dlk1/FA1 (delta-like 1/fetal antigen-1) is a member of the epidermal growth factor-like homeotic protein family whose expression is known to modulate the differentiation signals of mesenchymal and hematopoietic stem cells in bone marrow. We have demonstrated previously that Dlk1 can maintain the human bone marrow mesenchymal stem cells (hMSC) in an undifferentiated state. To identify the molecular mechanisms underlying these effects, we compared the basal gene expression pattern in Dlk1-overexpressing hMSC cells (hMSC-dlk1) versus control hMSC (negative for Dlk1 expression) by using Affymetrix HG-U133A microarrays. In response to Dlk1 expression, 128 genes were significantly up-regulated (with >2-fold; p < 0.001), and 24% of these genes were annotated as immune response-related factors, including pro-inflammatory cytokines, in addition to factors involved in the complement system, apoptosis, and cell adhesion. Also, addition of purified FA1 to hMSC up-regulated the same factors in a dose-dependent manner. As biological consequences of up-regulating these immune response-related factors, we showed that the inhibitory effects of dlk1 on osteoblast and adipocyte differentiation of hMSC are associated with Dlk1-induced cytokine expression. Furthermore, Dlk1 promoted B cell proliferation, synergized the immune response effects of the bacterial endotoxin lipopolysaccharide on hMSC, and led to marked transactivation of the NF-kappaB. Our data suggest a new role for Dlk1 in regulating the multiple biological functions of hMSC by influencing the composition of their microenvironment "niche." Our findings also demonstrate a role for Dlk1 in mediating the immune response.  相似文献

As equine musculoskeletal models become common, it is important to determine their sensitivity to the simplifications used. A subject-specific distal forelimb model was created using bones extracted from CT scans to examine movement from in-vivo invasive-marker motion capture. The movements of the sesamoid bones were simulated using the constraints of maintaining an isometric virtual ligament and maintaining contact between the appropriate articular surfaces, creating a variable moment arm for the tendons. The simulation of the proximal sesamoid bones was compared to movement recorded in-vitro. The paths and origins used for the deep digital flexor tendon (DDFT), superficial digital flexor tendon (SDFT) and suspensory ligament (SL) were altered and the effects on their calculated strains during trot stance were examined. The most influential alteration tested was the dorsopalmar changes to the tendon paths at the level of the proximal sesamoid bones, which led to a maximum length reduction of 4 and 2 mm for the SDFT and DDFT, respectively. Alterations to the virtual origins of the SDFT and DDFT were not influential leading to up to a 0.01% effect on strains for a 1cm dorsopalmar shift. In the SL, the choice of the proximal or distal edge of the proximal attachment site varied the strains calculated by up to 1% (3 mm). These results show that within the anatomically realistic spectrum, changes to tendon paths can have an appreciable effect on calculated strains; however the origin sites chosen are not as influential as changes to paths at the metacarpo-phalangeal joint.  相似文献

Aneuploidy is frequent in cancers. Recently it was found that pituitary tumor transforming gene (PTTG; also called Pds1p or securin) is overexpressed in many different tumors. Human T-cell leukemia virus type 1 (HTLV-1) is a retrovirus that primarily infects CD4+ T lymphocytes and causes adult T-cell leukemia. Here, we report that overexpression of human PTTG cooperated with the HTLV-I Tax oncoprotein in cellular transformation. Coexpression of Tax and PTTG enhanced chromosomal instability and neoplastic changes to levels greater than overexpression of either factor singularly. Cells that overexpressed both PTTG and Tax induced tumors more robustly in nude mice than cells that expressed either PTTG alone or Tax alone.  相似文献

Background

Understanding the role of seascape in shaping genetic and demographic population structure is highly challenging for marine pelagic species such as cetaceans for which there is generally little evidence of what could effectively restrict their dispersal. In the present work, we applied a combination of recent individual-based landscape genetic approaches to investigate the population genetic structure of a highly mobile extensive range cetacean, the harbour porpoise in the eastern North Atlantic, with regards to oceanographic characteristics that could constrain its dispersal.

Results

Analyses of 10 microsatellite loci for 752 individuals revealed that most of the sampled range in the eastern North Atlantic behaves as a 'continuous' population that widely extends over thousands of kilometres with significant isolation by distance (IBD). However, strong barriers to gene flow were detected in the south-eastern part of the range. These barriers coincided with profound changes in environmental characteristics and isolated, on a relatively small scale, porpoises from Iberian waters and on a larger scale porpoises from the Black Sea.

Conclusion

The presence of these barriers to gene flow that coincide with profound changes in oceanographic features, together with the spatial variation in IBD strength, provide for the first time strong evidence that physical processes have a major impact on the demographic and genetic structure of a cetacean. This genetic pattern further suggests habitat-related fragmentation of the porpoise range that is likely to intensify with predicted surface ocean warming.  相似文献

The Western capercaillie (Tetrao urogallus) is a keystone species of Palearctic boreal and altitude coniferous forests. With the increase of mountain leisure activities and habitat loss, populations are declining in most mountain ranges in Western Europe. Recent work has shown that the populations from the Pyrenees and Cantabrian Mountains survived a severe bottleneck during the 19th century, and are still considered as threatened due to habitat fragmentation and isolation with other populations. We present an extensive phylogeographic study based on mitochondrial DNA sequence (control region) extracted non-invasively from faeces collected throughout the species range (from western European mountains to central and eastern Europe, Fenno-Scandia, Russia and Siberia). We also compared our results with DNA sequences of closely related black-billed capercaillie (T. parvirostris). We found that populations from Pyrenees and Cantabrians are closely related but are different from all other capercaillie populations that form a homogenous clade. Therefore, we consider that these South-Western populations should be considered as forming an Evolutionary Significant Unit that needs an appropriate management at a local scale. We also discuss the possible locations of glacial refugia and subsequent colonisation routes in Eurasia, with a Western “aquitanus” lineage from Iberia and Balkans, and an Eastern “urogallus” lineage from Southern Asia. This work might have important implication for capercaillie conservation strategies to define important areas for conservation, and to prevent possible exchange or introductions of individuals originated from other lineages. This study aimed to characterize the bacterium-destroying properties of a gliding arc plasma device during electric discharges and also under temporal postdischarge conditions (i.e., when the discharge was switched off). This phenomenon was reported for the first time in the literature in the case of the plasma destruction of microorganisms. When cells of a model bacterium, Hafnia alvei, were exposed to electric discharges, followed or not followed by temporal postdischarges, the survival curves exhibited a shoulder and then log-linear decay. These destruction kinetics were modeled using GinaFiT, a freeware tool to assess microbial survival curves, and adjustment parameters were determined. The efficiency of postdischarge treatments was clearly affected by the discharge time (t*); both the shoulder length and the inactivation rate k(max) were linearly modified as a function of t*. Nevertheless, all conditions tested (t* ranging from 2 to 5 min) made it possible to achieve an abatement of at least 7 decimal logarithm units. Postdischarge treatment was also efficient against bacteria not subjected to direct discharge, and the disinfecting properties of "plasma-activated water" were dependent on the treatment time for the solution. Water treated with plasma for 2 min achieved a 3.7-decimal-logarithm-unit reduction in 20 min after application to cells, and abatement greater than 7 decimal logarithm units resulted from the same contact time with water activated with plasma for 10 min. These disinfecting properties were maintained during storage of activated water for 30 min. After that, they declined as the storage time increased.  相似文献

The ability to adjust reproductive output to environmental conditions is important to the fitness of a species. The semelparous, chordate, Oikopleura dioica, is particularly adept in producing a highly variable number of oocytes in its short life cycle. Here we show that this entails an original reproductive strategy in which the entire female germline is contained in a single multinucleate cell, the "coenocyst". After an initial phase of syncytial nuclear proliferation half of the nuclei entered meiosis whereas the other half became highly polyploid. The inner F-actin network, with associated plasma membranes, formed a highly ramified infrastructure in which each meiotic nucleus was contained in a pseudo-compartmentalized pro-oocyte linked to the common cytoplasm via ring canals. At a set developmental time, a subset of the pro-oocytes was selected for synchronous growth and the common coenocyst cytoplasm was equally partitioned by transfer through the ring canals. Examination of related species indicated that the coenocyst arrangement is a conserved feature of Appendicularian oogenesis allowing efficient numerical adjustment of oocyte production. As Appendicularia are the second most abundant class of zooplankton, with a world-wide distribution, the coenocyst is clearly a common and successful reproductive strategy on a global scale.  相似文献

Conformational changes of human plasma apolipoprotein B100 (apoB) during oxidative modification of low-density lipoproteins (LDL) have been investigated. Emphasis has been put on the early stages of LDL oxidation and the modification of apoB. We have applied two different modes of LDL oxidation initiation in order to approach the problem from different perspectives. To study conformational changes of the protein and the phospholipids surface monolayer, we have applied attenuated total reflection infrared as well as fluorescence spectroscopy. We have found for the first time that conformational changes of apoB occur even in the earliest stages of oxidation process and that those are located predominantly in the beta-sheet regions. The dynamics of changes has also been described and related to different stages of oxidation. After initial increase in particle surface accessibility and mobility, by entering into the propagation phase of oxidation process, LDL surface accessibility and mobility are decreased. Finally, in the decomposition phase of LDL oxidation, as the particle faces large chemical and physical changes, surface mobility and accessibility is increased again. These observations provide new insights into the modifications of LDL particles upon oxidation.  相似文献
