Structural and ultrastructural studies on the gut of the chaetognath Spadella cephaloptera, as well as observations on the feeding behavior of specimens bred in the laboratory, were conducted. The gut displays four distinct zones: pharynx, esophagus, and intestine, to which are connected a pair of diverticula, and the rectum, differing in length, shape, and cellular composition. The intestine alone represents ∼90% of the gut length. Upon ingestion, food in the intestine is submitted to successive backward and forward peristaltic movements until digestion has ended. Ultrastructural observations have identified five distinct cell types from granule morphology and the presence or absence of cilia at the apex of the cells. Three of the types undoubtedly correspond to secretory cells. They are the (1) pharyngeal, (2) esophageal, and (3) light intestinal ciliated cells, which could be, respectively, implicated in (1) mucous, (2) enzyme, and (3) both mucous and enzyme secretions. The fourth type, which corresponds to dense intestinal ciliated cells, displays all the characteristics of cells specialized in the absorption of macromolecules and intracellular digestion. The products of this digestion could be temporarily stored inside dense granules before being utilized during vitellogenesis. Except for the presence of cilia, the fifth type, which is localized in the short rectum, represents a common polyhedral epithelial cell type. © 1996 Wiley-Liss, Inc.  相似文献

The potential pluripotency of rabbit fetal germ cells has been investigated by using them to make chimeric embryos. Gonial cells, isolated from enzyme-dispersed male and female transgenic fetal rabbit gonads of 18–22 days gestation, were microinjected in groups of about 10 into 640 nontransgenic rabbit embryos between the two-cell and expanded blastocyst stages. Injections were made with primary isolations of gonial cells, within 48 hr of their collection. The injected embryos were transferred, with or without non-injected control embryos, into 49 recipient rabbits. Tissues from 159 resulting fetuses, implantation sites, and a few liveborn young were examined by PCR analysis for the two transgenes used (α-antitrypsin or luciferase). The overall pregnancy rate (about 80%) was not affected by the stage of development of the embryo injected, nor by co-transfer of control embryos. The survival rate of injected embryos (18% overall, 23.6% in pregnant recipients) was almost identical to that of 243 control embryos. Chimerism was detectable in tissues produced from 4 of 159 (2.5%) of the injected embryos, all four of which had been injected at the 8 to 16-cell stage. This low rate of success indicates that, although passage of rabbit gonial cells is not an absolute requirement for pluripotency, further investigation should pay particular attention to improving culture conditions with a view to deriving EG cell lines. © 1996 Wiley-Liss, Inc.  相似文献

Growth inhibition of Lactococcus lactis provoked by increasing osmolarity is reversed when glycine betaine (GB) or its analogs are added to a defined medium. Lacticin 481 production increased sharply with growth medium osmolarity in the absence of osmoprotectant but remained unaffected when GB was supplied in media of increasing osmolarity.  相似文献

We report here the first case of queen dimorphism in a ponerine ant species. A total of 550 colonies of the Neotropical ponerine ant Ectatomma ruidum, from two natural populations in southeastern Mexico, were investigated for polymorphism and reproductive function within the queen caste. The distribution of different morphological traits (head and thorax widths, and scutum and alitrunk lengths) was shown to be bimodal. Thorax width and head width were significantly isometric for both macro- and microgynes and thorax proportions did not change with the category of the queen. Microgynes appear as an isometric reduction of the normal queens. On average, macrogynes were found to be approximately 20% larger in size and twice as heavy as microgynes (and up to three times heavier in terms of dry weight). Their wing surface was 72% greater. There was no difference between the two morphs in terms of their capacity to be inseminated and all dealate females present in natural colonies could be considered as true potentially reproductive queens whatever the class to which they belonged. Ovary size, number of ovarioles per ovary, and number of mature oocytes produced were significantly higher for macrogynes than for microgynes, but small queens were able to function as active egg-layers. However, the number of egg-laying individuals was significantly lower for this form, although egg-laying activity appeared to be independent of queen weight. Despite its markedly reduced reproductive capacity compared to macrogynes, the distribution of the microgyne form of E. ruidum in both studied populations was very broad, reaching one-third of all alate and dealate females and extending to one-third of all colonies. In 72.2% of the colonies where alate and/or dealate microgynes were present, both forms co-occurred. Fat content analysis results supported the idea that semi-claustral independent colony founding, typical for this species, would essentially be achieved by macrogynes, while microgynes would be adopted by established colonies. We argue that queen dimorphism in E. ruidum could represent an attractive alternative dispersal strategy for female sexuals. The small females may constitute an important reserve of potential reproductives at a very low energetic cost to the colony, the production of a macrogyne being about 7.5 times more costly than a microgyne. The significantly greater `wing surface/body weight' ratio of microgynes also suggests their greater capacity for dispersion. The combination of such a capacity for microgynes, along with their ability to contribute in the production of both female morphs, including the reproductively efficient macrogynes, would contribute to insuring genetic reassortment at the population level and could explain, in part, the ecological success of E. ruidum in Neotropical zones. In the central sector of the Main Ethiopian Rift, the Ziway–Shala lake basin system includes four present-day residual lakes, from north to south, lakes Ziway, Langano, Abijata, and Shala. This region of East Africa is under the influence of the Intertropical Convergence Zone seasonal migration. Thus it has been designated as a potential core site by the ERICA Project (‘Environmental Research for Intertropical Climate in Africa'). The four lakes have been subjected to strong changes in water level and water salinity at least during the Late Pleistocene. The purpose of this study is to produce a model of basin formation and sediment accumulation for this system of lakes, in order to separate the effects of climatic change from environmental variations induced by local or regional factors such as volcano-tectonic forcings. In addition to an exhaustive synthesis of available data, various investigations have been used to develop this model: 3D remote sensing, high-resolution seismics, coring, and structural, sedimentological, and hydrological field studies. New AMS radiocarbon dating helped to refine the pre-existing stratigraphic framework for this region, and basin age estimations were calculated using mean sediment accumulation rates. The history of the Ziway–Shala lake basin system has been reconstructed from the Late Pliocene–Early Pleistocene period (106 yr), mainly characterized by catastrophic explosive volcanic eruptions. The early-middle Pleistocene–Late Pleistocene period (104–106 yr) was marked by a regional volcano-tectonic paroxysm, resulting in major changes in the morphology of the area, with the formation of the Abijata, Ziway and Shala lake basins. From 0.20 Ma, the Ziway–Shala basin history is marked by the eastward migration of volcano-tectonic activity, resulting in the development of the youngest basin of the Ziway–Shala system, the Langano Basin. The joint history of sedimentation in the Ziway, Langano, Abijata, and Shala lake basins started during the early-Late Pleistocene period (101–104 yr) and is characterized from this period up to the present-day by a series of climatically controlled rises and falls of lake level.  相似文献

The interrelationships between proteasomes and viral gene products are very complex. 20S proteasomes associate with a number of viral mRNAs which are cleaved by proteasome's associated endonuclease activity. In addition proteasome's endopeptidase activities are involved in the presentation of viral antigens. Viral proteins of different origin associate with the 20S and 26S complexes and interfere with their enzymatic activities. A major part of this review deals with the interactions between 20S proteasomes and the gene products of the human immunodeficiency virus (HIV) which has been studied in detail by our group.  相似文献

A bacterial strain (strain IFP 2173) was selected from a gasoline-polluted aquifer on the basis of its capacity to use 2,2,4-trimethylpentane (isooctane) as a sole carbon and energy source. This isolate, the first isolate with this capacity to be characterized, was identified by 16S ribosomal DNA analysis, and 100% sequence identity with a reference strain of Mycobacterium austroafricanum was found. Mycobacterium sp. strain IFP 2173 used an unusually wide spectrum of hydrocarbons as growth substrates, including n-alkanes and multimethyl-substituted isoalkanes with chains ranging from 5 to 16 carbon atoms long, as well as substituted monoaromatic hydrocarbons. It also attacked ethers, such as methyl t-butyl ether. During growth on gasoline, it degraded 86% of the substrate. Our results indicated that strain IFP 2173 was capable of degrading 3-methyl groups, possibly by a carboxylation and deacetylation mechanism. Evidence that it attacked the quaternary carbon atom structure by an as-yet-undefined mechanism during growth on 2,2,4-trimethylpentane and 2,2-dimethylpentane was also obtained.  相似文献

The consequences of feeding a protein-free (PF) diet, as compared to casein, on gut characteristics were studied in slightly energy-restricted rats fed similar amounts of feed for 10 d. The weight and pH of fresh digesta in the stomach were lower (P = 0.045 and P = 0.016). However, the weight of fresh digesta in the other segments and gut tissue weight were not significantly affected by the diet (P > 0.05). Small intestinal crypt depth, width and area were reduced by 13, 23 and 37%, respectively (P = 0.011, P = 0.004 and P = 0.001), and villus width tended to be smaller (P = 0.057), with the PF diet. Villus height to crypt depth ratio was also lower with the PF diet in the duodenum and ileum, respectively (P < 0.05). Finally, the specific activities of alkaline phosphatase and aminopeptidase N were reduced by 36 to 38% at different sites of the small intestine in the rats fed the PF diet (P < 0.05). In conclusion, chronic consumption of a protein-free diet altered the intestinal villus-crypt architecture and enzyme activities in rats.  相似文献

The wine bacterium Oenococcus oeni has to cope with harsh environmental conditions, including an acidic pH, a high alcoholic content, nonoptimal growth temperatures, and growth-inhibitory compounds such as fatty acids, phenolic acids, and tannins. We describe the characterization and cloning of the O. oeni ftsH gene, encoding a protease belonging to the ATP binding cassette protein superfamily. The O. oeni FtsH protein is closest in sequence similarity to the FtsH homologue of Lactococcus lactis. The O. oeni ftsH gene proved to be stress-responsive, since its expression increased at high temperatures or under osmotic shock. O. oeni FtsH protein function was tested in an Escherichia coli ftsH mutant strain, and consistent with the O. oeni ftsH gene expression pattern, the O. oeni FtsH protein provided protection for the E. coli ftsH mutant against heat shock. O. oeni and Bradyrhizobium japonicum FtsH proteins also triggered E. coli resistance to wine toxicity. Genes homologous to O. oeni ftsH were detected in many other lactic acid bacteria found in wine, suggesting that this type of gene constitutes a well-conserved stress-protective molecular device.  相似文献
