The effects of a cold pretreatment, the concentration of different auxins (2,4-D, NAA and IAA) and the type of carbon source (maltose and sucrose) on the induction of callus from anthers of three parental lines and four rice F1 hybrids (Japonica × Indica, Indica × Japonica) were studied. The results indicated that a cold pretreatment was essential for the induction of callus from anthers of the parental lines and the F1 hybrids. These effects were genotype dependent. Auxins were essential for the induction of callus, and the type and concentration of auxins also influenced this process, as well as the type of carbon source. The greatest induction of callus was by the hybrid Morelos A92 × Koshihikari after a cold pretreatment of 8 days using 10.74 M –napthaleneacetic acid and 30 g l–1 maltose.  相似文献

The European Centre for the Validation of Alternative Methods (ECVAM) Skin Irritation Task Force was established in 1996, to review the status of the development and validation of alternative tests for skin irritation and corrosion, and to identify appropriate non-animal tests for predicting human skin irritation that were sufficiently well-developed to be prevalidated and validated by ECVAM. The EpiDerm method, based on a reconstituted human skin model, was proposed as being sufficiently well advanced to enter a prevalidation (PV) study. Based on a review of test protocols, prediction models (PMs), and data submitted by test developers on ten specified chemicals, with 20% sodium lauryl sulphate as a reference standard, the task force recommended the inclusion of four other tests: EPISKIN and PREDISKIN, based on reconstituted human epidermis or on human skin; the non-perfused pig-ear test, based on pig skin; and the skin integrity function test (SIFT), with ex vivo mouse skin. The prevalidation study on these methods was funded by ECVAM, and took place during 1999-2000. The outcome of the PV study was that none of the methods was ready to enter a formal validation study, and that the protocols and PMs of the methods had to be improved in order to increase their predictive abilities. Improved protocols and PMs for the EpiDerm and EPISKIN methods, the pig ear test, and the SIFT were presented at an extended Task Force meeting held in May 2001. It was agreed that, in the short term, the performance of the revised and harmonised EpiDerm and EPISKIN methods, as well as the modified SIFT, should be evaluated in a further study with a new set of 20 test chemicals. In addition, it was decided that the SIFT and the pig ear test would be compared to see if common endpoints (transepidermal water loss, methyl green-pyronine stain) could be identified.  相似文献

Terfezia and Tirmania, so called desert truffles, are mycorrhizal fungi mostly endemic to arid and semi-arid areas of the Mediterranean Region, where they are associated with Helianthemum species. The aim of this work was to study the phylogenetic relationships in these pezizalean hypogeous fungi. The restriction fragment length polymorphism (RFLP) and DNA sequences of internal transcribed spacers (ITS) of the nuclear rDNA were studied for several morphological species, Terfezia arenaria, T. boudieri, T. claveryi, T. leptoderma, T. terfezioides (=Mattirolomyces terfezioides), Tirmania nivea and T. pinoyi. The sequences were analyzed with distance and parsimony methods. Phylogenetic analyses indicated a close genetic relationship between Tirmania and Terfezia. They may have arisen from a single evolutionary lineage of pezizalean fungi that developed the hypogeous habit as an adaptation to heat and drought in Mediterranean ecosystems. This analysis also supports the re-establishment of the genus Mattirolomyces. The genera Tirmania and Terfezia were monophyletic, and morphological species corresponded to phylogenetic species. The Tirmania clade comprises desert truffles with smooth spores and amyloid asci, which were found in deserts. The Terfezia clade grouped species found in semi-arid habitats having ornamented and spherical spores. These species are adapted to exploit different types of soil (either acid or basic soils) in association with specific hosts (either basophilous or acidophilous species). Although other factors might also play a role, host specialization and edaphic tolerances (fungus and/or host tolerances) might be the key in the species diversity of these genera.  相似文献

Alpha-helix structure in Alzheimer's disease aggregates of tau-protein   总被引:5,自引:0,他引:5  
The discovery of beta-sheet structure in Alzheimer's amyloid fibrils, and then in many other disease-related protein fibrils, has led to the widely believed view that beta-sheet formation is the general mechanism of aberrant protein aggregation leading to disease. This notion is further reinforced by recent findings, which indicate that normal proteins can be induced to form beta-sheet fibrils in vitro. Alzheimer's disease, a paradigm proteopathy, is accompanied by the formation of two distinct aggregates, amyloid fibrils and paired helical filaments (PHFs). Electron microscope images of PHFs show pairs of twisted ribbons with 80 nm periodicity. However, there is little information of the molecular structure of PHFs, as previous studies have failed to identify signs of regular structure. Using far-UV circular dichroism and Fourier-transformed infrared spectroscopy, we find that PHFs are comprised of alpha-helices. This is remarkable as tau-protein, PHF's primary constituent, has a high abundance of helix-breaking amino acids and is unstructured in solution. We also find that PHFs are very stable, as judged by their high melting temperature and resistance to protease digestion. PHFs are the first example of pathological aggregation associated to the formation of alpha-helix.  相似文献

Since 1999 we have observed three cases of imported histoplasmosis in our hospital. One was an immunocompetent individual and two further patients were immunosuppressed (renal transplantation and HIV infection C3). Two patients were born in endemic areas (Equatorial Guinea and Ecuador) and a third patient had a history of previous travel to the Peruvian Amazonia. The different clinical presentations and diagnostic tools of histoplasmosis are discussed.  相似文献

This work analyses the behaviour of Poaceae pollen during a 3-year sampling period (1993–1995) in the city of Santiago de Compostela (Spain). This taxon is shown to be one of the most abundant in the atmosphere, with representation percentages greater than or equal to 20% of total pollen registered during the said period. Its main pollination period is centred on the months of June and July, with slight modifications in the beginning and finalization dates of the years 1993 and 1994 and a significantly earlier beginning in the year 1995. Overall results show that its presence may be considered significant from the end of May to the first days of August. The differences in Poaceae pollen representation are related to variations in temperature, precipitation and hours of sunshine.  相似文献

In a previous controlled study, 21 participants with essential hypertension were treated with a program based on education, relaxation and D'Zurilla problem-solving training, and another 21 participants were assigned to a waiting list control condition (García-Vera, Labrador, & Sanz, 1997). In this report, the pre-post-treatment psychological changes accompanying those conditions were examined with the Jenkins Activity Survey, the Rosenbaum Self-Control Schedule, the Spielberger State-Trait Anxiety Inventory, and the D'Zurilla-Nezu Social Problem-Solving Inventory. Treatment yielded significant psychological changes that included an increase of problem-solving abilities. Moreover, correlation and multiple regression analyses revealed that, when clinic blood pressure (BP) values were considered, increases in problem-solving abilities were correlated with systolic and diastolic BP reductions for participants in the stress-management condition, and they mediated partially the antihypertensive effects of stress-management training on BP. No significant correlations were found between psychological changes and self-measured systolic or diastolic BP reductions.  相似文献

Chromosome numbers of twenty taxa, almost all of them Iberian endemics, of the generaTeucrium, Sideritis, Thymus andSalvia are reported. New chromosome numbers are given for the following taxa:Teucrium homotrichum (2n=78),T. hifacense (2n=26),T. rivasii (2n=26),T. rivas-martinezii (2n=26),Sideritis edetana (2n=30),S. murgetana subsp.littoralis (2n=28), and the hybridS. xviciosoi (2n=27). The first count on Iberian material forSalvia pratensis (2n=18) is reported. Chromosome numbers have been confirmed for the rest of the taxa studied on material from SE Spain. The 22-oxocholestanes compounds have shown an outstanding plant growth promoting activity; they have similar bioactivity as brassinosteroids, so they are normally named as brassinosteroid analogs thinking that they also impact on the known receptor BRI1. However, in silico studies allow us to predict interactions with other receptors and thus it's possible to evaluate them, through receptors of gibberellins, auxins, jasmonates, strigolactones and the protein associated with the BRI1 gene. This article describes the bioactivity of structures SPGP4 and SPGP8 as plant growth-promoting compounds. Both structures present coupling energies and interactions at the same level as epibrassinolide in the protein associated with BRI1 gene. Additionally, interactions through the auxin pathway and to strigolactone receptor were found using selected tests. In the rice lamina tilt, a higher effect was obtained when SPGP4 and SPGP8 were compared to epibrassinolide, although in a lesser level vis à vis to homobrassinolide. In the same way, when SPGP4 and SPGP8 were tested in the Growth Root Model an activity as strigonolactones was observed, enhancing the relationship between the main and secondary roots. However, the growth of coleptiles, when applying auxins, compounds SPGP4 and SPGP8 did not reach the same level as controls. In the tests associated to gibberellins and jasmonic acid, an increased bioactivity was observed, although this behavior was not reflected from the in silico study, possibly due to secondary signaling cascades. This work demonstrates that the 22-oxocolestane compounds SPGP4 and SPGP8 could be used as plant growth hormones, promoting several pathways.  相似文献
