An Exact theoretical expression for the apparent diffusion coefficient Dapp(K) of a thin rigid rod with arbitrary anisotropy of its translational diffusion diffusion coefficient is derived from the first cumulant of its dynamic structure factor. Dapp(K) is predicted to reach a limiting plateau value at extermely large values of KL, where K is the scattering vector and L the rod length. Howerver, that limiting plateau value is approached only very slowly along a quasi-plateau with a very gradual slope. Dynamic light-scattering studies have been performed on tobacco mosaic virus from K2 = (0.4–20) × 1010 cm?2 using 632-8-nm laser radiation. The present data yield D0 = (4.19 ± 0.10) × 10?8 cm2/s (corrected to 20,w conditions) and, with literature data to establish L = 2980 Å and the rotational diffusion coefficient DR = 318s?1, yield also Δ ≡ D ? D = (1.79 ± 0.38) × 10?8 cm2/s. The experimental data closely follow the curve of Dapp(K) vs K2 calcuated for these parameters. The present value of D0 substantially exceeds all previous dynamic light-scattering values, but is in good aggreement with previous sedimentation data, which were confirmed for the presemt sample. The anisotropy ratio Δ/D0 = 0.43 ± 0.09 is in accord with theoretical predictions based on the modified Kirkwood algorithm, despite the fact the D0 lies significantly below its corresponding theoretical value. The present data largely predlude the possibility that both D0 and Δ/D0 could simultaneously match their theoretical predictions. We present a detailed comparison of the experimental data with the calculations of Tirado and Garcia de la Torre based on the modified Kirkwood algorithm and with the Broersma formulas.  相似文献

Circular dichroism (CD) is being increasingly recognised as a valuable technique for examining the structure of proteins in solution. However, the value of many studies using CD is compromised either by inappropriate experimental design or by lack of attention to key aspects of instrument calibration or sample characterisation. In this article, we summarise the basis of the CD approach and its application to the study of proteins, and then present clear guidelines on how reliable data can be obtained and analysed.  相似文献

In small-scale experiments, the predatory mites, Hypoaspis aculeifer (Canestrini) and H. miles Berlese, applied at 700 mites m(-2), and the entomopathogenic nematode, Steinernema feltiae (Filipjev) applied at 3 x 10(-6) nematodes m(-2) controlled sciarids and phorids in mushroom compost and casing substrates. For both mite species, earliest application to the growing substrate following sciarid infestation reduced sciarid emergence. In contrast, later application of each biological control agent provided more effective control of phorid emergence. The behaviour of adult mites suggested that H. aculeifer were more positively geotactic than H. miles although both species could penetrate compost and casing substrates to a depth of 2-12 cm. A majority of S. feltiae nematodes resided at a depth of 2-4 cm in both substrate types. Independent application of H. aculeifer provided more comprehensive control of sciarids and phorids than the other biological agents studied, owing to its better dispersal within compost and casing, and ability to attack larvae of differing ages.  相似文献

Diploid cells of Saccharomyces cerevisiae were grown under controlled conditions with a Bioscreen instrument, which permitted the essentially continuous registration of their growth via optical density measurements. Some cultures were exposed to concentrations of a number of antifungal substances with different targets or modes of action (sterol biosynthesis, respiratory chain, amino acid synthesis, and the uncoupler). Culture supernatants were taken and analyzed for their "metabolic footprints" by using direct-injection mass spectrometry. Discriminant function analysis and hierarchical cluster analysis allowed these antifungal compounds to be distinguished and classified according to their modes of action. Genetic programming, a rule-evolving machine learning strategy, allowed respiratory inhibitors to be discriminated from others by using just two masses. Metabolic footprinting thus represents a rapid, convenient, and information-rich method for classifying the modes of action of antifungal substances.  相似文献

Clinical, epidemiological and mechanistic studies support the role of cranberry (Vaccinium macrocarpon Ait.) in maintaining urinary tract health. Cranberry proanthocyanidins contain A-type linkages and have been associated with preventing adhesion of P-fimbriated uropathogenic Escherichia coli to uroepithelial cells. It is not known if the presence of the A-type linkage is a prerequisite for anti-adhesion activity. Other commercial sources of proanthocyanidins with all B-type linkages have not previously been screened for this activity. The goals of this study were to compare the in vitro anti-adhesion activity of A-linked proanthocyanidins from cranberry juice cocktail with the anti-adhesion activities of B-linked proanthocyanidins from commercial grape and apple juices, green tea and dark chocolate, and determine if anti-adhesion activity is detectable in human urine following consumption of single servings of each commercial food product. Structural heterogeneity and presence of the A-type linkage in cranberry proanthocyanidins was confirmed utilizing MALDI-TOF/MS and DI/ESI MS, as was the presence of all B-type linkages in the proanthocyanidins from the other commercial products. The isolated A-type proanthocyanidins from cranberry juice cocktail elicited in vitro anti-adhesion activity at 60 microg/ml, the B-type proanthocyanidins from grape exhibited minor activity at 1200 microg/ml, while other B-type proanthocyanidins were not active. Anti-adhesion activity in human urine was detected following cranberry juice cocktail consumption, but not after consumption of the non-cranberry food products. Results suggest that presence of the A-type linkage in cranberry proanthocyanidins may enhance both in vitro and urinary bacterial anti-adhesion activities and aid in maintaining urinary tract health.  相似文献

Herbivory by copepods was studied from the coast towards theocean, during a bloom in May 1994 off NW Spain.Ingestion rateswere estimated by the gut chlorophyll content method in threesize fractions. The chlorophyll content displayed significantdaily cycles. Three different water bodies were described: coastal,shelf break and oceanic; the latter two zones separated by athermo haline frontal structure. Marked differences in planktonspecies composition, vertical distribution and biological rateswere found between zones. Theiiighest phytoplankton biomass,dominated by chain- forming diatoms, occurred in the oceaniczone associated with low primary production rates. Copepod feedinghad a low effect on oceanic phytoplankton; up to 0.2% of carbonstock and<3% of carbon production was consumed daily. Incontrast, medium-sized and large copepods removed 3% of carbonstock and 12% of primary production daily near the coast, wherephytoplankton were dominated by small flagellates in activegrowth. The highest variability in both plankton compositionand ingestionrates was found in the shelf-break zone, probablydue to displacements of the front. Copepods exerted a moderatepredation pressure on phytoplankton in coastal waters. Meanwhile,the impact of copepods on the offshore bloom was negligibleand the fate of the accumulated particulate carbon would bemostly determined by sedimentationand water dynamics.  相似文献

Human activities such as logging and agriculture can severely damage forest ecosystems by changing forest structure, ecosystem function, and biodiversity. These changes may have long‐lasting consequences, which influence forest recovery. We investigated the effect of past human disturbance on the current distribution of an understory, achlorophyllous orchid, Wullschlaegelia calcarata in Puerto Rico's tropical rain forest after 70 yr of recovery. Our study site was the 16‐ha Luquillo Forest Dynamics Plot located in the Luquillo Experimental Forest, which has four areas with differing intensity of land use that have been distinguished from variation in canopy cover seen in aerial photographs taken in 1936. We recorded orchids in six 10‐m‐wide, 500‐m‐long transects across four different areas of land‐use history. We found that the orchid was not present in an area of the plot which had <20 percent canopy cover in 1936, and was most abundant in the area with >80 percent canopy cover, which had been minimally impacted by human activity. Tree species composition varied among land‐use history areas, and our observations suggested that this variation might be influencing the local distribution of W. calcarata. We also measured leaf litter biomass and identified the leaves of litter in areas with and without the orchid. Litter with a high proportion of Buchenavia tetraphylla leaves had more orchids. Even though human disturbance ceased in 1932, land‐use history in the Luquillo Forest still casts a shadow over the distribution of W. calcarata.  相似文献

The rapid rise of antimicrobial resistance is one of the greatest challenges currently facing medical science. The most common cause of resistance to β-lactam antibiotics is the expression of β-lactamase enzymes, such as KPC-2. As such the development of novel inhibitors of KPC-2 and related enzymes is of the upmost importance. We report the design and synthesis of novel boronic acid transition state analogs containing a 1,4-substituted 1,2,3-triazole linker based on the known inhibitor 3-nitrophenyl boronic acid and demonstrate that they are promising scaffolds for the development inhibitors of KPC-2 with the ability to recover sensitivity to the antibiotic cefotaxime.  相似文献

Bounded by ocean and desert, the isolated, predominately Mediterranean‐climate region of south‐western Australia (SWA) includes nine bioregions (circa 44 million hectares). The ecological integrity of the landscapes in this global biodiversity hotspot has been compromised by deforestation, fragmentation, exploitation, and introduced biota. Nature and degree of transformation varies between four interconnected landscapes (Swan Coastal Plain; South‐west Forests; Wandoo Woodlands; and Great Western Woodlands). A Gondwanan perspective emphasizes a venerable biota and a cultural component to deep time. The particular importance of remnants and protected areas is recognized in restoring ecological integrity to Gondwanan landscapes. The nature and magnitude of the restoration task in these ancient, and neighboring, landscapes require higher levels of investment and more time than do recent landscapes. The protection, conservation, restoration, and rehabilitation of ecological integrity require multiple approaches in each landscape as well as consideration of the whole. Active conservation of biota and minimizing the impact of industrial‐ and agricultural‐use are priorities. Integrating a climate focus and rethinking fire are critical restoration considerations to future trajectories under anthropogenic climate change. A legislative mandate to coordinate industrial‐scale restoration and active conservation to build from protected areas must become a societal priority to restore ecological integrity.  相似文献
