Kar2p, an essential Hsp70 chaperone in the endoplasmic reticulum of Saccharomyces cerevisiae, facilitates the transport and folding of nascent polypeptides within the endoplasmic reticulum lumen. The chaperone activity of Kar2p is regulated by its intrinsic ATPase activity that can be stimulated by two different nucleotide exchange factors, namely Sil1p and Lhs1p. Here, we demonstrate that the binding requirements for Lhs1p are complex, requiring both the nucleotide binding domain plus the linker domain of Kar2p. In contrast, the IIB domain of Kar2p is sufficient for binding of Sil1p, and point mutations within IIB specifically blocked Sil1p-dependent activation while remaining competent for activation by Lhs1p. Taken together, these results demonstrate that the interactions between Kar2p and its two nucleotide exchange factors can be functionally resolved and are thus mechanistically distinct.  相似文献

Phylogenetic analyses of the Dasyaceae based on sequence analysis of the large subunit of ribulose-1,5-bisphosphate carboxylase/oxygenase (rbcL) and 42 morphological characters are presented. Comparative sequence analysis confirms the general view of the Ceramiaceae as a primitive, paraphyletic group giving rise to the Rhodomelaceae, Delesseriaceae and Dasyaceae within the monophyletic Ceramiales. On the basis of both data sets, the Heterosiphonia-like genera (Heterosiphonia, Colacodasya and Dasyella) are the most primitive members of the Dasyaceae, whereas the Dasya-like genera (Dasya, Pogonophorella, Eupogodon and Rhodoptilum) and Thuretia and Dictyurus are of more recent origin. On the basis of morphological data only, Thuretia and Dictyurus form a sister group to Heterosiphonia, and Eupogodon is monophyletic whereas Dasya and Heterosiphonia are not. Primary radial symmetry has arisen once in the Dasya clade but is secondarily obscured in some species by heavy, asymmetrical cortication that gives the appearance of bilateral symmetry. By using an original method, the authors injected the system of canals and then studied microscopically their network in the femoral diaphysis of man and dog. In the human bone, the density varies between 1.09 and 2.63 canals per mm2, with an average of 1.83; in dogs the limits of variation are 1.98--2.86 canals per mm2. The canal calibre is of 68.8 microns in man as against 48.5 microns in dogs. Both parameters present a wider variability in the femoral compact bone of man than in dog. By calculation it was found that the canal density in dogs is very significantly increased as compared to man; on the contrary, the diameter presents in dogs very significantly decreased values as compared to those of the diaphyseal compact bone of man.  相似文献

In the design of potent and selective sphingosine-1-phosphate receptor agonists, we were able to identify two series of molecules based on phenylamide and phenylimidazole analogs of FTY-720. Several designed molecules in these scaffolds have demonstrated selectivity for S1P receptor subtype 1 versus 3 and excellent in vivo activity in mouse. Two molecules PPI-4621 (4b) and PPI-4691 (10a), demonstrated dose responsive lymphopenia, when administered orally.  相似文献

In this study several investigations and tests were performed to determine the antioxidant activity and the acetylcholinesterase and tyrosinase inhibitory potential of Pulmonaria officinalis and Centarium umbellatum aqueous extracts (10% mass) and ethanolic extracts (10% mass and 70% ethanol), respectively. Moreover, for each type of the prepared extracts of P. officinalis and of C. umbellatum the content in the biologically active compounds – polyphenols, flavones and proanthocyanidins was determined. The antioxidant activity was assessed using two methods, namely the 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay and reducing power assay. The analyzed plant extracts showed a high acetylcholinesterase and tyrosinase inhibitory activity in the range of 72.24–94.24% (at the highest used dose – 3 mg/mL), 66.96% and 94.03% (at 3 mg/mL), respectively correlated with a high DPPH radical inhibition – 70.29–84.9% (at 3 mg/mL). These medicinal plants could provide a potential natural source of bioactive compounds and could be beneficial to the human health, especially in the neurodegenerative disorders and as sources of natural antioxidants in food industry.  相似文献

Baseline genotypes were established for 256 individuals of Caulerpa collected from 27 field locations in Florida (including the Keys), the Bahamas, US Virgin Islands, and Honduras, nearly doubling the number of available GenBank sequences. On the basis of sequences from the nuclear rDNA‐ITS 1+2 and the chloroplast tufA regions, the phylogeny of Caulerpa was reassessed and the presence of invasive strains was determined. Surveys in central Florida and southern California of >100 saltwater aquarium shops and 90 internet sites revealed that >50% sold Caulerpa. Of the 14 Caulerpa species encountered, Caulerpa racemosa was the most common, followed by Caulerpa sertularioides, Caulerpa prolifera, Caulerpa mexicana, and Caulerpa serrulata. None of the >180 field‐collected individuals (representing 13 species) was the invasive strain of Caulerpa taxifolia or C. racemosa. With one exception (a sample of C. racemosa from a shop in southern California belonged to the invasive Clade III strain), no invasive strains were found in saltwater aquarium stores in Florida or on any of the internet sites. Although these results are encouraging, we recommend a ban on the sale of all Caulerpa species (including “live rock”) because: morphological identification of Caulerpa species is unreliable (>12% misidentification rate) and invasive strains can only be identified by their aligned DNA sequences, and because the potential capacity for invasive behavior in other Caulerpa species is far from clear. The addition of the Florida region to the genetic data base for Caulerpa provides a valuable proactive resource for invasion biologists as well as researchers interested in the evolution and speciation of Caulerpa.  相似文献
