Les observations faites sur deux cucurbitacées africaines, Lagenaria siceraria Molina (Standl.) et Citrullus lanatus Thumb (Matsum & Nakai) ont permis d’inventorier l’entomofaune (utiles et ravageurs) associée à la pratique culturale. L’évaluation des dégâts occasionnés par les ravageurs a mis en évidence leur impact sur la production. Suivant un schéma expérimental, des parcelles de 49 m2 traitées au CYPERCAL 50 EC (50 g/l de Cypermétrine) et au FURADAN 5G (50 g/kg de Carbofuran) et non traitées ont été disposées en blocs aléatoires. Les observations ont porté sur l’identification et le dénombrement des insectes, sur le pourcentage de plants attaqués ainsi que sur la production de baies mûres et de graines. Au total, 71 espèces ont été identifiées, réparties en 41 familles et 10 ordres (Coléoptères, Hyménoptères, Diptères, Lépidoptères, Orthoptères, Homoptères, Hétéroptères, Dictyoptères, Odonates et Dermaptères). Les principaux ravageurs appartiennent à la famille des Chrysomelidae (Lamprocopa occidentalis, Aulacophora foveicolis, Aulacophora africana, Asbecesta cyanipennis, Ootheca mutabilis et Lilioceris livida), des Coccinellidae (Henosepilachna elaterii et Henosepilachna reticulata) et des Meloidae (Mylabris holosericea). Plus de 50 % de plants sains ont été recensés dans les parcelles traitées contre 0 à 33 % dans les parcelles témoins. Les nombres moyens des baies obtenues dans les parcelles traitées (37,16 ± 2,99 pour L. siceraria et 42,33 ± 2,58 pour C. lanatus) sont significativement (P < 0,001) plus importants que ceux observés dans les parcelles témoins (29,33 ± 4,67 pour L. siceraria et 31,33 ± 3,5 pour C. lanatus). Les rendements moyens en graines dans les parcelles traitées (222,39 ± 15,78 kg/ha pour L. siceraria et 127,4 ± 8,16 kg/ha pour C. lanatus) sont significativement (P < 0,001) supérieurs à ceux observés dans les parcelles témoins (144,4 ± 14,52 kg/ha pour L. siceraria et 88,18 ± 8,09 kg/ha pour C. lanatus). Les traitements insecticides, en réduisant les populations de ravageurs et les dégâts sur les plants, ont entrainé une augmentation du rendement à l’hectare de plus de 40 %.  相似文献

Culicoides biting midges (Diptera: Ceratopogonidae) serve as biological vectors for several pathogens, including the Bluetongue virus and the recently described Schmallenberg virus in northern Europe. These diseases have caused considerable direct and indirect economic losses to the sheep and cattle industries. This study undertaken between August and December 2007 on a sheep farm in the Namur province (Belgium) aims to evaluate Culicoides populations present inside a partially opened sheepfold and in a nearby sheep meadow, using light traps. The comparative analysis of insects trapped at 18 dates at regular intervals showed that Culicoides were most abundant inside this livestock building (17,450 midges) than in surrounding meadow (1,121 midges); this meadow had however a greater species diversity. The two species C. obsoletus and C. scoticus constituting the Obsoletus complex predominated for all trappings and females were much more numerous than males. Important capture of engorged females of the Obsoletus complex inside the sheepfold seems to reflect the possibility of an opportunistic endophagous behavior. Maintaining sheep inside livestock buildings in order to reduce the risk of Culicoides bites – and thus of pathogens transmission – however requires to limit biting midge populations which are likely to enter or to develop inside these buildings. Implementation of effective sanitation and hygiene measures against midges present inside farms, as well as establishing of measures to protect livestock against intrusion and improvement of “midge-proofing” of animal housing are therefore highly recommended.  相似文献

Poly(ADP-ribosyl)ation is a posttranslational modification catalyzed by the poly(ADP-ribose) polymerases (PARPs). These enzymes covalently modify glutamic, aspartic and lysine amino acid side chains of acceptor proteins by the sequential addition of ADP-ribose (ADPr) units. The poly(ADP-ribose) (pADPr) polymers formed alter the physico-chemical characteristics of the substrate with functional consequences on its biological activities. Recently, non-covalent binding to pADPr has emerged as a key mechanism to modulate and coordinate several intracellular pathways including the DNA damage response, protein stability and cell death. In this review, we describe the basis of non-covalent binding to pADPr that has led to the emerging concept of pADPr-responsive signaling pathways. This review emphasizes the structural elements and the modular strategies developed by pADPr-binding proteins to exert a fine-tuned control of a variety of pathways. Poly(ADP-ribosyl)ation reactions are highly regulated processes, both spatially and temporally, for which at least four specialized pADPr-binding modules accommodate different pADPr structures and reprogram protein functions. In this review, we highlight the role of well-characterized and newly discovered pADPr-binding modules in a diverse set of physiological functions.  相似文献

Objectives. To determine the antioxidant activities of nonsteroidal anti-inflammatory drugs (NSAIDS), we examined by chemiluminescence (CL) and electron spin resonance (ESR) their scavenging properties towards lipid peroxides, hypochlorous acid and peroxynitrite.

Methods. The antioxidant properties of nimesulide (NIM), 4-hydroxynimesulide (4-HONIM), aceclofenac (ACLO), 4-hydroxyaceclofenac (4-HOA-CLO), diclofenac (DICLO) and indomethacin (INDO) were tested on four different reactive oxygen species (ROS) generating systems: (I) phorbol-myristate acetate (PMA)-activated neutrophils, (II) Fe2+/ascorbate-induced lipid peroxidation, (III) HOCl-induced light emission, (IV) the kinetics of ONOO- decomposition followed by spectrophotometry. ROS production was monitored by luminol-enhanced CL or by ESR using two different spin traps.

Results. At 10 μM, ACLO, NIM, 4-HONIM, 4-HOA-CLO, and DICLO decreased luminol-enhanced CL generated by PMA-activated neutrophils. Inversely, INDO increased the luminol enhanced CL. Interestingly, hydroxylated metabolites were more potent antioxidants than the parent drugs. Furthermore, all drugs tested, excepted ACLO, lowered lipid peroxidation induced by Fe2+/ascorbate system. ACLO and DICLO, even at the highest concentration tested (100 μM), did not significantly lower HOCl induced CL, whereas the other drugs were potent scavengers. Finally, all the NSAIDS accelerated decomposition of ONOO-, suggesting a potential capacity of the molecules to scavenge peroxynitrite.

Conclusion. The NSAIDs possess variable degrees of antioxidant activities, linked to their ability to react with HOCl, lipid peroxides or ONOO-. These antioxidant activities could offer interesting targeted side-effects in the treatment of joint inflammatory diseases.  相似文献

High levels of antibiotic tolerance are a hallmark of bacterial biofilms. In contrast to well-characterized inherited antibiotic resistance, molecular mechanisms leading to reversible and transient antibiotic tolerance displayed by biofilm bacteria are still poorly understood. The physiological heterogeneity of biofilms influences the formation of transient specialized subpopulations that may be more tolerant to antibiotics. In this study, we used random transposon mutagenesis to identify biofilm-specific tolerant mutants normally exhibited by subpopulations located in specialized niches of heterogeneous biofilms. Using Escherichia coli as a model organism, we demonstrated, through identification of amino acid auxotroph mutants, that starved biofilms exhibited significantly greater tolerance towards fluoroquinolone ofloxacin than their planktonic counterparts. We demonstrated that the biofilm-associated tolerance to ofloxacin was fully dependent on a functional SOS response upon starvation to both amino acids and carbon source and partially dependent on the stringent response upon leucine starvation. However, the biofilm-specific ofloxacin increased tolerance did not involve any of the SOS-induced toxin–antitoxin systems previously associated with formation of highly tolerant persisters. We further demonstrated that ofloxacin tolerance was induced as a function of biofilm age, which was dependent on the SOS response. Our results therefore show that the SOS stress response induced in heterogeneous and nutrient-deprived biofilm microenvironments is a molecular mechanism leading to biofilm-specific high tolerance to the fluoroquinolone ofloxacin.  相似文献

Hydrolysis of ATP by partition ATPases, although considered a key step in the segregation mechanism that assures stable inheritance of plasmids, is intrinsically very weak. The cognate centromere-binding protein (CBP), together with DNA, stimulates the ATPase to hydrolyse ATP and to undertake the relocation that incites plasmid movement, apparently confirming the need for hydrolysis in partition. However, ATP-binding alone changes ATPase conformation and properties, making it difficult to rigorously distinguish the substrate and cofactor roles of ATP in vivo. We had shown that mutation of arginines R36 and R42 in the F plasmid CBP, SopB, reduces stimulation of SopA-catalyzed ATP hydrolysis without changing SopA-SopB affinity, suggesting the role of hydrolysis could be analyzed using SopA with normal conformational responses to ATP. Here, we report that strongly reducing SopB-mediated stimulation of ATP hydrolysis results in only slight destabilization of mini-F, although the instability, as well as an increase in mini-F clustering, is proportional to the ATPase deficit. Unexpectedly, the reduced stimulation also increased the frequency of SopA relocation over the nucleoid. The increase was due to drastic shortening of the period spent by SopA at nucleoid ends; average speed of migration per se was unchanged. Reduced ATP hydrolysis was also associated with pronounced deviations in positioning of mini-F, though time-averaged positions changed only modestly. Thus, by specifically targeting SopB-stimulated ATP hydrolysis our study reveals that even at levels of ATPase which reduce the efficiency of splitting clusters and the constancy of plasmid positioning, SopB still activates SopA mobility and plasmid positioning, and sustains near wild type levels of plasmid stability.  相似文献

In vitro nickel (II) complex presents antimitotic effects. In this work, we have studied the in vivo seasonal effects of nickel (II), ligand and the complex [NiCl2(M5FTSC)2] in male Swiss mice. During spring, an intra peritoneal (i.p.) injection of NiCl2 in aqueous NaCl up to 1.0.10-2 mmol.kg-1 body weight (b.w.) killed 10% of the rodents after 6 days. Lethal dose 100% (LD100) was up 1.91.10-2 mmol.kg-1 b.w.; ligand was less toxic than Ni (II), while the complex was 25% lethal at 1.37.10-2 mmol.kg-1 b.w. In autumn, mice were less sensitive to NiCl2. The ligand and the complex, on the contrary, were more toxic. This leads us to the conclusion that, in vivo, chronotolerance of nickel (II), ligand and complex in aqueous solution, are quite different in spring and autumn seasons.  相似文献
