Recent evidence suggests that ceramide regulates stress signaling via reorganization of the plasma membrane. The focus of this review will be to discuss the mechanism by which acid sphingomyelinase (ASMase)-generated ceramide initiates transmembrane signaling in the plasma membrane exoplasmic leaflet. In particular, we review the unique biophysical properties of ceramide that render it proficient in formation of signaling domains termed ceramide-rich platforms (CRPs), and the role of CRPs in the pathophysiology of various diseases. The biomedical significance of CRPs makes these structures an attractive therapeutic target.  相似文献

Ramonda nathaliae (Gesneriaceae) is a rare desiccation tolerant flowering plant species of the Northern Hemisphere. This, mainly calcicole, preglacial relict species is endemic in the Balkan Peninsula, where it has survived in its refugial habitats of gorges and mountain slopes. At present, only two localities within its narrow range are known where it thrives in hostile serpentine habitats, and the adverse serpentine environment is bound to present further challenge to the adaptive capacity of R. nathaliae. In general, the occurrence of a resurrection flowering plant on serpentine soil is exceptional and the soil-plant relation of R. nathaliae in those circumstances is described here for the first time. The aim of this study was (i) to analyze mineral elements composition in soil from both serpentine and limestone habitats of the species and to compare the way peculiarities of the substrates are reflected in roots and leaves of plants from the respective soils; (ii) to evaluate the effect of heavy metal overload on the habit of serpentine R. nathaliae individuals. Serpentine soil, characterized by high levels of phytotoxic heavy metals (Ni, Cr, Co, Mn), hosts plants showing elevated metal contents in their organs. Ramonda plants from serpentine populations are able to maintain balance of Ca to Mg favourable to Ca (2.0 in roots, 2.7 in leaves) despite a strongly unfavourable Ca/Mg ratio in the soil (0.09). The greatest difference in concentrations was observed for the Ni content in plant tissues: serpentine plants had 57 and 20 times more Ni in their roots and leaves than the plants from limestone. Aluminium was present in similar concentrations in both soils, and was highly accumulated in plant tissues of the plants from both substrates. Metal-specific metabolic activity is demonstrated in bioaccumulation indices of several essential minerals (Ca, Mg, Cu, Zn). A significantly higher metal content found in roots in relation to leaves might indicate the plant's ability to immobilize the metals within the root tissues. Mycorrhizal fungi colonize plant roots from both substrates and apparently are important in improving the supply of nutrients, but they could also take part in toxic metal immobilization. The price of adaptation to the hostile environment is evident in the habit of R. nathaliae plants growing on serpentine: reduced size of rosettes and leaves, as well as chlorotic and necrotic leaf tips and margins.  相似文献

The aim was to investigate the detection rates of periodontal bacteria (Porphyromonas gingivalis, Tannerella forsythia, Prevotella intermedia, and Aggregatibacter actinomycetemcomitans) and herpesviruses (herpes simplex virus-1 [HSV-1], cytomegalovirus [CMV], and Epstein-Barr virus [EBV]) in different forms and severity of periodontal disease, and to compare them with those in periodontally healthy subjects. One hundred and twenty-nine patients participated in the study: 39 diagnosed with periodontal abscess (PA), 33 with necrotizing ulcerative periodontitis (NUP), 27 with chronic periodontitis (CP), and 30 participants with healthy periodontal tissue represented a healthy control group. All patients with periodontal disease (PA, NUP, and CP) were also divided into two groups according to the severity of their disease: moderate and severe periodontitis. The subgingival samples were collected from the periodontitis active sites and the detection of microorganisms was performed by end-point polymerase chain reaction analyses. The results revealed significantly higher detection rates of P. gingivalis, T. forsythia, and P. intermedia in all three groups of patients with periodontitis than in healthy participants. The highest detection rate of A. actinomycetemcomitans was noticed in CP, which was significantly higher than that in PA, NUP, and healthy control. The occurrence of EBV was significantly higher in NUP than in CP and healthy participants. CMV was detected significantly more frequently in PA and NUP than in CP and healthy participants. Comparisons among healthy participants and patients with moderate and severe periodontitis showed significantly higher detection rates of EBV and CMV in patients with severe forms of periodontitis than in healthy participants and those with moderate periodontitis.  相似文献

The TGF-betas are multifunctional proteins whose activities are believed to be controlled by interaction with the latent TGF-beta binding proteins (LTBPs). In spite of substantial effort, the precise in vivo significance of this interaction remains unknown. To examine the role of the Ltbp-3, we made an Ltbp-3-null mutation in the mouse by gene targeting. Homozygous mutant animals develop cranio-facial malformations by day 10. At 2 mo, there is a pronounced rounding of the cranial vault, extension of the mandible beyond the maxilla, and kyphosis. Histological examination of the skulls from null animals revealed ossification of the synchondroses within 2 wk of birth, in contrast to the wild-type synchondroses, which never ossify. Between 6 and 9 mo of age, mutant animals also develop osteosclerosis and osteoarthritis. The pathological changes of the Ltbp-3-null mice are consistent with perturbed TGF-beta signaling in the skull and long bones. These observations give support to the notion that LTBP-3 is important for the control of TGF-beta action. Moreover, the results provide the first in vivo indication for a role of LTBP in modulating TGF-beta bioavailability.  相似文献

The growing demand to fulfill the needs of present-day medicine in terms of novel effective molecules has lead to reexamining some of the old and known bacterial secondary metabolites. Bacterial prodigiosins (prodiginines) have a long history of being re markable multipurpose compounds, best examined for their anticancer and antimalarial activities. Production of prodigiosin in the most common producer strain Serratia marcescens has been described in great detail. However, few reports have discussed the ecophysiological roles of these molecules in the producing strains, as well as their antibiotic and UV-protective properties. This review describes recent advances in the production process, biosynthesis, properties, and applications of bacterial prodigiosins. Special emphasis is put on undecylprodigiosin which has generally been a less studied member of the prodigiosin family. In addition, it has been suggested that proteins involved in undecylprodigiosin synthesis, RedG and RedH, could be a useful addition to the biocatalytic toolbox being able to mediate regio- and stereoselective oxidative cyclization. Judging by the number of recent references (216 for the 2007–2013 period), it has become clear that undecylprodigiosin and other bacterial prodigiosins still hold surprises in terms of valuable properties and applicative potential to medical and other industrial fields and that they still deserve continuing research curiosity.  相似文献

The mechanism of resistance to aminoglycosides based on methylation of their target, 16S rRNA, was until recently described only in antibiotic producing microorganisms. However, equivalent methyltransferases have now also been identified among numerous clinical Gram-negative pathogenic isolates. We have cloned, expressed, and purified GrmA, the aminoglycoside-resistance methyltransferase from Micromonospora purpurea, producer of gentamicin complex. Two vectors were created that express protein with an N-terminal 6× histidine tag with and without an enterokinase recognition producing proteins His6-EK-GrmA and His6-GrmA, respectively. The activity of both recombinant proteins was demonstrated in vivo. After optimized expression and native purification both protein variants proved to be active in in vitro methylation assays. This work lays a foundation for future detailed biochemical, structural and pharmacological studies with this member of an important group of aminoglycoside-resistance enzymes.  相似文献
