Wood ants are a dominant and ecologically important component of northern coniferous forests with interactions at many trophic levels. Each species exhibits specific habitat preferences which need to be understood if conservation measures are to be successful. In Britain, the rare narrow-headed ant Formica exsecta has disappeared from much of its former range and is now largely restricted to the highlands of Scotland where it is found in open canopy woodland and along forest edges. Nest locale of one small and vulnerable population at the edge of its current range, were compared with those of random locations within the same habitat and with actual nests within the stronghold of the Abernethy-Glenmore complex in Strathspey. After data exploration with Redundancy Analysis, stepwise multiple regression was used to create a model which best estimated the variance in nest location using a parsimonious selection of vegetation and environmental variables. The input variables included were light, soil moisture, altitude, tree stature and distribution, vegetation structure and composition, and ground characteristics. F. exsecta clearly exhibited preferences for the position of nest mounds in relation to light, vegetation and tree cover. Forest location was also important in determining which variables nests were affected by. This study highlights the importance of maintaining a dynamic mosaic of different-aged woodland enabling early successional species such as F. exsecta, with suitable areas to move to as conditions change and allowing the co-existence of all wood ant species.  相似文献

Ascidians are invertebrate chordates that form tadpole larvae with a surprisingly small number of cells. Recently, the emergence of powerful molecular tools to study cell fate determination in ascidians has been complemented by studies, often at cellular resolution, of morphogenetic processes. These studies point to a complex interplay among mechanisms that control cell fate and polarity and those that govern cell shape change and morphogenesis. The relative simplicity and stereotypy of ascidian development suggests that it will be possible to understand, and possibly to mathematically model, this dynamic coupling between cell fate and shape change.  相似文献

Corticosteroid-binding globulin (CBG) is a plasma glycoprotein that is primarily synthesized in the liver and binds cortisol and progesterone with high affinity. In this study, a CBG secreting hepatocellular carcinoma derived cell line (HepG2) was used to investigate the hormonal regulation of hepatic CBG synthesis. HepG2 cells were grown for 72 h in 30, 300 and 3000 nM concentrations of estradiol (E2), testosterone (T), insulin, thyroxin (T4) and dexamethasone (DMZ) and the secreted CBG quantified by a novel enzyme-linked immunosorbent assay (ELISA). Two-dimensional polyacrylamide gel electrophoresis (2D-PAGE) was carried out to determine the effects of these hormones on the relative distribution of CBG glycoforms.

Insulin, T4 and high concentrations of E2 decreased the secretion of CBG by HepG2 cells (p < 0.05). Ethanol, the solvent used for E2, T and DMZ, also significantly attenuated CBG secretion. 2D-PAGE resolved 13–14 glycoforms of CBG produced by HepG2 cells. Insulin caused a reduction in the synthesis of more acidic, while T4 and DMZ decreased the production of more basic CBG glycoforms. Stimulation with E2 resulted in the synthesis of additional isoforms of increased acidity, which may represent a type of CBG only seen during pregnancy in vivo. Possible physiological implications of these findings are discussed.  相似文献

Oxadiazole derivatives were synthesized and evaluated for their ability to inhibit tubulin polymerization and to cause mitotic arrest in tumor cells. The most potent compounds inhibited tubulin polymerization at concentrations below 1 microM. Lead analogs caused mitotic arrest of A431 human epidermoid cells and cells derived from multi-drug resistant tumors (10, EC(50)=7.8 nM). Competition for the colchicine binding site and pharmacokinetic properties of selected potent compounds were also investigated and are reported herein, along with structure-activity relationships for this novel series of antimitotic agents.  相似文献

Specifying synaptic partners and regulating synaptic numbers are at least partly activity-dependent processes during visual map formation in all systems investigated to date . In Drosophila, six photoreceptors that view the same point in visual space have to be sorted into synaptic modules called cartridges in order to form a visuotopically correct map . Synapse numbers per photoreceptor terminal and cartridge are both precisely regulated . However, it is unknown whether an activity-dependent mechanism or a genetically encoded developmental program regulates synapse numbers. We performed a large-scale quantitative ultrastructural analysis of photoreceptor synapses in mutants affecting the generation of electrical potentials (norpA, trp;trpl), neurotransmitter release (hdc, syt), vesicle endocytosis (synj), the trafficking of specific guidance molecules during photoreceptor targeting (sec15), a specific guidance receptor required for visual map formation (Dlar), and 57 other novel synaptic mutants affecting 43 genes. Remarkably, in all these mutants, individual photoreceptors form the correct number of synapses per presynaptic terminal independently of cartridge composition. Hence, our data show that each photoreceptor forms a precise and constant number of afferent synapses independently of neuronal activity and partner accuracy. Our data suggest cell-autonomous control of synapse numbers as part of a developmental program of activity-independent steps that lead to a "hard-wired" visual map in the fly brain.  相似文献

Phagocytic cells, such as neutrophils and macrophages, perform a critical role in protecting organisms from infection by engulfing and destroying invading microbes . Although some bacteria and fungi have evolved strategies to survive within a phagocyte after uptake, most of these pathogens must eventually kill the host cell if they are to escape and infect other tissues . However, we now demonstrate that the human fungal pathogen Cryptococcus neoformans is able to escape from within macrophages without killing the host cell by a novel expulsive mechanism. This process occurs in both murine J774 cells and primary human macrophages. It is extremely rapid and yet can occur many hours after phagocytosis of the pathogen. Expulsion occurs independently of the initial route of phagocytic uptake and does not require phagosome maturation . After the expulsive event, both the host macrophage and the expelled C. neoformans appear morphologically normal and continue to proliferate, suggesting that this process may represent an important mechanism by which pathogens are able to escape from phagocytic cells without triggering host cell death and thus inflammation .  相似文献

Living males of the 'ancient asexual' Darwinulidae (Ostracoda: Crustacea)   总被引:2,自引:0,他引:2  
Three living male darwinulid ostracods of a new species of the genus Vestalenula have been found in Yakushima, Japan. This is the first report of living darwinulid males for over 100 years and their morphology casts doubt on the two previous records from the late 1800' s. The presence of male darwinulids also calls into question the hypothesis that the family Darwinulidae is an exclusively ancient asexual group, reproducing without sex for over 200 million years (Myr). Male carapaces are of similar size and shape to A-1 juvenile females of the same species, suggesting that males may have been dismissed as A-1 juveniles in other living and fossil species. The antennae and fifth limbs are sexually dimorphic: the male antennae have six segments compared with five in the female and a series of putative chemical receptors originating at the extra segment boundary, while the male fifth limbs have well-developed grasping hooks, as in males of many ostracod groups. The lack of Zenker's Organ and of complex internal structures within the hemipenis contradicts previous hypotheses of the phylogenetic position of darwinulids.  相似文献

We compared the longevity of 29 strains representing 11 entomopathogenic nematode species in soil over 42 to 56 d. A series of five laboratory experiments were conducted with six to eight nematode strains in each and one or more nematode strains in common, so that qualitative comparisons could be made across experiments. Nematodes included Heterorhabditis bacteriophora (four strains), H. indica (Homl), H. marelatus (Point Reyes), H megidis (UK211), H. mexicana (MX4), Steinernema carpocapsae (eight strains), S. diaprepesi, S. feltiae (SN), S. glaseri (NJ43), S. rarum (17C&E), and S. riobrave (nine strains). Substantial within-species variation in longevity was observed in S. carpocapsae, with the Sal strain exhibiting the greatest survival. The Sal strain was used as a standard in all inter-species comparisons. In contrast, little intra-species variation was observed in S. riobrave. Overall, we estimated S. carpocapsae (Sal) and S. diaprepesi to have the highest survival capability. A second level of longevity was observed in H. bacteriophora (Lewiston), H. megidis, S. feltiae, and S. riobrave (3–3 and 355). Lower levels of survivability were observed in other H. bacteriophora strains (Hb, HP88, and Oswego), as well as S. glaseri and S. rarum. Relative to S. glaseri and S. rarum, a lower tier of longevity was observed in H. indica and H. marelatus, and in H. mexicana, respectively. Although nematode persistence can vary under differing soil biotic and abiotic conditions, baseline data on longevity such as those reported herein may be helpful when choosing the best match for a particular target pest.  相似文献
