Poly(A)-containing RNA from frozen adult rat brain were fractionated by centrifugation in a formamide/sucrose gradient. Individual fractions were used to program protein synthesis in vitro in a reticulocyte lysate. The cell-free translation products were analyzed by two-dimensional electrophoresis in polyacrylamide slab gels. We observed a heterodispersion of the mRNA translation activity coding for the beta-tubulin subunit which contrasts with a relatively homogeneous distribution of the alpha-tubulin subunit mRNA. These last mRNA species are present in a peak which sediments near the 18-S region of the gradient whereas the beta-tubulin mRNA activity is predominant in the fractions corresponding to the heaviest mRNA species. When these heaviest RNAs were separated again by centrifugation in a second formamide/sucrose gradient, a poly(A)-rich RNA population was obtained that was enriched in RNA for programming the beta-tubulin subunit. Analysis of the products whose synthesis in vitro was directed by this mRNA population revealed that beta tubulin was the main protein formed, the ratio beta/alpha being more than tenfold greater than in the products translated in vitro using total poly(A)-rich RNA. 相似文献
Bromadiolone damaged the erythrocytes, resulting in a probable saturation of transferrin, a deposit of iron in the connective tissue and in a few cells of the proximal tubules of the kidneys and an increased storage of ferritin in the spleen. In the hepatocytes, mitochondria were distorted, their lipid inclusions being granular; a large depletion of glycogen may be considered a reflection of an elevated phosphorylase a ascribable to the proliferation of the smooth endoplasmic reticulum. In the kidneys, pyelonephritis may be irrelevant to the poisoning of the animals. Bromine could not be detected using microanalytical methods. 相似文献
Tubulin heterogeneity was analyzed during morphological differentiation of a mouse neuroblastoma clone (C 1300). One form of β-tubulin was found to be specific of the differentiated cells. The synthesis of this isoform is strictly related to neurite outgrowth process and has been shown to be regulated at the post-translational level. 相似文献
The aim of the present work is to gather new information on the ferritin molecule. Natural crystals of ferritin occurring in the yolk platelets of a mollusc oocyte were studied. Their crystallographic structure was found to be equivalent to one of the structures obtained by artificial crystallization (fcc; a = 15 nm). Individual ferritin particles isolated from horse spleen were studied by microdiffraction techniques, using field emission gun transmission electron microscopy. The iron core crystals display a hexagonal structure; our results confirm the value of the unit cell parameter a (0.51 nm) and, for the first time, we have been able to extract the value of the unit cell parameter c (0.95 nm). Thus, among the three models described in the literature for the crystalline structure of the iron complex, our results corroborate that of Towe and Bradley (J. Colloid. Interf. Analysis of mouse brain myelin by sodium dodecyl sulfate-polyacrylamide gel electrophoresis showed that in the high-molecular-weight range it contained, besides the Wolfgram protein doublet, proteins comigrating with actin and with both subunits of tubulin. The occurrence of these alpha and beta subunits was confirmed by peptide mapping in myelin analyzed by two-dimensional electrophoresis. This tubulin did not arise from an artifactual binding of soluble brain tubulin to the myelin fraction: addition of exogenously labeled tubulin to brain homogenates proved that during myelin isolation by the procedure of Norton and Poduslo (1973) the contaminating tubulin was washed out. On the other hand, the distribution of tubulin isoforms in myelin was investigated by isoelectric focusing and compared with the distribution of the 21 isoforms listed for the whole brain soluble tubulin. It was shown that many isoforms were found in myelin (three isoforms for the alpha subunit and nine for the beta subunit), and that some isoforms were represented both in myelin and in soluble tubulin, but in different relative proportions. 相似文献
The advantages of nanofiltration membranes coupled with a CSTR were demonstrated for the semicontinuous production of lactic acid from whey permeate. Lactic acid was removed from the growth medium while lactose was kept in the bioreactor with the bacterial cells; moreover, Mg2+ ions were also recycled in the bioreactor at 96% and the nanofiltrate color was greatly reduced. The highest volumetric productivity achieved with this device was 7.1 g l−1 h−1 and the lactate concentration was 55 g l−1. The specific productivity was 3.54 h−1. More than 99% of the membrane fouling after 44 h of fermentation was reversible. The initial permeate flux was restored easily by a water rinse. The performance of this type of membrane bioreactor was discussed. 相似文献
Fibropapillomatosis (FP) threatens the survival of green turtle (Chelonia mydas) populations at a global scale, and human activities are regularly pointed as causes of high FP prevalence. However, the association of ecological factors with the disease’s severity in complex coastal systems has not been well established and requires further studies. Based on a set of 405 individuals caught over ten years, this preliminary study provides the first insight of FP in Martinique Island, which is a critical development area for immature green turtles. Our main results are: (i) 12.8% of the individuals were affected by FP, (ii) FP has different prevalence and temporal evolution between very close sites, (iii) green turtles are more frequently affected on the upper body part such as eyes (41.4%), fore flippers (21.9%), and the neck (9.4%), and (iv) high densities of individuals are observed on restricted areas. We hypothesise that turtle’s aggregation enhances horizontal transmission of the disease. FP could represent a risk for immature green turtles’ survival in the French West Indies, a critical development area, which replenishes the entire Atlantic population. Continuing scientific monitoring is required to identify which factors are implicated in this panzootic disease and ensure the conservation of the green turtle at an international scale.
