The productivity of stable Chinese hamster ovary cell lines secreting HIV-1 monomeric (IIIB gp120) and oligomeric (UG21 gp140) recombinant envelope glycoproteins was compared in serum-containing (S+), serum-free (S−) and protein-free (P−) culture media. UG21 gp140 expression was greatest in S+ medium, while IIIBgp120 production was lower than gp140 in all three media but highest in S−. UG21 gp140 production was highest in standard 850-cm2 roller bottle cultures in S+ media, peaking after 14 days of incubation, while expression levels in the three media were 0.5 (S+), 0.4 (S−) and 0.2 (P−) mg/l, from which 90, 80 and 12% of gp140, respectively, could be purified by immunoaffinity chromatography. Purified UG21 gp140 from S+ and S− media possessed biological functionality as evidenced by CD4 and monoclonal antibody (Mab) binding. In contrast, UG21 gp140 from P− medium appears to be misfolded and non-functional. Despite the possession of a different N-linked glycan profile, UG21 gp140 from S− media shows very similar CD4 and Mab binding characteristics to S+ UG21 gp140. The relevance of these findings to HIV vaccine development is discussed. 相似文献

Juvenile and adult reef fishes often undergo migration, ontogenic habitat shifts, and nocturnal foraging movements. The orientation
cues used for these behaviours are largely unknown. In this study, the use of sound as an orientation cue guiding the nocturnal
movements of adult and juvenile reef fishes at Lizard Island, Great Barrier Reef was examined. The first experiment compared
the movements of fishes to small patch reefs where reef noise was broadcast, with those to silent reefs. No significant responses
were found in the 79 adults that were collected, but the 166 juveniles collected showed an increased diversity each morning
on the reefs with broadcast noise, and significantly greater numbers of juveniles from three taxa (Apogonidae, Gobiidae and
Pinguipedidae) were collected from reefs with broadcast noise. The second experiment compared the movement of adult and juvenile
fishes to reefs broadcasting high (>570 Hz), or low (<570 Hz) frequency reef noise, or to silent reefs. Of the 122 adults
collected, the highest diversity was seen at the low frequency reefs; and adults from two families (Gobiidae and Blenniidae)
preferred these reefs. A similar trend was observed in the 372 juveniles collected, with higher diversity at the reefs with
low frequency noises. This preference was seen in the juvenile apogonids; however, juvenile gobiids were attracted to both
high and low sound treatments equally, and juvenile stage Acanthuridae preferred the high frequency noises. This evidence
that juvenile and adult reef fishes orientate with respect to the soundscape raises important issues for management, conservation

Recombinant expression systems differ in the type of glycosylation they impart on expressed antigens such as the human immunodeficiency virus type 1 (HIV-1) envelope glycoproteins, potentially affecting their biological properties. We performed head-to-head antigenic, immunogenic and molecular profiling of two distantly related Env surface (gp120) antigens produced in different systems: (a) mammalian (293 FreeStyle™ cells; 293F) cells in the presence of kifunensine, which impart only high-mannose glycans; (b) insect cells (Spodoptera frugiperda, Sf9), which confer mainly paucimannosidic glycans; (c) Sf9 cells recombinant for mammalian glycosylation enzymes (Sf9 Mimic™), which impart high-mannose, hybrid and complex glycans without sialic acid; and (d) 293F cells, which impart high-mannose, hybrid and complex glycans with sialic acid. Molecular models revealed a significant difference in gp120 glycan coverage between the Sf9-derived and wild-type mammalian-cell-derived material that is predicted to affect ligand binding sites proximal to glycans. Modeling of solvent-exposed surface electrostatic potentials showed that sialic acid imparts a significant negative surface charge that may influence gp120 antigenicity and immunogenicity. Gp120 expressed in systems that do not incorporate sialic acid displayed increased ligand binding to the CD4 binding and CD4-induced sites compared to those expressed in the system that do, and imparted other more subtle differences in antigenicity in a gp120 subtype-specific manner. Non-sialic-acid-containing gp120 was significantly more immunogenic than the sialylated version when administered in two different adjuvants, and induced higher titers of antibodies competing for CD4 binding site ligand-gp120 interaction. These findings suggest that non-sialic-acid-imparting systems yield gp120 immunogens with modified antigenic and immunogenic properties, considerations that should be considered when selecting expression systems for glycosylated antigens to be used for structure-function studies and for vaccine use. 相似文献

By the implantation of wedges containing indol-3-ylacetic acid and sucrose into blocks of undifferentiated bean-callus tissue it has been possible to induce the formation of xylem and phloem cells. 2. The differentiation has been investigated cytologically and measured chemically. 3. The optimum concentrations of the nutrients in the wedge, which gave differentiation closely resembling the vascular development found in the stem of the intact plant, was 0.1mg. of indol-3-ylacetic acid/l. and 2% sucrose. 4. The ratios of the xylose/arabinose concentrations of the tissues increased in the differentiated callus tissue compared with those of the undifferentiated tissue. A similar increase has been found for the ratios determined for xylem tissue compared with those for cambium. 5. The lignin content of the differentiated tissue compared with the undifferentiated tissue was greater in both the callus and stem tissue. 6. Chemical analysis of lignin showed that in the differentiated callus tissue it consisted of sub-units based on p-hydroxybenzaldehyde and vanillin. This was compared with the lignin obtained from undifferentiated callus tissue and that obtained from the tissues of the intact stem. 7. The results of the investigation have been discussed with reference to the problems of cell growth and differentiation and related to the changing patterns of the ultrastructure of the cell during its development. 相似文献

A new photoactive beta-adrenergic antagonist, p-azidobenzylcarazolol (pABC) has been synthesized by combining a carbazole moiety with a p-azido-benzyl substituent. The compound has been labeled with tritium to a specific activity of 26 Ci/mmol. In frog erythrocyte membranes, [3H]p-azido-benzylcarazolol binds to the beta-adrenergic receptor with the expected beta 2 specificity and with high affinity (KD congruent to 100 +/- 10 pM). Unlabeled p-azido-benzylcarazolol can irreversibly inactivate the [3H]dihydroalprenolol-binding activity of frog erythrocyte membranes in a photodependent manner which can be prevented by beta-adrenergic agents. Incubation of frog erythrocyte membranes or digitonin-solubilized preparations of these membranes or digitonin-solubilized preparations of these membranes which had been enriched in beta-adrenergic receptors by a Sepharose-alprenolol chromatography step led to covalent incorporation of radioactivity into a Mr = 58,000 peptide. Specific incorporation of [3H]pABC into the Mr = 58,000 peptide could be prevented by both beta-adrenergic agonists and antagonists. This peptide has previously been purified and shown to contain the beta-adrenergic receptor-binding site (Shorr, R. G. L., Lefkowitz, R. J., and Caron, M. G. (1981) J. Biol. Chem. 256, 5820-5826). Thus, photoaffinity labeling of the beta-adrenergic receptor protein directly identifies the same hormone-binding subunit as has been isolated by conventional purification techniques. 相似文献

Cycloleucine uptake by metacestodes of H. diminuta of various ages was investigated. Absorption occurs by active mediated transport, mean Kt = 0.28 mM. Vmax values are age-related, and can be correlated to developmental changes.
