Summary Carpophores were developed on defined medium from explants of stipes produced in the dark using an expansionless mutant (no. 190) ofCoprinus cinereus. Explants taken from stipes within 24 h of formation developed multiple fruit within 3–12 days without formation of mycelia. Fruiting levels were affected by stipe age (24 h), medium composition, explant size, and polarity of the explant on fruiting medium. This method offers a new tool for developmental studies and may also be of use to commercial mushroom growers.  相似文献

Two genes (PPH21 and PPH22) encoding the yeast homologues of protein serine-threonine phosphatase 2A have been cloned from a Saccharomyces cerevisiae genomic library using a rabbit protein phosphatase 2A cDNA as a hybridization probe. The PPH genes are genetically linked on chromosome IV and are predicted to encode polypeptides each with 74% amino acid sequence identity to rabbit type 2A protein phosphatase, indicating once again the extraordinarily high degree of sequence conservation shown by protein-phosphatases from different species. The two PPH genes show less than 10% amino acid sequence divergence from each other and while disruption of either PPH gene alone is without any major effect, the double disruption is lethal. This indicates that protein phosphatase 2A activity is an essential cellular function in yeast. Measurement of type 2A protein phosphatase activity in yeast strains lacking one or other of the genes indicates that they account for most, if not all, protein phosphatase 2A activity in the cell.  相似文献

Calcium oxalate crystal growth and aggregation leads to the formation of renal calculi. It is known to be inhibited by several compounds both in vitro and in vivo conditions. The present study highlights the inhibitory potential of sodium pentosan polysulphate (SPP), a semi-synthetic glycosaminoglycan (GAG) on calcium oxalate crystal growth in vitro. Its efficacy was compared with those of known inhibitors like pyrophosphate, heparin and chondroitin-4-sulphate. Of the above compounds pyrophosphate was found to be the most potent inhibitor. Among the GAGs, SPP exhibited 80% inhibitory activity as compared to heparin. A lesser degree of inhibition was observed with chondroitin-4-sulphate.  相似文献

Two different genes encoding class II chitinases from peanut (Arachis hypogaea L. cv. NC4), A.h.Chi2;1 and A.h.Chi2;2, have been cloned. In peanut cell suspension cultures, mRNA levels of A.h.Chi2;2 increased after ethylene or salicylate treatment and in the presence of conidia from Botrytis cinerea. The second gene, A.h.Chi2;1, was only expressed after treatment with the fungal spores. Transgenic tobacco plants containing the complete peanut A.h.Chi2;1 gene exhibited essentially the same expression pattern in leaves as observed in peanut cell cultures. Expression characteristics of transgenic tobacco carrying a promoter-GUS fusion of A.h.Chi2;1 are described.  相似文献

To model nitrification rates in soils, it is necessary to have equations that accurately describe the effect of environmental variables on nitrification rates. A variety of equations have been used previously to describe the effect of temperature on rates of microbial processes. It is not clear which of these best describes the influence of temperature on nitrification rates in soil. I compared five equations for describing the effects of temperature on nitrification in two soils with very different temperature optima from a California oak woodland-annual grassland. The most appropriate equation depended on the range of temperatures being evaluated. A generalized Poisson density function best described temperature effects on nitrification rates in both soils over the range of 5 to 50 °C; however, the Arrhenius equation best described temperature effects over the narrower range of soil temperatures that normally occurs in the ecosystem (5 to 28 °C). Temperature optima for nitrification in most of the soils were greater than even the highest soil temperatures recorded at the sites. A model accounting for increased maintenance energy requirements at higher temperatures demonstrates how net energy production, rather than the gross energy production from nitrification, is maximized during adaptation by nitrifier populations to soil temperatures.  相似文献

This paper explores the consequences of the theoretical forward activation enzymatic pathway A 0 A 1 A 2 A 3 where E 1 convents A 0 to A 1, E 2 converts A 1 to A 2 and E 3 converts A 2 to A 3. A 0, which is environmentally determined, also serves to activate (or modulate) the activity of E 3 in such a way as to keep the concentration of A 2 ([A 2]) constant at a particular set-point value. For mathematical simplicity, first order rate kinetics are used where k 1, k 2 and k 3 are the rate constants for E 1, E 2, and E 3 respectively. It is shown that if k 3 is modulated appropriately so as keep [A 2] at the setpoint value with a changing upstream [A 0], then the modulation of k 3 must be anticipatory of the dynamics of the biochemical pathway. In other words, the rate of change of k 3, will be a function of [A 0], k 1, k 2, k 3, and the set-point value of [A 2]. If the modulation of k 3 does not perfectly model (anticipate) the reaction pathway of which it is a part, then the actual [A 2] will deviate from the set-point value. This type of anticipatory feed-forward activation may represent an important aspect of biological organization.  相似文献

Five allozyme polymorphisms were used to analyze the mating system in a Sitka spruce seed orchard in Saanichton, British Columbia. Allelic frequencies differed between the pollen and maternal pools at three of the five loci, with alleles rare in the maternal pool being even rarer in the effective pollen pool. Minor differences in pollen allelic frequencies were observed in the upper vs. lower crown. The multilocus outcrossing rate of the upper crown (tm = 0.909) exceeded that of the lower crown (tm = 0.764). Single-locus estimates of the outcrossing rate were significantly heterogeneous, with the lowest estimate of outcrossing, t = 0.773, observed for PGM-2 locus. Analyses of the mating system for the three maternal PGM-2 genotypes revealed heterogeneous pollen allelic frequencies and heterogeneous outcrossing rates, possibly due to assortative mating at this locus.  相似文献

We review the recent history of the Mute Swan in Great Britain and discuss the factors known to be affecting the population. Following a rise in the national population during the 1950s, numbers decreased sharply during the 1960s, and changed relatively little between 1970/71 and 1984/85. However, there has been considerable regional variation in the fortunes of Mute Swan populations during this period, with dramatic declines in some areas. Although several factors were thought to be contributing to such declines, poisoning from the ingestion of lead fishing weights was shown to be the largest single cause of death amongst swans in a number of areas. Voluntary measures to address this problem were initiated in 1982 and culminated in the banning by law of use of lead weights in 1987.Winter counts were used to investigate the current status and distribution of the Mute Swan in Great Britain and to examine long-term regional trends. The maximum total count reached 12600 birds in January 1990, which compares with an average of 9550 for the previous five winters. However, accounting for birds missed, the population may now number at least 25 000. Peak total numbers have mostly occurred in September, after which numbers remain approximately stable until December and then decline. Patterns of seasonal abundance vary between regions and habitats and these are discussed.The British population has increased dramatically since 1986/87 and reached its highest level for 27 years in 1987/88. There have been recent increases in most regions with record levels being reached mostly in 1987 or 1988, and there has been growth in the numbers on all habitat types, especially on reservoirs, gravel extraction pits and freshwater marshes. The timing of these increases corresponds very closely with the introduction of legislation against the use of lead fishing weights, and the incidence of lead poisoning is known to have been considerably reduced by such measures.  相似文献

Phylogenetic and physiological methods were used to study the evolution of the opsin gene family in Drosophila. A phylogeny based on DNA sequences from 13 opsin genes including representatives from the two major subgenera of Drosophila shows six major, well-supported clades: The blue opsin clade includes all of the Rhl and Rh2 genes and is separated into two distinct subclades of Rhl sequences and Rh2 sequences; the ultraviolet opsin clade includes all Rh3 and Rh4 genes and bifurcates into separate Rh3 and Rh4 clades. The duplications that generated this gene family most likely took place before the evolution of the subgenera Drosophila and Sophophora and their component species groups. Numerous changes have occurred in these genes since the duplications, including the loss and/or gain of introns in the different genes and even within the Rhl and Rh4 clades. Despite these changes, the spectral sensitivity of each of the opsins has remained remarkably fixed in a sample of four species representing two species groups in each of the two subgenera. All of the strains that were investigated had R1-6 (Rhl) spectral sensitivity curves that peaked at or near 480 nm, R7 (Rh3 and Rh4) peaks in the ultraviolet range, and ocellar (Rh2) peaks near 420 nm. Each of the four gene clades on the phylogeny exhibits very conservative patterns of amino acid replacement in domains of the protein thought to influence spectral sen sitivity, reflecting strong constraints on the spectrum of light visible to Drosophila.  相似文献

We studied an imported host-parasitoid community in Hawaii, asking to what extent the species covaried in a systematic fashion even though all species were exotic to Hawaii, and occurred in an artificial agroecosystem (a commercial guava, Psidium guajava L., orchard). Using knock-down pyrethrin sprays we were able to accurately quantify numbers of the host, [oriental fruit fly, Bactrocera dorsalis (Hendel)] and its four major parasitoid species [Biosteres arisanus (Sonan), Diachasmimorpha longicaudata (Ashmead), Psyttalia incisi (Silvestri), and Bi. vandenboschi (Fullaway)] at hourly intervals. We found that the parasitoids' activity and abundance was well correlated with the activity and abundance of their host, and that all four parasitoid species covaried in concert with one another. In fact, the magnitude of correlation between the different species in this system was greater than the correlation with temperature. This show clearly that an entirely exotic community, reassembled piecemeal as a result of biocontrol efforts, can end up with patterns of temporal covariation that are highly coincident. One other interesting result concerns the speed with which sprayed trees were recolonized by the fruit fly and its parasitoids. The time that it took each species to reach its mean density prior to removal by the first pyrethrin spray at 0600 hours varied. It took 2 h for female B. dorsalis to recolonize guava trees to pre-spray levels. It took 3 h for Bi. arisanus, 4 h for D. longicaudata, 7 h for Bi. vandenboschi and 14 h for P. incisi to reach pre-spray levels. The fact that Bi. arisanus recolonized vacant trees almost as rapidly as did the fruit fly pest suggest that there is little opportunity for the fruit fly to escape in space and time by staying one step ahead of its enemies.  相似文献
