Nicotiana tabacum BY-2 suspension cells have several advantages that make them suitable for the production of full-size monoclonal antibodies which can be purified directly from the culture medium. Carbohydrate characterization of an antibody (Lo-BM2) expressed in N. tabacum BY-2 cells showed that the purified Lo-BM2 displays N-glycan homogeneity with a high proportion (>70%) of the complex GnGnXF glycoform. The stable co-expression of a human β-1,4-galactosyltransferase targeted to different Golgi sub-compartments altered Lo-BM2N-glycosylation and resulted in the production of an antibody that exhibited either hybrid structures containing a low abundance of the plant epitopes (α-1,3-fucose and β-1,2-xylose), or a large amount of galactose-extended N-glycan structures. These results demonstrate the suitability of stable N-glycoengineered N. tabacum BY-2 cell lines for the production of human-like antibodies.  相似文献

Many spiders depend upon webs to capture prey. Web function results from architecture and mechanical performance of the silk. We hypothesized that the common house spider, Achaearanea tepidariorum, would alter the mechanical performance of its cobweb in response to different prey by varying the structural and material properties of its silk. We fed spiders either large, high kinetic energy crickets or small, low kinetic energy pillbugs for 1 week and then examined their freshly spun silk. We separated mechanical performance into structural and material effects. We measured both types of properties for silk threads collected directly from cobwebs to test for "tuning" of silk performance to different aspects of prey capture. We compared silk from two different functional regions of the cobweb-sticky gumfooted threads that adhere directly to prey and supporting threads that maintain web integrity. Supporting threads from cricket-fed spiders were stiffer and tougher than supporting threads from pillbug-fed spiders. Both types of silk from cricket-fed spiders broke at higher loads than silk from pillbug-fed spiders. We explain this variation using a simple model of forces exerted by prey and spiders on single threads and propose potential mechanisms for this change in material properties. Two alternative, nonexclusive, hypotheses are suggested by our data. Spiders may tune silk to different types of prey by spinning threads that are able to hold prey without deforming permanently. Alternatively, as spider's body mass differed dramatically between the two feeding regimes, spiders may tune silk to their own body mass.  相似文献

The antifungal activity of several di(hetero)arylamine derivatives of the benzo[b]thiophene system was evaluated against clinically relevant Candida, Aspergillus, and dermatophyte species by a broth macrodilution test based on CLSI (formerly NCCLS) guidelines. The most active compound showed a broad spectrum of activity (against all tested fungal strains, including fluconazole-resistant fungi), with particularly low MICs for dermatophytes. Results from the inhibition of the dimorphic transition in Candida albicans and flow cytometry studies further confirmed their biological activity. With this study it was possible to establish some structure-activity relationships (SARs). The hydroxy groups proved to be essential for the activity in the aryl derivatives. Furthermore, the spectrum of activity in the pyridine derivatives was broadened by the absence of the ester group on position 2 of the benzo[b]thiophene system.  相似文献

The aim of this study was to provide the information necessary to enable the comparison of exposure conditions in different human volunteer studies published by the research groups at the Universities of Turku, Swinburne, and Zurich. The latter applied a setup optimized for human volunteer studies in the context of risk assessment while the first two applied a modified commercial mobile phone for which detailed dosimetric data were lacking. While the Zurich Setup exposed the entire cortex of the target hemisphere, the other two setups resulted in only very localized exposure of the upper cheek, and concentrated on a limited area of the middle temporal gyrus just above the ear. The resulting peak spatial SAR averaged over 1 g of the cortex was 0.19 W/kg of the Swinburne Setup, and 0.31 W/kg for the Turku Setup, compared to 1 W/kg for the Zurich Setup. The average exposure of the thalamus was 5% and 9% of the Zurich Setup results for the Swinburne and Turku Setups, respectively. In general, the phone-based setup results in only reasonably defined exposures in a very limited area around the maximum exposure; the exposure of the rest of the cortex was low, and may vary greatly as a function of the setup, position, and local anatomy. The analysis confirms the need for a carefully designed exposure setup that exposes the relevant brain areas to a well-defined level in human volunteer studies, and shows that studies can only be properly compared and replicated if sufficiently detailed dosimetric information is available.  相似文献

The plasma membrane proton pump ATPase (H(+)-ATPase) plays a central role in transport across the plasma membrane. As a primary transporter, it mediates ATP-dependent H(+) extrusion to the extracellular space, thus creating pH and potential differences across the plasma membrane that activate a large set of secondary transporters. In several species, the H(+)-ATPase is encoded by a family of approximately 10 genes, classified into 5 gene subfamilies and we might ask what can this tell us about the concept, and the evolution, of gene families in plants. All the highly expressed H(+)-ATPase genes are classified into only two gene subfamilies, which diverged before the emergence of present plant species, raising the questions of the significance of the existence of these two well-conserved subfamilies and whether this is related to different kinetic or regulatory properties. Finally, what can we learn from experimental approaches that silence specific genes? In this review, we would like to discuss these questions in the light of recent data.  相似文献

Focal adhesion kinase (FAK) is activated following integrin engagement or stimulation of transmembrane receptors. Autophosphorylation of FAK on Tyr-397 is a critical event, allowing binding of Src family kinases and activation of signal transduction pathways. Tissue-specific alternative splicing generates several isoforms of FAK with different autophosphorylation rates. Despite its importance, the mechanisms of FAK autophosphorylation and the basis for differences between isoforms are not known. We addressed these questions using isoforms of FAK expressed in brain. Autophosphorylation of FAK(+), which is identical to that of "standard" FAK, was intermolecular in transfected cells, although it did not involve the formation of stable multimeric complexes. Coumermycin-induced dimerization of gyrase B-FAK(+) chimeras triggered autophosphorylation of Tyr-397. This was independent of cell adhesion but required the C terminus of the protein. In contrast, the elevated autophosphorylation of FAK(+6,7), the major neuronal splice isoform, was not accounted for by transphosphorylation. Specifically designed immune precipitate kinase assays confirmed that autophosphorylation of FAK(+) was intermolecular, whereas autophosphorylation of FAK(+6,7) or FAK(+7) was predominantly intramolecular and insensitive to the inhibitory effects of the N-terminal domain. Our results clarify the mechanisms of FAK activation and show how alternative splicing can dramatically alter the mechanism of autophosphorylation of a protein kinase.  相似文献

The longitudinal distribution of Trichoptera at 37 sampling sites along 800 km of the Loire River (France) was analyzed in order to better understand large-scale changes in community structure occurring on a stream continuum. Different simple ordination methods — centered PCA and Correspondence Analysis — were performed to investigate these changes. Both analyses showed that a major change occurs in the community in the upper reaches of the Loire River between the Serre de la Fare and Vorey sampling sites with the appearance of Hydropsyche contubernalis and H. Exocellata. Even though other changes in community structure could exist, revealed either by Correspondence Analysis at the first sampling sites (crenal), or by PCA at sampling sites influenced by dams, this Trichoptera community appears to be stable enough in the latter part of the Loire River with the addition of a few species, Hydropsyche bulgaromanorum, Ecnomus tenellus and E. deceptor, rather than changes in species. The results are discussed in the light of different concepts in stream ecology, and the validity of such data analyses to describe some community patterns.  相似文献

Synthesis and biological evaluation of novel and potent cyclohexylamine-based histamine H3 receptor inverse agonists are described. Compounds in this newly identified series exhibited subnanomolar binding affinities for human receptor and no significant interaction with hERG channel. One derivative (10t) demonstrated enhanced in vivo efficiency and preferential brain distribution, both properties suitable for potential clinical evaluation.  相似文献
