Microbial extracts continue to be a productive source of new molecules with biotechnological importance. Fungi of the genus Penicillium are known to produce biologically active secondary metabolites. The goal of this work is verify the production of antimicrobial metabolites by Penicillium chrysogenum IFL1 using agro-industrial residues. P. chrysogenum IFL1 produced active metabolites growing on the agro-industrial residues, grape waste and cheese whey. The 7-day cultures showed antimicrobial activities against bacteria, fungi and amoebae. The filtrate of the cheese whey culture inhibited the growth of the bacteria Staphylococcus aureus, Bacillus cereus and Pseudomonas aeruginosa, the fungus Fusarium oxysporum and the amoeba Acanthamoeba polyphaga. Due to the greater antimicrobial activity of the cheese whey culture, a footprinting profile was carried out using the ESI-MS and ESI-MS/MS techniques. The presence of penicillin G and other metabolites that have antimicrobial activity such as penicillin V and rugulosin can be suggested. P. chrysogenum IFL1 was able to produce a wide variety of antimicrobial compounds on agro-industrial residues, which makes the process ecologically friendly.  相似文献

Howler monkey capture is an arduous and expensive task requiring trained and specialized professionals. We compared strategies and methods to most efficiently capture Alouatta guariba clamitans in remnants of the Atlantic Forest in Rio de Janeiro and its bordering states of Minas Gerais and São Paulo. We tested whether or not the success of expeditions in the forest with anesthetic darts, nets, and baited traps differed with and without the support of an information network, a contact chain built with key institutions and inhabitants to continuously monitor howler monkey presence. The influence of forest conditions (vegetation type and fragment size) upon darting success was also evaluated. We captured 24 free‐living A. guariba clamitans. No howler monkey was caught with traps, probably due to the predominantly folivore feeding to high local plant diversity providing a great variety of food options. Captures based on an information network were significantly more efficient in terms of numbers of caught monkeys than without it. Captures with darts were considerably more efficient when performed in semideciduous forests and small forest fragments as opposed to ombrophilous forests or large woods. Although we walked great distances within the forest searching for howler monkeys, all but one animal were captured at the forest fringes. Hindrances to search and the darting method in the Atlantic Forest, for example, the steep terrain, high tree canopies, hunt pressure, and low A. guariba clamitans population density, were mitigated with the use of the information network in this monkey capture. Moreover, the information network enhanced the surveillance of zoonotic diseases, which howler monkeys and other nonhuman primates are reservoirs in Brazil, such as malaria and yellow fever.  相似文献

The long-term soil management effects on C and N stocks of soil physical fractions are still poorly understood for South American subtropical soils. This study aimed (i) to evaluate the influence of cereal- and legume-based cropping systems and N fertilisation on C and N stocks of the sand-, silt- and clay-size fractions of a no-tilled subtropical Acrisol in southern Brazil, (ii) to compute the Carbon Management Index (CMI) for those cropping systems using physical fractionation data, and (iii) to investigate the possible existence of finite capacity of those soil physical fractions to store C and N. Soil samples of a long-term experiment were collected from the 0–2.5 and 2.5–7.5 cm layers of three no-till cropping systems [fallow bare soil, oat/maize (O/M) and pigeon pea+maize (P+M)] under two N fertilisation levels (0 and 180 kg N ha–1). However, for fallow bare soil, only the non-fertilised sub-plot was sampled. An adjacent native grassland soil was sampled as a reference. The C and N stocks of the three soil physical fractions were higher in the legume-based cropping system (P+M) than in O/M and bare soil, because of the higher residue input in the former. The P+M cropping system restored the C and N stocks in sand- and silt-size fractions to the same levels found in grassland soil. Higher C and N stocks in all physical fractions were also obtained with N fertilisation. The C and N stocks and the C:N ratio were most affected by cropping systems in the sand- and least in the clay-size fraction. Particulate organic matter was found in the silt-size fraction, showing this fraction is not only constituted by mineral-associated organic mater, as commonly believed. Taking grassland soil as reference (CMI = 100), the CMI ranged from 46, in O/M no N, to 517, in P+M with N, pointing to a better soil management in the latter. The clay-size fraction tended to show a finite capacity to store C and N (48.8 g C kg–1 and 4.9 g N kg–1 of clay), which was not verified in sand- and silt-size fractions. The adoption of no-tillage and legume-based cropping systems with high residue input are adequate soil management strategies to improve soil quality and make the agricultural production systems more sustainable in subtropical regions.  相似文献

Seed predators that severely affect seed germination rates are well known for many plant species. Here, we hypothesised that due to differences in resource allocation within fruits, seed predation can negatively affect non-predated seeds in infested fruits when predation occurs during fruit maturation (a ‘top-down’ effect). We addressed this question using a system of bruchid beetles on Mimosa trees and we also investigated whether seed quality (nitrogen concentration) affects beetle body mass, which would have implications for adult fitness (‘bottom-up’ effect). To assess spatial variation, bottom-up and top-down effects were investigated in two plant populations. Nitrogen concentration was significantly higher in seeds from non-infested fruits than from infested fruits. This supports the hypothesis that resource allocation may differ between seeds from infested and non-infested fruits. Germination experiments showed that seeds from non-infested fruits germinated better than non-predated seeds from infested fruits. It was also confirmed that seed quality affected bruchid body mass. There was also evidence that more resources were taken from well-developed seeds. These results showed that seed predation can damage non-predated seeds.  相似文献

The ant Mycocepurus goeldii (Forel) is known for having a relict karyotype with low chromosome number and the present study help the understanding of this ant cytogenetics by describing the occurrence of pre-nucleolar bodies in their chromosomes using impregnation with silver nitrate (Ag-NOR) and the location of 45S rDNA sites by means of the FISH (fluorescent in situ hybridization) technique. Several spots were observed surrounding all chromosomes when submitted to the Ag-NOR technique. These unusual markings were observed in both chromatids of metaphase and early anaphase chromosomes, and are associated to the presence of pre-nucleolar bodies, allowing the observation of the phenomenon of nucleologenesis. Although recent studies have shown that all chromosomes of M. goeldii exhibit centromeric or pericentromeric markings for the CMA(3) fluorochrome, the FISH technique indicated the presence of 45S rDNA in only one pair of chromosomes that differed in the number of CMA(3) markings observed for this species, pointing that the other markings observed with this fluorochrome do not match the sequences in ribosomal genes.  相似文献

Background  

Theories about eukaryote origins (eukaryogenesis) need to provide unified explanations for the emergence of diverse complex features that define this lineage. Models that propose a prokaryote-to-eukaryote transition are gridlocked between the opposing "phagocytosis first" and "mitochondria as seed" paradigms, neither of which fully explain the origins of eukaryote cell complexity. Sex (outcrossing with meiosis) is an example of an elaborate trait not yet satisfactorily addressed in theories about eukaryogenesis. The ancestral nature of meiosis and its dependence on eukaryote cell biology suggest that the emergence of sex and eukaryogenesis were simultaneous and synergic and may be explained by a common selective pressure.  相似文献

The genus Trachymyrmex is a key group in the tribe Attini because of its close phylogenetic relationship to leaf-cutter ants, Acromyrmex and Atta. Cytogenetic data are only available for five taxa of Trachymyrmex, with chromosome numbers of 2n = 12, 18, 20 and 22, and morphology with predominantly metacentric chromosomes. The aim of the present study was to characterize the karyotype of the ant Trachymyrmex fuscus Emery, 1934, by means of the number and morphology of its chromosomes, heterochromatin pattern, CMA3 and DAPI fluorochromes in the population of two nests collected at Paraopeba, state of Minas Gerais, Brazil. Nineteen females presented 2n = 18 chromosomes (16m + 2sm) and a single male presented n = 9 (8m + 1sm). A size chromosomal polymorphism involving the short arm of the submetacentric pair was confirmed by statistical analysis, with three character conditions: heterozygous SB (with a difference in size between the short arms), standard SS (smaller short arms) and homozygote BB (bigger short arms). In the first nest, both SB and SS workers were observed. The other nest contained heterozygous (SB), homozygous (BB), and a male carrying the B chromosome (larger size). The presence of heterochromatin on all centromeric and pericentromeric chromosomes of T. fuscus suggests that the size difference observed in the submetacentric pair in the SB and BB workers is not related to the heterochromatin but to a duplication of euchromatic regions through intra- or inter- chromosomal rearrangements. The fluorochrome CMA3 matched the C-banding markings, indicating that the heterochromatin is rich in GC base pairs. As far as we know, this is the first chromosomal polymorphism reported in the tribe Attini.  相似文献
