Human genes governing innate immunity provide a valuable tool for the study of the selective pressure imposed by microorganisms on host genomes. A comprehensive, genome-wide study of how selective constraints and adaptations have driven the evolution of innate immunity genes is missing. Using full-genome sequence variation from the 1000 Genomes Project, we first show that innate immunity genes have globally evolved under stronger purifying selection than the remainder of protein-coding genes. We identify a gene set under the strongest selective constraints, mutations in which are likely to predispose individuals to life-threatening disease, as illustrated by STAT1 and TRAF3. We then evaluate the occurrence of local adaptation and detect 57 high-scoring signals of positive selection at innate immunity genes, variation in which has been associated with susceptibility to common infectious or autoimmune diseases. Furthermore, we show that most adaptations targeting coding variation have occurred in the last 6,000–13,000 years, the period at which populations shifted from hunting and gathering to farming. Finally, we show that innate immunity genes present higher Neandertal introgression than the remainder of the coding genome. Notably, among the genes presenting the highest Neandertal ancestry, we find the TLR6-TLR1-TLR10 cluster, which also contains functional adaptive variation in Europeans. This study identifies highly constrained genes that fulfill essential, non-redundant functions in host survival and reveals others that are more permissive to change—containing variation acquired from archaic hominins or adaptive variants in specific populations—improving our understanding of the relative biological importance of innate immunity pathways in natural conditions.  相似文献

Assembly of plant Shaker subunits as heterotetramers, increasing channel functional diversity, has been reported. Here we focus on a new interaction, between AKT2 and KAT2 subunits. The assembly as AKT2/KAT2 heterotetramers is demonstrated by (i) a strong signal in two-hybrid tests with intracytoplasmic C-terminal regions, (ii) the effect of KAT2 on AKT2 subunit targeting in tobacco cells, (iii) the complete inhibition of AKT2 currents by co-expression with a dominant-negative KAT2 subunit in Xenopus oocytes, and reciprocally, and (iv) the appearance, upon co-expression of wild-type AKT2 and KAT2 subunits, of new channel functional properties that cannot be explained by the co-existence of two kinds of homotetrameric channels. In particular, the instantaneous current, characteristic of AKT2, displayed new functional features when compared with those of AKT2 homotetramers: activation by external acidification (instead of inhibition) and weak inhibition by calcium. Single channel current measurements in oocytes co-expressing AKT2 and KAT2 revealed a strong preference for incorporation of subunits into heteromultimers and a diversity of individual channels. In planta, these new channels, which may undergo specific regulations, are likely to be formed in guard cells and in the phloem, where they could participate in the control of membrane potential and potassium fluxes.  相似文献

Beticolin 3 (B3) belongs to a family of nonpeptidic phytotoxins produced by the fungus Cercospora beticola, which present a broad spectrum of cytotoxic effects. We report here that, at cytotoxic concentration (10 microM), B3 formed voltage-independent, weakly selective ion channels with multiple conductance levels in planar lipid bilayers. In symmetrical standard solutions, conductance values of the first levels were, respectively, 16 +/- 1 pS, 32 +/- 2 pS, and 57 +/- 2 pS (n = 4) and so on, any conductance level being roughly twice the lower one. Whether a cluster organization of elementary channels or different channel structures underlies this particular property was addressed by investigating the ionic selectivity and the pore size corresponding to the first three conductance levels. Both selectivity and pore size were found to be almost independent of the conductance level. This indicated that multiple conductance behavior resulted from a cluster organization of "B3 elementary channels." According to the estimated pore size and analyses of x-ray diffraction of B3 microcrystals, a structural model for "B3 elementary channels" is proposed. The ability to form channels is likely to be involved in the biological activity of beticolins.  相似文献

Laboratory microcosms were used to determine how leaf degradation processes (C–CO2 release and leaf mass loss) are affected by the diversity of soil macro-invertebrate assemblages (comprising earthworms, woodlice and millipedes). The relationships were tested using different measures of diversity (number of species and trait dissimilarities). Net diversity effects and transgressive overyielding were calculated to disentangle the relative impacts of negative, neutral and positive species interactions on the relationships observed.Minimum trait dissimilarities were found to be correlated with leaf mass loss and, to a lesser extent, with C–CO2 release. This suggests that process rates increase as trait similarity in species assemblage decreases. Conversely, increasing the number of species resulted in the saturation of process rates after 1–2 species had been added to a microcosm. The process rates observed could mainly be explained by positive species interactions (40–50% of microcosms being process-dependent), followed by neutral interactions (20–40%) and selection effects or negative interactions (less than 20%). These results thus suggest that the effects of a soil macro-invertebrate assemblage on soil processes are more likely to be driven by complementarity or facilitation than by competition.  相似文献

Potassium translocation in plants is accomplished by a large variety of transport systems. Most of the available molecular information on these proteins concerns voltage-gated potassium channels (Kv channels). The Arabidopsis genome comprises nine genes encoding α-subunits of Kv channels. Based on knowledge of their animal homologues, and on biochemical investigations, it is broadly admitted that four such polypeptides must assemble to yield a functional Kv channel. The intrinsic functional properties of Kv channel α-subunits have been described by expressing them in suitable heterologous contexts where homo-tetrameric channels could be characterized. However, due to the high similarity of both the polypeptidic sequence and the structural scheme of Kv channel α-subunits, formation of heteromeric Kv channels by at least two types of α-subunits is conceivable. Several examples of such heteromeric plant Kv channels have been studied in heterologous expression systems and evidence that heteromerization actually occurs in planta has now been published. It is therefore challenging to uncover the physiological role of this heteromerization. Fine tuning of Kv channels by heteromerisation could be relevant not only to potassium transport but also to electrical signaling within the plant.Key words: heteromerization, voltage-gated channels, membrane potential  相似文献

We investigated roosting and hunting site fidelity of Daubenton's bats Myotis daubentonii (Kuhl, 1817) in the Forêt de Soignes, an old-stand forest dominated by 150–200-year-old beeches, during the summers of 2003 and 2004. Roosting behaviour and hunting activity over ponds of adult females and juveniles were monitored using radio-telemetry. Eighteen roosts were located, all in natural cavities. The bats occupied a limited number of trees located in a specific and small roosting area. This roost aggregation was not linked to the distribution of hollow trees. Furthermore, whereas all 11 ponds in the study area were exploited by Daubenton's bats, monitored individuals were limited to two ponds to which they showed high fidelity. These two ponds were not the closest to the roosting area. Overall, these results show that at least for the time we conducted our experiment, female and juvenile Daubenton's bats are highly faithful to specific roosting and hunting grounds.  相似文献

An inward Shaker K(+) channel identified in Zea mays (maize), ZmK2.1, displays strong regulation by external K(+) when expressed in Xenopus laevis (African clawed frog) oocytes or COS cells. ZmK2.1 is specifically activated by K(+) with an apparent K(m) close to 15 mM independent of the membrane hyperpolarization level. In the absence of K(+), ZmK2.1 appears to enter a nonconducting state. Thus, whatever the membrane potential, this maize channel cannot mediate K(+) influx in the submillimolar concentration range, unlike its relatives in Arabidopsis thaliana. Its expression is restricted to the shoots, the strongest signal (RT-PCR) being associated with vascular/bundle sheath strands. Based on sequence and gene structure, the closest relatives of ZmK2.1 in Arabidopsis are K(+) Arabidopsis Transporter 1 (KAT1) (expressed in guard cells) and KAT2 (expressed in guard cells and leaf phloem). Patch-clamp analyses of guard cell protoplasts reveal a higher functional diversity of K(+) channels in maize than in Arabidopsis. Channels endowed with regulation by external K(+) similar to that of ZmK2.1 (channel activity regulated by external K(+) with a K(m) close to 15 mM, regulation independent of external Ca(2+)) constitute a major component of the maize guard cell inward K(+) channel population. The presence of such channels in maize might reflect physiological traits of C4 and/or monocotyledonous plants.  相似文献
