We previously reported that dehydroevodiamine.HCl (DHED) has anticholinesterase and antiamnesic activities. To verify the effects of DHED on cognitive deficits further, we tested it on the scopolamine-induced amnesia model of the rat using the passive avoidance and eight-arm radial maze tests. A single (20 mg/kg p.o.) and repeated (10 mg/kg p.o.) administrations of DHED could significantly reverse the latency time shortened by scopolamine (1 mg/kg i.p.) to control level. The impaired spatial working memory induced by scopolamine (1 mg/kg i.p.) was also improved significantly by a single injection (6.25 mg/kg i.p.) and repeated administrations of DHED (10 mg/kg p.o.) in the eight-arm radial maze test. In addition, we examined the effects of DHED on the memory impairment and the histological changes of the brain after unilateral electrolytic lesion of the entorhinal cortex (EC) and middle cerebral artery occlusion in rats. The cognitive deficits caused by EC lesion and middle cerebral artery occlusion were improved significantly by repeated administrations of DHED (6.25 mg/kg i.p.) after EC lesion or ischemic insult once a day for 7 days in the passive avoidance test. Histological analysis showed that the neuronal loss in the DHED-treated group was notably reduced in the hippocampal area (CA1) of ischemic rats and in the dentate gyrus and hippocampal area (CA1 and CA3) of EC-lesioned rats compared with the nontreated group. The infarction area was decreased significantly by a single administration of DHED (6.25 mg/kg i.p.) 30 min before ischemic insult for 6 h. These results suggest that DHED might be an effective drug for not only the Alzheimer's disease type, but also the vascular type of dementia. 相似文献

Proteoglycans were extracted with 4 M guanidine–HCl from the zone of maturing chondrocytes, the site of endochondral ossification of growing antlers of wapiti (Cervus elaphus). Proteoglycans were isolated by DEAE-Sephacel chromatography and separated by Sepharose CL-4B chromatography into three fractions. Fraction I contained a high molecular mass (>1000 kDa) chondroitin sulfate proteoglycan capable of interacting with hyaluronic acid. Its amino acid composition resembled that of the cartilage proteoglycan, aggrecan. Fraction II contained proteoglycans with intermediate molecular weight which were recognized by monoclonal antibodies specific to chondroitin sulfate and keratan sulfate. Fraction III contained a low molecular mass (<160 kDa) proteoglycan, decorin, with a glucuronate-rich glycosaminoglycan chain. 相似文献

Hemolymph is oxidized and darkens visibly during the collection from silkworms due to the activity of tyrosinase in it. Toxic
quinones are produced by the oxidation and consequently inhibit the cell growth. Heat treatment can be used to prevent the
oxidation; however, the oxidation may occur during the collection of hemolymph before it is heat-treated. It makes the hemolymph
collection difficult especially on a large-scale preparation. Hemolymphs collected from 257 different strains of silkworms
were examined to select the slowly oxidized hemolymphs. Hemolymphs collected from mutant strains such as Lemone, TBO, Cre,
Y4, and wEb showed relatively slow color changes. Oxidation rates of the hemolymphs were measured by the absorbance change using a spectrophotometer.
The hemolymph of wEb showed the slowest oxidation. The absorbance of this mutant hemolymph reached the saturation value at 20°C in 450 min, whereas
the total oxidation time of the wild-type (Baekokjam) hemolymph at the same temperature was 120 min. We tested if this mutant
hemolymph is useful as a medium supplement for insect cell culture. Cell growth rate and final cell concentration in the medium
supplemented with the wEb hemolymph were almost same as those in the medium supplemented with the wild-type hemolymph. Hemolymph is collected on a
small scale by clipping the abdominal leg; however, this method is not appropriate for large scale preparation. Centrifugation
after chopping the silkworm hemolymph by a blending mixer is a more appropriate procedure for large scale collection. Slowly
oxidized wEb hemolymph resulted in higher cell concentration than the wild-type hemolymph when hemolymph was collected by the large scale

A new ion-pair high-performance liquid chromatographic method with column-switching has been developed for the determination of paraquat in human serum samples. The diluted serum sample was injected onto a precolumn packed with LiChroprep RP-8 (25-40 μm) and polar serum components were washed out by 3% acetonitrile in 0.05 M phosphate buffer (pH 2.0) containing 5 mM sodium octanesulfonate. After valve switching to inject position, concentrated compounds were eluted in the back-flush mode and separated on an Inertsil ODS-2 column with 17% acetonitrile in 0.05 M phosphate buffer (pH 2.0) containing 10 mM sodium octanesulfonate. The total analysis time per sample was about 30 min and mean recovery was 98.5±2.8% with a linear range of 0.1–100 μg/ml. To produce xylobiose from xylan, high-level expression of an endoxylanase gene from Bacillus sp. was carried out in Bacillus subtilis DB104. A 1.62-kb SmaI DNA fragment, coding for an endoxylanase of Bacillus sp., was ligated into the Escherichia coli/B. subtilis shuttle vector pJH27Δ88, producing pJHKJ4, which was subsequently transformed into B. subtilis DB104. A maximum endoxylanase activity of 105 U/ml was obtained from the supernatant of B. subtilis DB104 harboring pJHKJ4. The endoxylanase was purified to homogeneity by ion-exchange chromatography and the production profile
of xylooligosaccharides from xylan by the endoxylanase was examined by HPLC with a carbohydrate analysis column. Xylobiose
was the major product from xylan at 40 °C and its proportion in the xylan hydrolyzates increased with the reaction time; at
12 h, over 60% of the reaction products was xylobiose. These results suggest that xylobiose, which has a stimulatory effect
on the selective growth of the intestinal bacterium Bifidobacterium, can be mass-produced effectively by the endoxylanase of Bacillus sp. cloned in B. subtilis.

Fourteen species of fossil wood belonging to eleven genera and seven families were identified from 38 well preserved specimens collected from the Lower Coal-bearing Formation of Janggi Group at Donghae-myeon, Pohang City, Gyeongsangbuk-do Prefecture, Korea. Seven new taxa were found and described; they are Carya koreana Jeong et Kim, Betula janggiensis Jeong et Kim, Carpinus donghaensis Jeong et Kim, Ostrya geumgwangensis Jeong et Kim, Stewartia pseudo-camellioxylon Jeong et Kim, Acer minokamoensis Jeong, Kim et Suzuki and Acer pohangensis Jeong et Kim. The most abundant taxa are Betulaceae, Aceraceae, Ulmaceae, and Wataria. Except Wataria of which the habitat preference has not been confirmed, these taxa are major elements of cool-temperate vegetation and are similar to the Aniai-type flora of Japan. Previous studies of fossil plants from the Geumgwangdong Shale (leaves and seeds), the Upper Coal-bearing Formation (fossil woods) and the Yeonil Group (leaves and seeds) and this study, show that the climate of the Pohang Basin changed from cool-temperate to warm-temperate and subtropical during the Miocene. 相似文献

Highly efficient solar cells with sustainable performance under severe mechanical deformations are in great demand for future wearable power supply devices. In this regard, numerous studies have progressed to implement flexible architecture to high‐performance devices such as perovskite solar cells. However, the absence of suitable flexible and stretchable materials has been a great obstacle in the replacement of largely utilized transparent conducting oxides that are limited in flexibility. Here, a shape recoverable polymer, Noland Optical Adhesive 63, is utilized as a substrate of perovskite solar cell to enable complete shape recovery of the device upon sub‐millimeter bending radii. The employment of stretchable electrodes prevents mechanical damage of the perovskite layer. Before and after bending at a radius of 1 mm, power conversion efficiency (PCE) is measured to be 10.75% and 10.4%, respectively. Additionally, the shape recoverable device demonstrates a PCE of 6.07% after crumpling. The mechanical properties of all the layers are characterized by nanoindentation. Finite element analysis reveals that the outstanding flexibility of the perovskite layer enables small plastic strain distribution on the deformed device. These results clearly demonstrated that this device has great potential to be utilized in stretchable power supply applications. 相似文献

A small molecular metal‐chelate complex, tris(8‐hydroxyquinoline‐5‐sulfonic acid) aluminum (AlQSA3), that has three sulfonic acid groups per molecule leading to an excellent solubility in water is reported as a liquid‐free perfect solid‐state electrolyte for flexible film‐type all‐solid‐state energy storage devices. The AlQSA3 material is synthesized by one‐step reaction of aluminum triisopropoxide and 8‐hydroxyquinoline‐5‐sulfonic acid. The aqueous solutions of AlQSA3 are applied to fabricate flexible film‐type all‐solid state electric double layer capacitors with indium‐tin oxide thin film electrodes. The ion conductivity of the AlQSA3 film reaches 0.116 mS cm?1, while a pronounced hysteresis are obtained in the cyclic voltammetry measurement. The AlQSA3 film capacitors exhibit an output voltage of 1.5 V at 20 μA cm?2, which is considerably stable by the repeated operation. In particular, the peak output voltage is well kept even after 180° bending for 500 times in the case of the flexible AlQSA3 film capacitors. 相似文献
