little restriction-map variation. Eight six-cutter restriction
endonucleases were used to locate sequence variation in 14- and 16.5-kb
regions in 109 lines sampled from North America, Africa, and Europe. The
overall level of heterozygosity is estimated as 0.29%. Nine large
insertions, all presumed to be transposable elements, were observed.
Base-pair heterozygosity appears to be reduced compared with regions having
normal levels of recombination. The estimated heterozygosity is much higher
than reported in earlier studies of restriction-map variation among visible
mutations in the complex. The incidence of large insertions is not elevated
compared with that in other regions of the genome. This suggests that
asymmetric synapsis and exchange is not an important mechanism for the
elimination of transposable elements.

Tandem Scanning Confocal Microscopy (TSCM) allows one to section optically into and record real-time images of living organs and tissues in a noninvasive fashion. In this paper, we will present some initial TSCM observations of subcapsular nephrons in the living, intact kidneys of Munich-Wistar rats and evaluate the nephron's responses to temporary ischemia and to intravenous infusion of mannitol. The rats were anesthetized with Inactin and a laparotomy performed to expose the kidneys. Using a TSCM equipped with a 20 x water-immersion objective, we optically sectioned through the intact kidney capsule and recorded real-time images of living subcapsular glomeruli and uriniferous tubules. The proximal tubule brush border was highly reflective and allowed us to distinguish between the first and second segments of the proximal tubules as well as the distal tubules. Cellular elements of the blood could be seen passing rapidly through peritubular capillaries and individual glomerular capillary loops. With fluorescent filters in place, intravenously injected carboxyfluorescein was seen to pass through the glomerular capillary loops and then progressively through the different segments of the uriniferous tubules. Ligation of the renal artery resulted in rapid swelling of proximal tubule cells into the tubular lumens, loss of reflectiveness of the microvillous brush borders, and closure of the peritubular capillary spaces. Upon release of the ligature, the proximal tubule lumens again became patent, often opening up abruptly and in a zipper-like fashion down the length of the tubules. Increasing the glomerular filtration rate by intravenous infusion of mannitol resulted in increases in tubular luminal and perimeter dimensions. Mannitol also acted as an effective impermeant osmotic agent and prevented most of the cellular swelling which was otherwise seen in response to renal ischemia. 相似文献

This study evaluated whether the intensity of previously smelled odors
could unintentionally influence the subsequent judgement of odor intensity.
The predicted context effect was based on the adaptation- level theory.
Before and 25 min after either WEAK or STRONG biasing odor concentrations,
51 subjects were required to rate the intensity of 10 different odor
concentrations of California Orange Oil. After the WEAK bias, subjects
judged the odor intensity as being stronger than they did after the STRONG
bias. Thus the intensity of odors smelled 25 min earlier can
unintentionally influence subsequent odor intensity judgement. The findings
are discussed in the light of two alternative explanations, namely, a
central implicit memory process and a stimulus- level-based change at the
peripheral level.

Aminoacyl-tRNAs (aa-tRNAs) are the essential substrates for translation. Most aa-tRNAs are formed by direct aminoacylation of tRNA catalyzed by aminoacyl-tRNA synthetases. However, a smaller number of aa-tRNAs (Asn-tRNA, Gln-tRNA, Cys-tRNA and Sec-tRNA) are made by synthesizing the amino acid on the tRNA by first attaching a non-cognate amino acid to the tRNA, which is then converted to the cognate one catalyzed by tRNA-dependent modifying enzymes. Asn-tRNA or Gln-tRNA formation in most prokaryotes requires amidation of Asp-tRNA or Glu-tRNA by amidotransferases that couple an amidase or an asparaginase to liberate ammonia with a tRNA-dependent kinase. Both archaeal and eukaryotic Sec-tRNA biosynthesis and Cys-tRNA synthesis in methanogens require O-phosophoseryl-tRNA formation. For tRNA-dependent Cys biosynthesis, O-phosphoseryl-tRNA synthetase directly attaches the amino acid to the tRNA which is then converted to Cys by Sep-tRNA: Cys-tRNA synthase. In Sec-tRNA synthesis, O-phosphoseryl-tRNA kinase phosphorylates Ser-tRNA to form the intermediate which is then modified to Sec-tRNA by Sep-tRNA:Sec-tRNA synthase. Complex formation between enzymes in the same pathway may protect the fidelity of protein synthesis. How these tRNA-dependent amino acid biosynthetic routes are integrated into overall metabolism may explain why they are still retained in so many organisms. 相似文献

Neonatal development of the rat testis involves a number of critical events including re-entry of gonocytes into the cell cycle and eventual loss of many of these cells and their progeny via apoptosis. Since surviving gonocytes give rise to subsequent generations of germ cells, regulation of their fate is critical for adult testicular function. Here, we have identified a role for short-type PB-cadherin (STPB-C) in promoting survival of gonocytes in neonatal rats and we have linked its expression to the JAK-STAT signaling pathway. These findings were obtained with varied approaches including use of transgenic rats overexpressing STPB-C which were studied with protein microarrays and other techniques, direct examination of germ cell apoptosis and survival in gonocyte-Sertoli cell co-cultures, and direct study of the JAK-STAT pathway in these models and in L cells transfected with STPB-C. These data provide new information on the regulation of gonocyte fate and exciting new evidence supporting a link between the JAK-STAT pathway and cadherin-based cell-cell interactions. 相似文献

Background
Lysyl-tRNA synthetase (LysRS) is unique within the aminoacyl-tRNA synthetase family in that both class I (LysRS1) and class II (LysRS2) enzymes exist. LysRS1 enzymes are found in Archaebacteria and some eubacteria while all other organisms have LysRS2 enzymes. All sequenced strains of Bacillus cereus (except AH820) and Bacillus thuringiensis however encode both a class I and a class II LysRS. The lysK gene (encoding LysRS1) of B. cereus strain 14579 has an associated T box element, the first reported instance of potential T box control of LysRS expression. 相似文献58.
