The aim of this study was to present data about ovary organization and oogenesis in two small groups of clitellate annelids, i.e. in representatives of Acanthobdellida (Acanthobdella peledina) and Branchiobdellida (Branchiobdella pentodonta and Branchiobdella parasitica), and to compare them to ovaries known from true leeches and oligochaetous clitellates. In A. peledina, the ovaries have the form of elongated cords, termed ovary cords, and are enveloped by coelomic sacs, the so-called ovisacs. The ovisacs are paired and each one contains only one ovary cord. The morphology and structure of the ovary cords depend on the maturity level of the animal. In young specimens the ovary cords are short and contain mainly oogonial cells and germ cells entering meiosis. Oogonia divide mitotically without full cytokineses, and as a result germ-line cysts are formed. As the animals grow, the cords become more elongated and the germ cells within the cords differentiate into nurse cells and oocytes. Oocytes gather cell organelles and, finally, detach from the ovary cord and float freely in the ovisac lumen.In both examined branchiobdellidans the ovaries are also paired. They are short and conical and are not enclosed within ovisacs. The narrow end of each ovary is connected to the intersegmental septum via a ligament, whereas the outermost (broad) end of the ovary extends freely into the coelom. The ovaries are polarized. Their narrow ends contain oogonia, whereas nurse cells and growing oocytes, gradually projecting from the ovary, can be found in their middle and outermost parts. Early vitellogenic oocytes detach from the ovary and float freely in the coelom.In all of the species studied, the ovaries are made up of germ-line cysts associated with somatic (follicular) cells. The architecture of a germ-line cyst is exactly the same as in other clitellate annelids that have been studied to date. Each germ cell in a cyst has one stable cytoplasmic bridge connecting it with a central anuclear cytoplasmic mass, a cytophore. The fate of germ cells constituting cysts is diverse. The majority of the cells withdraw from meiosis and become nurse cells; only a few continue meiosis, grow and become oocytes. The meroistic mode of oogenesis is suggested. We suggest also that the formation of germ-line cysts and ovary meroism should be regarded as basal conditions for all Clitellata. The occurrence of ovisacs enveloping the ovaries in A. peledina and Hirudinida is regarded as a synapomorphy of both groups, whereas ovaries found in B. pentodonta and B. parasitica have no ovisacs and resemble ovaries described in Oligochaeta sensu stricto.  相似文献

The colonization pattern of Vitis vinifera L. by Burkholderia phytofirmans strain PsJN was determined using grapevine fruiting cuttings with emphasis on putative inflorescence colonization under nonsterile conditions. Two-week-old rooted plants harbouring flower bud initials, grown in nonsterile soil, were inoculated with PsJN:gfp2x. Plant colonization was subsequently monitored at various times after inoculation with plate counts and epifluorescence and/or confocal microscopy. Strain PsJN was chronologically detected on the root surfaces, in the endorhiza, inside grape inflorescence stalks, not inside preflower buds and flowers but rather as an endophyte inside young berries. Data demonstrated low endophytic populations of strain PsJN in inflorescence organs, i.e. grape stalks and immature berries with inconsistency among plants for bacterial colonization of inflorescences. Nevertheless, endophytic colonization of inflorescences by strain PsJN was substantial for some plants. Microscopic analysis revealed PsJN as a thriving endophyte in inflorescence organs after the colonization process. Strain PsJN was visualized colonizing the root surface, entering the endorhiza and spreading to grape inflorescence stalks, pedicels and then to immature berries through xylem vessels. In parallel to these observations, a natural microbial communities was also detected on and inside plants, demonstrating the colonization of grapevine by strain PsJN in the presence of other microorganisms.  相似文献

Some properties of transitions generated during an imprecise replication of nucleic acids are described here. A possible role of these transitions in the evolution of the genetic code with respect to the evolution of the tertiary structure of proteins is suggested. The data supporting this hypothesis are obtained from the analysis of certain regularities present in the genetic code and in the proportions of amino acid residues buried in the interior of globular proteins.  相似文献

Proteins without readily available three-dimensional structural data present a difficult problem in the exploration of structure/function relationships. Saturation mutagenesis using contaminated oligonucleotides can identify potentially interesting regions of such a protein. This technique, in which synthesized oligonucleotides contain low-level base substitutions, allows random mutations to be placed throughout a gene sequence. Using double-stranded cassettes, a region of the human interleukin-1 alpha gene has been altered using such mutagenic oligonucleotides. However, instead of contaminating both strands of the gene sequence at the same level, each strand of the insert was contaminated at a different level. Several recombinants were sequenced and the effects of the mutations on the activity of the proteins were examined. Contaminating the two oligonucleotides at different levels produced a significantly different distribution of nucleotide changes from that seen if both strands were contaminated at the same level. The observed distribution followed the average of the distributions for each of the two contamination levels. This resulted in roughly equal frequencies of 1 to 5 nucleotide changes per clone with very few clones containing the wild-type nucleotide sequence. This helped overcome the redundancy in the genetic code, resulting in a high frequency of amino acid changes, and allowed changes at every amino acid to be sampled in a small number of mutants. This procedure can allow a gene sequence to be screened rapidly by removing most wild-type sequences from analysis while making sure that there are many amino acid changes in the resultant mutants.  相似文献

The relationship between alcohol consumption and glycoconjugate metabolism is complex and multidimensional. This review summarizes the advances in basic and clinical research on the molecular and cellular events involved in the metabolic effects of alcohol on glycoconjugates (glycoproteins, glycolipids, and proteoglycans). We summarize the action of ethanol, acetaldehyde, reactive oxygen species (ROS), nonoxidative metabolite of alcohol--fatty acid ethyl esters (FAEEs), and the ethanol-water competition mechanism, on glycoconjugate biosynthesis, modification, transport and secretion, as well as on elimination and catabolism processes. As the majority of changes in the cellular metabolism of glycoconjugates are generally ascribed to alterations in synthesis, transport, glycosylation and secretion, the degradation and elimination processes, of which the former occurs also in extracellular matrix, seem to be underappreciated. The pathomechanisms are additionally complicated by the fact that the effect of alcohol intoxication on the glycoconjugate metabolism depends not only on the duration of ethanol exposure, but also demonstrates dose- and regional-sensitivity. Further research is needed to bridge the gap in transdisciplinary research and enhance our understanding of alcohol- and glycoconjugate-related diseases.  相似文献

汞(Hg)是危害人类健康的主要重金属元素之一,多数食用菌对Hg有很强的富集能力,测定食用菌中Hg含量,并对其进行食用安全性评价具有重要意义。采用冷原子吸收直接测汞仪系统测定85份云南常见牛肝菌属真菌菌盖、菌柄中总Hg含量;以同一牛肝菌子实体菌盖与菌柄总Hg含量比(Q(C/S))分析牛肝菌属真菌对Hg的富集特征;根据联合国粮农组织和世界卫生组织(FAO/WHO)现行每周Hg允许摄入量(provisional tolerable weekly intake,PTWI)标准和中国GB 2762—2012规定的食用菌中Hg限量标准评价样品的食用安全性。结果表明,菌盖和菌柄中总Hg含量分别在0.13~22.00、0.20~8.40 mg·kg-1DW,不同产地、种类及不同采集年份的样品中总Hg含量存在明显差异;同一牛肝菌菌盖、菌柄总Hg含量比(Q(C/S))在0.28~4.08,92%的样品Q(C/S)1,表明多数样品中菌盖对Hg的富集能力强于菌柄。根据GB 2762—2012规定的食用菌及其制品中总Hg含量限量标准(≤0.1 mg·kg-1),发现所有测试样品的总Hg含量均超标;根据PTWI标准,假设成年人(60 kg)每周食用300 g新鲜牛肝菌,则多数样品Hg摄入量低于允许摄入量,无安全风险,而少数样品Hg摄入量高于允许摄入量,食用有一定的潜在风险。同时,牛肝菌总Hg含量与种类、产地、采集时间等因素密切相关,采食及安全评价需综合考虑这些因素。  相似文献

 Immunohistochemical studies have been performed to investigate the occurrence and coexistence of two catecholamine-synthesising enzymes, tyrosine hydroxylase and dopamine-β-hydroxylase, and several neuropeptides, including neuropeptide Y, vasoactive intestinal polypeptide, Leu5-enkephalin, somatostatin, calcitonin gene-related peptide and substance P, in nerve fibres supplying porcine accessory genital glands, the seminal vesicles, prostate (body and the disseminated part) and bulbourethral glands. Three major populations of nerve fibres supplying non-vascular elements of the glands have been distinguished (from the largest to the smallest one): (1) noradrenergic fibres, the majority of which contain Leu5-enkephalin, neuropeptide Y or, to a lesser extent, somatostatin, (2) non-noradrenergic, putative cholinergic fibres containing vasoactive intestinal polypeptide, neuropeptide Y and/or somatostatin and, (3) non-noradrenergic, presumably sensory fibres, containing calcitonin gene-related peptide and substance P. Whilst the coexistence patterns within nerves supplying particular glands are similar, the density of innervation varies between the organs. The innervation of the seminal vesicles and prostatic body is more developed than that of the disseminated part of the prostate and bulbourethral glands. The majority of noradrenergic fibres related to blood vessels contain neuropeptide Y only, while the non-noradrenergic nerves contain mainly vasoactive intestinal polypeptide. Summary Various cytotoxic functions of blood lymphocytes were studied in 101 patients undergoing daily treatment with human interferon- (IFN). Antibody-dependent cellular cytotoxicity and lectin-dependent cellular cytotoxicity were not altered to any major extent after 1 day or 1 week of IFN therapy. After 3 and 6 months of treatment a decrease in these functions was observed in most patients. Natural killer cell activity increased following the first injection of IFN and remained elevated during 1 year of IFN therapy.  相似文献
