The role of IL-12, IL-23 and IFN-gamma in immunity to viruses   总被引:2,自引:0,他引:2  
IL-12, IL-23 and IFN-gamma form a loop and have been thought to play a crucial role against infectious viruses, which are the prototype of "intracellular" pathogens. In the last 10 years, the generation of knock-out (KO) mice for genes that control IL-12/IL-23-dependent IFN-gamma-dependent mediated immunity (STAT1, IFN-gammaR1, IFNgammaR2, IL-12p40 and IL-12Rbeta1) and the identification of patients with spontaneous germline mutations in these genes has led to a re-examination of the role of these cytokines in anti-viral immunity. We here review viral infections in mice and humans with genetic defects in the IL-12/IL-23-IFN-gamma axis. A comparison of the phenotypes observed in KO mice and deficient patients suggests that the human IL-12/IL-23-IFN-gamma axis plays a redundant role in immunity to most viruses, whereas its mouse counterparts play a more important role against several viruses.  相似文献

Purpose

To examine the frequency and potential of dose-volume predictors for chest wall (CW) toxicity (pain and/or rib fracture) for patients receiving lung stereotactic body radiotherapy (SBRT) using treatment planning methods to minimize CW dose and a risk-adapted fractionation scheme.

Methods

We reviewed data from 72 treatment plans, from 69 lung SBRT patients with at least one year of follow-up or CW toxicity, who were treated at our center between 2010 and 2013. Treatment plans were optimized to reduce CW dose and patients received a risk-adapted fractionation of 18 Gy×3 fractions (54 Gy total) if the CW V30 was less than 30 mL or 10–12 Gy×5 fractions (50–60 Gy total) otherwise. The association between CW toxicity and patient characteristics, treatment parameters and dose metrics, including biologically equivalent dose, were analyzed using logistic regression.

Results

With a median follow-up of 20 months, 6 (8.3%) patients developed CW pain including three (4.2%) grade 1, two (2.8%) grade 2 and one (1.4%) grade 3. Five (6.9%) patients developed rib fractures, one of which was symptomatic. No significant associations between CW toxicity and patient and dosimetric variables were identified on univariate nor multivariate analysis.

Conclusions

Optimization of treatment plans to reduce CW dose and a risk-adapted fractionation strategy of three or five fractions based on the CW V30 resulted in a low incidence of CW toxicity. Under these conditions, none of the patient characteristics or dose metrics we examined appeared to be predictive of CW pain.  相似文献

Quantifying and mapping ecosystem services (ES) is seen as one way to improve decision making and land management to better integrate environmental issues. This study aimed to characterize ES supply in deforestation context where an improvement of scientific knowledge should help develop more efficient environmental management. For three case studies in the Brazilian Amazon impacted by deforestation, seven indicators of potential ES supply were mapped at a spatial resolution of 30 × 30 m: biodiversity index (indicator of food web support); richness of pollinators (pollination); index of soil chemical quality (support to production); water available for plants (water regulation); soil carbon stocks (support to production and climate regulation); rate of water infiltration into the soil (soil erosion control); and vegetation carbon stocks (climate regulation). To map these indicators, in situ measurements of ES for 135 sampling points and remote sensing data were linked using regression methods. These methods were used to predict ES values and identify environmental factors that influence ES supply. The resulting maps help in understanding the influence of environmental factors on ES spatial distribution within the sites. The analyses illustrate the influence of land-use changes on ES supply and the role of context effects due to the heterogeneity of the biophysical environment, the temporality of deforestation and/or their diversified sociopolitical contexts. From a methodological viewpoint, the study highlights the importance of choices inherent in all cartographic practices and that need to be considered, especially in the context of rendering ES maps operational.  相似文献

During assembly of the bacterial flagellum, protein subunits that form the exterior structures are exported through a specialized secretion apparatus energized by the proton gradient. This category of protein transport, together with the similar process that occurs in the injectisomes of gram‐negative pathogens, is termed type‐III secretion. The membrane‐embedded part of the flagellar export apparatus contains five essential proteins: FlhA, FlhB, FliP, FliQ and FliR. Here, we have undertaken a variety of experiments that together support the proposal that the protein‐conducting conduit is formed primarily, and possibly entirely, by FliP. Chemical modification experiments demonstrate that positions near the center of certain FliP trans‐membrane (TM) segments are accessible to polar reagents. FliP expression sensitizes cells to a number of chemical agents, and mutations at predicted channel‐facing positions modulate this effect. Multiple assays are used to show that FliP suffices to form a channel that can conduct a variety of medium‐sized, polar molecules. Conductance properties are strongly modulated by mutations in a methionine‐rich loop that is predicted to lie at the inner mouth of the channel, which might form a gasket around cargo molecules undergoing export. The results are discussed in the framework of an hypothesis for the architecture and action of the cargo‐conducting part of the type‐III secretion apparatus.  相似文献

Landscape diversity slows the spread of an invasive forest pest species   总被引:1,自引:0,他引:1  
According to the associational resistance hypothesis, diverse habitats provide better resistance to biological invasions than monocultures. Host‐plant abundance has been shown to affect the range expansion of invasive pests, but the effect of landscape diversity (i.e. density of host/non‐host patches and diversity of forest habitat patches) on invasions remains largely untested. We used boundary displacement models and boosted regression tree analyses to investigate the effects of landscape diversity on the invasion of Corsica by the maritime pine bast scale Matsucoccus feytaudi over an 18‐yr period. Taking the passive wind dispersal of the scale into account, we showed that open habitats and connectivity between host patches accelerated spread by up to 13%, whereas landscapes with high tree diversity and a high density of non‐host trees decreased scale spread by up to 14%. We suggest a new mechanism for such associational resistance to pest invasion at the landscape level, which we term ‘the pitfall effect’.  相似文献

Infectious diseases have been paramount among the threats to health and survival throughout human evolutionary history. Natural selection is therefore expected to act strongly on host defense genes, particularly on innate immunity genes whose products mediate the direct interaction between the host and the microbial environment. In insects and mammals, the Toll-like receptors (TLRs) appear to play a major role in initiating innate immune responses against microbes. In humans, however, it has been speculated that the set of TLRs could be redundant for protective immunity. We investigated how natural selection has acted upon human TLRs, as an approach to assess their level of biological redundancy. We sequenced the ten human TLRs in a panel of 158 individuals from various populations worldwide and found that the intracellular TLRs—activated by nucleic acids and particularly specialized in viral recognition—have evolved under strong purifying selection, indicating their essential non-redundant role in host survival. Conversely, the selective constraints on the TLRs expressed on the cell surface—activated by compounds other than nucleic acids—have been much more relaxed, with higher rates of damaging nonsynonymous and stop mutations tolerated, suggesting their higher redundancy. Finally, we tested whether TLRs have experienced spatially-varying selection in human populations and found that the region encompassing TLR10-TLR1-TLR6 has been the target of recent positive selection among non-Africans. Our findings indicate that the different TLRs differ in their immunological redundancy, reflecting their distinct contributions to host defense. The insights gained in this study foster new hypotheses to be tested in clinical and epidemiological genetics of infectious disease.  相似文献

AIMS: Conflicting results exist regarding bone mineral density (BMD), metabolism and reproductive function of adult patients with congenital adrenal hyperplasia (CAH). We evaluated the long-term outcome and the impact of chronic glucocorticoid replacement in these patients. METHODS: Physical characteristics, serum hormone concentrations, BMD and metabolism were studied in 45 consecutive CAH adult patients. RESULTS: Among the 36 women, only 14 (39%) had regular menses. Among the 27 women with classical CAH, the mean number of surgical reconstructions of virilized genitalia was 2.1 +/- 0.2. Twenty of them (74%) were sexually active. Three men presented with testicular adrenal rest tumors. Twenty-five patients (55%) had decreased BMD at the femoral neck and/or at the lumbar spine. BMI was correlated with the BMD T-score at the femoral neck (p < 0.001) and at the lumbar spine (p < 0.01). Hydrocortisone dose was negatively correlated with the BMD T-score at the femoral neck (p = 0.04). Subjects with osteopenia had a significantly lower BMI and received higher hydrocortisone dose than those with normal BMD. Overweight was found in 21 patients (47%). There was a significantly positive correlation between HOMA and BMI (p < 0.001), and between HOMA and 17-OHP levels (p = 0.016). CONCLUSIONS: Adult patients with CAH treated with long-term glucocorticoids are at risk for decreased BMD, increased BMI, and disturbed reproductive function.  相似文献
