Keep all the pieces: Systematics 2000 and world conservation

Conserving biological diversity requires a major effort in conducting survey and inventories, establishing priorities, selecting protected areas, managing resources and monitoring the effects of management. Systematics has an important contribution to make to each of these five major activities. Further, the new Convention on Biological Diversity requires systematics information to support action under virtually all of its substantive conservation and sustainable use articles. It seems apparent that large reference collections contribute directly to development, and development assistance agencies should recognize that investing in maintaining these collections is a legitimate form of development assistance.     

Inhibition of surfactant release from isolated type II cells by compound 4880

Release of [³H]phosphatidylcholine from pulmonary Type II epithelial cells was stimulated by terbutaline, forskolin and cytochalasin D. Compound $\text{48}\text{80}$ inhibited both basal and agonist-stimulated release of [³H]PC. The IC₅₀ for inhibition by compound $\text{48}\text{80}$ was 1–2 μg/ml, and was similar for inhibition of both basal and stimulated release of [³H]phosphatidylcholine. Inhibitory effects of $\text{48}\text{80}$ were noted following a 1 h exposure to compound $\text{48}\text{80}$ and persisted up to 3 h. The inhibitory effect of compound $\text{48}\text{80}$ was entirely reversed by removing compound $\text{48}\text{80}$ from the external milieu. Compound $\text{48}\text{80}$ had no effect on cytosolic cyclic AMP levels or lactate dehydrogenase release. Inhibition of surfactant release produced by compound $\text{48}\text{80}$ was unaffected by changes in extracellular calcium concentrations. Compound $\text{48}\text{80}$ is a non-toxic inhibitor of phosphatidylcholine release from Type II epithelial cells.     

Measurement of the inorganic pyrophosphate in tissues of Pisum sativum L.

Purified pyrophosphate: fructose 6-phosphate 1-phosphotransferase (EC 2.7.1.90) was used to measure the inorganic pyrophosphate in unfractionated extracts of tissues of Pisum sativum L. The fructose 1,6-bisphosphate produced by the above enzyme was measured by coupling to NADH oxidation via aldolase (EC 4.1.2.13), triosephosphate isomerase (EC 5.3.1.1) and glycerol-3-phosphate dehydrogenase (EC 1.1.1.8). Amounts of pyrophosphate as low as 1 nmol could be measured. The contents of pyrophosphate in the developing embryo of pea, and in the apical 2 cm of the roots, were appreciable; 9.4 and 8.9 nmol g^-1 fresh weight, respectively. The possibility that pyrophosphate acts in vivo as an energy source for pyrophosphate: fructose 6-phosphate 1-phosphotransferase and for UDPglucose pyrophosphorylase (EC 2.7.7.9) is considered.     

Studies with a human plasma-derived immunosuppressive,anti-lymphoma factor

Summary A low-molecular-weight (1,400) factor isolated from a human plasma -globulin concentrate by acid-salt dissociation and ultrafiltration inhibits proliferation of mitogen-stimulated T cells and L1210 leukemia cells. The factor (UM05R) inhibits DNA, RNA, and protein synthesis in sensitive cells, acts in G1 of the cell cycle, and appears to suppress mitogen-responsive T cells without an accessory cell requirement. UM05R activity is enhanced by known cAMP-elevating agents and by sulfhydryl compounds. The results of the present study are consistent with the hypothesis that the plasma-derived agent inhibits lympho-proliferation as a result of elevation of intracellular cAMP.     

Measurement of diameters of estuarine bacteria and particulates in natural water samples by use of a submicron particle analyzer

Desulfovibrio vulgaris strain Madison outcompetedMethanobacterium strain ivanov for hydrogen when sulfate was in excess because of higher cell yield and growth rate and a greater affinity for hydrogen as a consequence of a lower Km and higher Vmax for in vivo hydrogenase activity.Desulfovibrio vulgaris displayed a growth yield of 1.1 g/mol H₂, a Km for tritium exchange of 4 M, and a specific in vivo hydrogenase activity of 2.17 DPM³H₂O×10³/g cell protein/h; whereasMethanobacterium strain ivanov had a yield of 0.6 g/mol H₂, a Km for tritium exchange of 14 M, and a specific in vivo hydrogenase activity of 0.38 DPM³H₂O×10³/g cell protein/h. Under these physiological conditions, the Gibbs free-energy change associated with methanogenesis and sulfidogenesis from H₂ was calculated to be-47.4 kJ/mol and-62.9 kJ/mol, respectively. When sulfidogenesis was limited by sulfate concentration, the methanogen was able to successfully compete with the sulfidogen for hydrogen. Competition between methanogens and sulfidogens for hydrogen is explained in terms of thermodynamic, kinetic, and other important considerations not discussed in the previous literature.     

Effects of cadmium accumulation on growth and respiration of a cadmium-sensitive strain of Bacillus subtilis and a selected cadmium resistant mutant

The effects of cadmium on the growth and respiration of two strains of Bacillus subtilis are compared to the accumulation of Cd by viable and cyanide-killed cells, protoplasts and cell fractions of the strains. Growth and respiration of strain 1A1 were significantly inhibited at 10g Cd²⁺/ml while the growth and respiration of strain 1A1R, a selected mutant of 1A1, were only slightly affected. Similarly, 1A1R protoplasts were more resistant to Cd than were 1A1 protoplasts. The differential resistance of the strains correlates with the accumulation of Cd by the two strains, with 1A1 accumulating approximately 10 times the level of Cd after a 4 h exposure to 1 g Cd²⁺/ml. The distributions of Cd throughout the cells, however, were similar between strains.
