Light dependent shift in the anti-predator response of a pyralid moth   总被引:1,自引:0,他引:1  
Male small china-mark moth Cataclysta lemnata (Pyralidae) swarming over shallow water show a flight activity that peaks during the afternoon but which sometimes is extended into the night. We exposed wild, naturally flying C. lemnata to simulated predator attacks consisting of a) bursts of ultrasound (26 kHz, simulating a bat) and b) a thrown stick (rapid movement, simulating a small bird), during day and night, respectively. We thus investigated the possibility that these moths are able to switch between defensive strategies as the predator regime shifts from insectivorous birds to bats in the evening. The defensive response differed qualitatively between day and night, as expected, but it was independent of the kind of stimulus. We thus demonstrate a previously unknown flexibility in the defensive strategy of moths.  相似文献

Flustra foliacea is a cheilostome marine bryozoan which, after initial horizontal growth during the first year, changes to erect growth thereafter. Appraising this slow-growing species for biotechnological use, required the development of cultivation methods for economical biomass production. Vegetative reproduction via cuttings, as employed in the horticulture of various plants, was tested. Fronds of F. foliacea were cut into pieces (10–150 mg fresh weight) and fixed in longitudinally cut silicon tubes. Bryozoa were fed with a mixture of Isochrysis galbana, Dunaliella tertiolecta and Cryptomonas sp. yielding growth rates of 0.3% ± 0.02 day?1of the initial fresh weight. Growth of the colony occurred firstly at the original growth margin, but later also at the transections, including the side fixed in the silicon tube, opposite to the growth margin. After three months of growth, the two layers of zooids split at the growth margin, forming two monolayers. This growth feature, not described previously for Flustra foliacea,was observed under experimental and natural conditions, and is interpreted as the beginning of one type of ramification. According to our findings, vegetative reproduction of F. foliacea under laboratory conditions is possible and might be an alternative to natural resources regarding biomass of this species.  相似文献

Confined within a volcanic caldera at 2000 m a.s.l., the sub-alpine desert of Tenerife, Canary Islands, harbors a distinct biota. At this altitude the climate is harsh and the growing season short. Hence, plant and animal communities, constituting the sub-alpine plant–flower-visitor network, are clearly delimited, both spatially and temporally. We investigated species composition and interaction structure of this system. A total of 11 plant species (91% endemics) and 37 flower-visiting animal species (62% endemics) formed 108 interactions. Numbers of interactions among species varied ten-fold within both plant and animal communities. Generalization level of a species was positively correlated with its local abundance. Two separate network analyses revealed a significantly nested structure. In relation to a plant–flower-visitor system, nestedness implies that specialized species (animals or plants) interact with a subset of the species pool visiting (animals) or being visited (plants) by more generalized species. Therefore, specialized, locally rare plants tend to be visited by generalized, locally abundant animals, and specialized, locally rare animals tend to utilize generalized, locally abundant food plants. Such patterns could have implications for conservation of the sub-alpine network, and stress the importance of preserving not only rare species, but also the more abundant ones, which may be key food resources or pollinators in the plant–flower-visitor network.  相似文献

Objective: The role of cortisol in the regulation of lipolysis is not clear. This study was undertaken to explore whether a standard dose of prednisolone for 1 week would influence lipolysis in abdominal and femoral tissue. Research Methods and Procedures: We used the microdialysis technique, the forearm technique, and indirect calorimetry, in the fasting state, after 1 week of treatment with prednisolone (30 mg daily) or placebo. Eight healthy young men (age: 25 ± 3 years; height: 181 ± 1 cm; body mass index [BMI]: 23.3 ± 0.7 kg/m2) were studied. Results: Treatment with prednisolone induced insulin resistance (Homeostasis Model Assessment index: placebo vs. prednisolone: 7.15 ± 1.63 vs. 17.00 ± 14.26, p = 0.03), hyperinsulinemia (p = 0.01), and hyperglucagonemia (p = 0.001), whereas growth hormone concentrations were unaffected. Abdominal adipose tissue interstitial glycerol was increased during treatment with prednisolone in the face of significant hyperinsulinemia, although it barely reached statistical significance (p = 0.06). At the femoral adipose tissue depot, no difference in lipolysis was found. Arterial and venous free fatty acids (FFA) were comparable in the two situations, whereas the arteriovenous difference across the forearm was significantly decreased during treatment with prednisolone, indicating increased uptake, or decreased release of FFA. Energy expenditure (p = 0.3), repiratory quotient (p = 0.9), glucose oxidation (p = 0.9), lipid oxidation (p = 1.0), and protein oxidation (p = 0.1) were unaltered on the 2 study days. Discussion: Short‐term treatment with a standard dose of corticosteroids induces increased abdominal adipose tissue lipolysis, as well as hyperinsulinemia, hyperglucagonemia, and insulin resistance.  相似文献

In order to meet increasingly stringentEuropean discharge standards, new applicationsand control strategies for the sustainableremoval of ammonia from wastewater have to beimplemented. In this paper we discuss anitrogen removal system based on the processesof partial nitrification and anoxic ammoniaoxidation (anammox). The anammox process offersgreat opportunities to remove ammonia in fullyautotrophic systems with biomass retention. Noorganic carbon is needed in such nitrogenremoval system, since ammonia is used aselectron donor for nitrite reduction. Thenitrite can be produced from ammonia inoxygen-limited biofilm systems or in continuousprocesses without biomass retention. Forsuccessful implementation of the combinedprocesses, accurate biosensors for measuringammonia and nitrite concentrations, insight inthe complex microbial communities involved, andnew control strategies have to be developed andevaluated.  相似文献

Hyaluronidases from diverse species and sources have different pH optima. Distinct mechanisms with regard to dynamic structural changes, which control hyaluronidase activity at varying pH, are unknown. Human serum hyaluronidase 1 (HYAL1) is active solely below pH 5.1. Here we report the design of a HYAL1 variant that degrades hyaluronan up to pH 5.9. Besides highly conserved residues in close proximity of the active site of most hyaluronidases, we identified a bulky loop formation located at the end of the substrate binding crevice of HYAL1 to be crucial for substrate hydrolysis. The stretch between cysteine residues 207 and 221, which normally contains 13 amino acids, could be replaced by a tetrapeptide sequence of alternating glycine serine residues, thereby yielding an active enzyme with an extended binding cleft. This variant exhibited hyaluronan degradation at elevated pH. This is indicative for appropriate substrate binding and proper positioning being decisively affected by sites far off from the active center.Hyaluronan (HA),3 a linear polysaccharide found in the extracellular matrix of most tissues and body fluids of vertebrates, is enzymatically degraded by hyaluronidases (1). Mammalian-type hyaluronidases are grouped into EC 3.2.1.35 (2, 3) or the glycoside hydrolase family 56 (4). Members of this enzyme family hydrolyze the 1,4-β-glycosidic linkage between N-acetyl-d-glucosamine and d-glucuronate within HA polymers (5). In mammalians, hyaluronidases have been found in testis, liver lysosomes, and serum. They are involved in controlling HA levels and are thus implicated in various diseases related to defects of HA metabolism (6).The crystal structures of hyaluronidase from bee (7), wasp (8), and only recently that of human serum hyaluronidase 1 (HYAL1) (9) have been deciphered. In addition to the N-terminal catalytic domain of the insect enzymes, which resembles a distorted (β/α)8 barrel, HYAL1 contains yet another domain. HA hydrolysis is achieved by a pair of acidic amino acids via a retaining double displacement mechanism and a substrate-assisted catalysis, in which the carbonyl oxygen of the N-acetyl group of the cleaved HA subunit acts as the catalytic nucleophile (7).Mammalian-type hyaluronidases display different pH optima. HYAL1 (10) and hyaluronidase 2 (HYAL2) (11) exhibit highest activities at acidic conditions, whereas the hyaluronidase found in Xenopus laevis kidney is only active at neutral pH (12). Bee venom hyaluronidase (13), as well as sperm hyaluronidase, PH20 (SPAM1) (14), are capable of degrading HA over a broad pH range. Up to three PH20 isoforms with greatly different pH optima could be found in protein preparations from bovine testis (15). Extensive analysis of hyaluronidase structures did not bring forward any insights as to what residues or regions of the enzymes specify a specific pH optimum.Profiles of pH-dependent activities can be assigned by computing the electrostatic interactions of the enzyme, which are primarily determined by the ionization states of its amino acid side chains. The pKa values of titratable groups of the enzyme reflect pH-dependent properties such as stability, enzymatic interaction, and substrate interactions (16). Here we present computational and experimental data on the replacement of a loop region located at the end of the substrate binding groove yielding a variant hyaluronidase with an altered pH profile.  相似文献

Scale-up from shake flasks to fermenters has been hampered by the lack of knowledge concerning the influence of operating conditions on mass transfer, hydromechanics, and power input. However, in recent years the properties of shake flasks have been described with empirical models. A practical scale-up strategy for everyday use is introduced for the scale-up of aerobic cultures from shake flasks to fermenters in batch and continuous mode. The strategy is based on empirical correlations of the volumetric mass transfer coefficient (k(L) a) and the pH. The accuracy of the empirical k(L) a correlations and the assumptions required to use these correlations for an arbitrary biological medium are discussed. To determine the optimal pH of the culture medium a simple laboratory method based on titration curves of the medium and a mechanistic pH model, which is solely based on the medium composition, is applied. The effectiveness of the scale-up strategy is demonstrated by comparing the behavior of Corynebacterium glutamicum on lactic acid in shake flasks and fermenters in batch and continuous mode. The maximum growth rate (micro(max) = 0.32 h(-1)) and the oxygen substrate coefficient (Y O2 /S= 0.0174 mol/l) of C. glutamicum on lactic acid were equal for shake flask, fermenter, batch, and continuous cultures. The biomass substrate yield was independent of the scale, but was lower in batch cultures (Y(X/S) = 0.36 g/g) than in continuous cultures (Y(X/S) = 0.45 g/g). The experimental data (biomass, respiration, pH) could be described with a simple biological model combined with a mechanistic pH model.  相似文献
