Summary The solution conformation of human big endothelin-1, a 38-residue peptide which serves as the putative precursor to the potent vasoconstrictor endothelin-1 has been examined by1H NMR. NOEs were utilized as distance restraints in the distance geometry program DSPACE to generate initial structures. Further refinement of these structures was accomplished through molecular mechanics/molecular dynamics in an iterative process involving the incorporation of stereospecific assignments of prochiral centers and the use of back-calculation of NOESY spectra. A family of structures consisting of a type 11 -turn for residues 5–8 and an -helix extending from residues 9–16 constitute a well-defined region, as reflected by the atomic root-mean-square (RMS) difference of 1.56 Å about the mean coordinate positions of the backbone atoms (N, C, Ca and O). This core region (residues 1-15) is very similar to the core residues of endothelin-1 (Donlan, M. et al. (1991)J. Cell. Biochemistry, S15G, 85). While the evidence from NOESY and coupling constant data suggests that the C-terminal region, residues 17–34, is not a mixture of randomly distributed chain conformations, it is also not consistent with a single chain conformation. Under the conditions studied, residues 17–38 in human big endothelin-1 in water at pH 3.0 between 20–30°C appear to be represented by a series of conformers in dynamic equilibrium.  相似文献

The success of restoration initiatives to restore bivalve beds relies on sufficient recruitment of larvae to offset mortality of re‐established populations. Individuals of the nearly extirpated green‐lipped mussel are capable of surviving within the current environment of the Hauraki Gulf, New Zealand; however, it is uncertain what potential factors might inhibit the establishment and persistence of restored mussel beds. Four experimental mussel beds were established within a shallow soft‐sediment embayment and assessments of population dynamics were conducted approximately every 6 months over a 2‐year period. Deployed mussels quickly congregated into contiguous mussel beds that persisted throughout the study; however, only 26.2% of mussels that were initially established survived until the end of the study. The cause of this overall loss of mussels can be attributed to a near lack of observed recruitment, with only three individual recruiting mussels observed throughout the entire study. Despite similar mortality rates within the restored mussel beds to that of natural populations, these populations will be unsustainable long term given the lack of recruitment. Potential causes of the observed mortality and lack of recruitment are discussed, including environmental factors affecting non‐natal mussel stock and sea star predation. This research provides a foundation for the development of best‐practice methods in the restoration of green‐lipped mussels. However, further investigation into recruitment pathways and sources of mortality for adult mussels will be necessary to overcome the observed limitations if future restoration is to be successful.  相似文献

The crude 5-year survival rate among children with Wilms'' tumour increased from 54% for those diagnosed from 1960 to 1965 to 81% for the period 1966 to 1971. This resulted from an increased ability to cure metastatic disease and, to a lesser extent, to an increased ability to prevent relapse. It is proposed that, after resection and postoperative irradiation, maintained combination chemotherapy with actinomycin D and vincristine should be used electively to prevent relapse, but that this use should also be selective in order that overall morbidity be minimized. Of urgent priority, therefore, is improved delineation of present-day prognostic factors in children with Wilms'' tumour.  相似文献

Underwater noise is increasing globally, largely due to increased vessel numbers and international ocean trade. Vessels are also a major vector for translocation of non-indigenous marine species which can have serious implications for biosecurity. The possibility that underwater noise from fishing vessels may promote settlement of biofouling on hulls was investigated for the ascidian Ciona intestinalis. Spatial differences in biofouling appear to be correlated with spatial differences in the intensity and frequency of the noise emitted by the vessel’s generator. This correlation was confirmed in laboratory experiments where C. intestinalis larvae showed significantly faster settlement and metamorphosis when exposed to the underwater noise produced by the vessel generator. Larval survival rates were also significantly higher in treatments exposed to vessel generator noise. Enhanced settlement attributable to vessel generator noise may indicate that vessels not only provide a suitable fouling substratum, but vessels running generators may be attracting larvae and enhancing their survival and growth.  相似文献

Both 3,4-dihydroxyphenylalanine and 2,4,5-trihydroxyphenylalanine were oxidized with periodate and mushroom tyrosinase to determine whether the latter compound is an intermediate in melanin biosynthesis. Matrix analysis of the spectra obtained with a rapid scan spectrophotometer and comparison of the spectra of quinone intermediates with model quinones disclosed that, although 2,4,5-trihydroxyphenylalanine can be oxidized to 2-carboxy-2,3-dihydroindole-5,6-quinone (dopachrome), this oxidation proceeds through a stable intermediate, 5-(2-carboxy-2-aminoethyl)-2-hydroxy-1,4-benzoquinone, which does not appear in the oxidation of 3,4-dihydroxyphenylalanine to dopachrome. Thus, these studies are in agreement with the original postulate, that 4-(2-carboxy-2-aminoethyl)-1,2-benzoquinone and leukodopachrome are the intermediates in the major pathway for dopachrome synthesis.  相似文献

The feeding of the late larval stages of the spiny lobster, Panulirus cygnus, and the energy reserves used by the non-feeding post-larvae for crossing the continental shelf of Western Australia were examined through their lipid composition. Lipid was a significant component of the biomass of all larval and post-larval samples (range 63-213 mg g(-1) of dry biomass). The fatty acid profiles of late stage larvae (8-9) suggest that they were feeding on salps and small crustaceans, such as euphausids, from oligotrophic pelagic communities where herbivorous and microbial grazing is an important basis to the food web. There was a marked decrease in lipid content of post-larvae progressively across the continental shelf, and this corresponded closely with decreasing dry mass, suggesting that post-larvae were using lipid as an energy source during cross-shelf movement at a rate of 1.6 J km(-1). This is considerably lower than for other spiny lobster species, suggesting that the post-larvae of the western rock lobster may use physical processes as well as active swimming for onshore transport. This may help to explain the large inter-annual variability in the post-larval settlement of this species, which is closely related to differences in weather patterns capable of greatly varying onshore advection processes.  相似文献

Predation by sea stars has the potential to cause elevated levels of mortality in reestablished populations of bivalves relative to levels of recruitment. Recent efforts to restore beds of the nearly extirpated green‐lipped mussel within the Hauraki Gulf, New Zealand, resulted in high abundances of sea stars occupying those beds and it is unknown whether predation poses a potential limitation to the success of restoration in this bivalve species. The contribution of predation by sea stars to the mortality of mussels across four experimental mussel beds over a 2‐year period was estimated using data from regular assessments of sea star abundance and an experimentally determined consumption rate of sea stars upon mussels. In addition, the potential size selection of mussels by sea stars was assessed to determine if large sea stars selected for recent settlers. Sea stars' abundance within the mussel beds grew to an average of 0.57 sea stars/m2 within 9 months, remaining at similar levels throughout the study. These predominantly large sea stars were estimated to have consumed 30.1% of the mussels over a 25‐month period, representing a contribution of 40.4% of the mussel mortality. Sea stars predominantly selected for larger mussels, and their predation likely contributes little to any lack in mussel recruitment. Sea star predation is clearly a limiting factor on the survival of transplanted adult mussels and the present study highlights the need to continually assess predation risk to determine if remediation is necessary for the persistence of the restored beds.  相似文献

A confirmed Ebola haemorrhagic fever (EHF) outbreak in Bundibugyo, Uganda, November 2007–February 2008, was caused by a putative new species (Bundibugyo ebolavirus). It included 93 putative cases, 56 laboratory-confirmed cases, and 37 deaths (CFR = 25%). Study objectives are to describe clinical manifestations and case management for 26 hospitalised laboratory-confirmed EHF patients. Clinical findings are congruous with previously reported EHF infections. The most frequently experienced symptoms were non-bloody diarrhoea (81%), severe headache (81%), and asthenia (77%). Seven patients reported or were observed with haemorrhagic symptoms, six of whom died. Ebola care remains difficult due to the resource-poor setting of outbreaks and the infection-control procedures required. However, quality data collection is essential to evaluate case definitions and therapeutic interventions, and needs improvement in future epidemics. Organizations usually involved in EHF case management have a particular responsibility in this respect.  相似文献

The mechanisms by which reef fish larvae locate settlement habitat at the end of their pelagic phase are unclear. We used an in situ binary choice chamber and an artificial source of reef sound to determine whether pomacentrid larvae can use ambient sound to locate reefs. Larvae were caught in light traps and then placed in a submerged binary choice chamber with an artificial source of reef sound ~80 m from one end of the chamber. At night, larvae moved towards the sound source; during the day, larvae showed no preference. These results suggest that pomacentrid larvae can detect reef sound and are capable of directional hearing. While other studies have shown that reef fish larvae respond to reef sound, and that the adults of some species can localize underwater sound sources, the localization of underwater sound by fish larvae has not been demonstrated previously.Communicated by Ecological Editor P.F. Altered temperatures affect insects’ life history traits, such as development period and fecundity, which ultimately determine population growth rates. Understanding insects’ thermal biology is therefore integral to population forecasting and pest management decision‐making such as when to utilise crop spraying or biological control. Aphids are important crop pests in temperate regions, causing considerable yield losses. The aphid thermal‐biology literature is, however, heavily biased towards the effects of rising mean temperatures, whereas the effects of fluctuating, extreme climatic events (e.g., heat waves and sub‐zero cold periods) are largely overlooked. This study assessed the effects of laboratory‐simulated heat waves and sub‐zero cold periods on the survival, development period, and fecundity of the grain aphid, Sitobion avenae (Fabricius) (Hemiptera: Aphididae: Microsiphini), in addition to assessing maternal effects on the birth weight and development period of the offspring of exposed individuals. Exposure to heat stress periods (total of 16 h at 30 °C) significantly reduced aphid fecundity and increased physiological development period (in day‐degrees) resulting in a reduced population growth rate. Cold exposure (total of 1.33 h at ?15 °C) reduced population growth rate due to an elongated development period (in days), but did not affect fecundity or physiological development period (in day‐degrees). Both cold and heat stress significantly reduced aphid survival. Maternal experience of heat stress reduced nymphal birth weight although nymphal development period was not affected by either cold or heat stress. The results suggest that including the effects of fluctuating, extreme temperature events on aphid life history in population forecast models is likely to be of great importance to pest management decision‐making. The demonstration of maternal effects on birth weight also suggests that cross‐generational effects of heat waves on population growth rates could occur.  相似文献
