  • 1 Although the successful management of the Colorado potato beetle Leptinotarsa decemlineata (Say) depends on the prevention of its dispersal, its walking pattern in the landscape remains poorly understood. In the present study, post‐diapause, early summer, late summer and colony adult beetles, both fed and unfed before release, were tracked with a harmonic radar to establish their walking movement pattern in a bare‐ground field.
  • 2 The random walk model successfully described the dispersal of all beetle types, whether fed or unfed.
  • 3 The diverse life history of this species was manifested by an increased distance travelled and deviations of individual paths from the random model. Starved post‐diapause beetles travelled furthest and individual paths deviating from random were both local and directed, probably aiming to maximize opportunities for host colonization. Starved early summer beetles also travelled further than fed beetles but relied more on random movement to disperse in the habitat. Starving had little impact on the distance travelled or the path deviations of late summer beetles that are searching for overwintering site rather than hosts.
  • 4 The increased displacement of starving beetles over fed beetles corresponded with an increased walking step and index of straightness.
  • 5 The impact of starvation on travel distance was greater than expected from laboratory tests.
  • 6 In conclusion, the results obtained in the present study suggest a random walking pattern to search arable land until host volatile or visual impulses trigger a more directed walk or flight.

The Hedgehog signaling is a determinant pathway for tumor progression. However, while inhibition of the Hedgehog canonical pathway—Patched–Smoothened–Gli—has proved efficient in human tumors with activating mutations in this pathway, recent clinical data have failed to show any benefit in other cancers, even though Sonic Hedgehog (SHH) expression is detected in these cancers. Cell-adhesion molecule-related/down-regulated by Oncogenes (CDON), a positive regulator of skeletal muscle development, was recently identified as a receptor for SHH. We show here that CDON behaves as a SHH dependence receptor: it actively triggers apoptosis in the absence of SHH. The pro-apoptotic activity of unbound CDON requires a proteolytic cleavage in its intracellular domain, allowing the recruitment and activation of caspase-9. We show that by inducing apoptosis in settings of SHH limitation, CDON expression constrains tumor progression, and as such, decreased CDON expression observed in a large fraction of human colorectal cancer is associated in mice with intestinal tumor progression. Reciprocally, we propose that the SHH expression, detected in human cancers and previously considered as a mechanism for activation of the canonical pathway in an autocrine or paracrine manner, actually provides a selective tumor growth advantage by blocking CDON-induced apoptosis. In support of this notion, we present the preclinical demonstration that interference with the SHH–CDON interaction triggers a CDON-dependent apoptosis in vitro and tumor growth inhibition in vivo. The latter observation qualifies CDON as a relevant alternative target for anticancer therapy in SHH-expressing tumors.  相似文献

Microbial mannanases are biotechnologically important enzymes since they target the hydrolysis of hemicellulosic polysaccharides of softwood biomass into simple molecules like manno-oligosaccharides and mannose. In this study, we have implemented a strategy of molecular engineering in the yeast Yarrowia lipolytica to improve the specific activity of two fungal endo-mannanases, PaMan5A and PaMan26A, which belong to the glycoside hydrolase (GH) families GH5 and GH26, respectively. Following random mutagenesis and two steps of high-throughput enzymatic screening, we identified several PaMan5A and PaMan26A mutants that displayed improved kinetic constants for the hydrolysis of galactomannan. Examination of the three-dimensional structures of PaMan5A and PaMan26A revealed which of the mutated residues are potentially important for enzyme function. Among them, the PaMan5A-G311S single mutant, which displayed an impressive 8.2-fold increase in kcat/KM due to a significant decrease of KM, is located within the core of the enzyme. The PaMan5A-K139R/Y223H double mutant revealed modification of hydrolysis products probably in relation to an amino-acid substitution located nearby one of the positive subsites. The PaMan26A-P140L/D416G double mutant yielded a 30% increase in kcat/KM compared to the parental enzyme. It displayed a mutation in the linker region (P140L) that may confer more flexibility to the linker and another mutation (D416G) located at the entrance of the catalytic cleft that may promote the entrance of the substrate into the active site. Taken together, these results show that the directed evolution strategy implemented in this study was very pertinent since a straightforward round of random mutagenesis yielded significantly improved variants, in terms of catalytic efiiciency (kcat/KM).  相似文献

Female mate-choice copying is a social learning phenomenon whereby a female's observation of a successful sexual interaction between a male and another female increases her likelihood of subsequently preferring that male. Although mate-choice copying has been documented in several vertebrate species, to our knowledge it has not yet been investigated in insects. Here, we investigated whether female mate-choice copying occurs in the fruit fly Drosophila serrata, a model system for the study of mate preferences and the sexual selection they generate. We used two complementary experiments in which focal females were given a choice between two males that differed in either their apparent (as determined visually by the focal female) or actual recent mating success. Mate-choice copying was evaluated by testing whether focal females mated more frequently with the ‘preferred’ male as opposed to the other male. In both experiments, however, we found no evidence for mate-choice copying. We discuss possible reasons for the apparent absence of mate-choice copying in this species.  相似文献

We investigated spatial distribution of fishes and decapods in eelgrass (Zostera marina L.) and non-vegetated sandy habitats in Kouchibouguac Estuary, New Brunswick, Canada. During the ice-free season in 1999 and 2000, mobile fauna were sampled using fyke nets, minnow traps and an epibenthic sled. In general, fishes and decapods were more abundant and diverse (species richness) in eelgrass beds than in nearby sandy habitats, although the trend in abundance of decapods was not statistically significant. Abundance was greater at night than during daylight hours for most species. Atlantic tomcod (Microgadus tomcod), winter flounder (Pleuronectes americanus) and sand shrimp (Crangon septemspinosa) dominated night/crepuscular catches, whereas threespine stickleback (Gasterosteus aculeatus) and cunner (Tautogolabrus adspersus) predominated in diurnal periods. The nursery function of eelgrass habitat was most evident for juvenile white hake (Urophycis tenuis) and small cunners (<3 cm), which were found only in such habitat. This study is one of the few to have investigated the ecological importance of eelgrass habitats for mobile faunal communities in eastern Canada.  相似文献

The flight take-off activity of Colorado potato beetles, Leptinotarsa decemlineata (Say) (Coleoptera: Chrysomelidae), was significantly higher at a landscape-protected than at semiexposed and exposed sites in a 2-yr field study. In both years, mean daylight temperature, solar radiation, and relative humidity were generally similar at all sites, but wind speed was lower at the protected site than at the exposed sites. Results suggest that wind was the limiting abiotic factor for flight take-off at the exposed site. Caged beetles exposed to constant wind speeds of 3.4, 4.7, and 7.0 m/s showed a significant corresponding decrease in number of flight take-off. There was no cumulative effect of wind exposure on the readiness of the beetles to fly, suggesting that wind acts as a physical barrier to flight take-off. It should be possible to reduce Colorado potato beetle flight dispersal by selecting fields most exposed to wind over landscape-protected fields when rotating potato, Solanum tuberosum L., crops.  相似文献

In order to study the effect of phosphorylation on the function of the steroidogenic acute regulatory protein (StAR), 10 putative phosphorylation sites were mutated in the hamster StAR. In pcDNA3.1-StAR transfected COS-1 cells, decreases in basal activity were found for the mutants S55A, S185A and S194A. Substitution of S185 by D or E to mimic phosphorylation resulted in decreased activity for all mutants; we concluded that S185 was not a phosphorylation site and we hypothesized that mutations on S185 created StAR conformational changes resulting in a decrease in its binding affinity for cholesterol. In contrast, the mutation S194D resulted in an increase in StAR activity. We have calculated the relative rate of pregnenolone formation (App. Vmax) in transfected COS-1 cells with wild type (WT) and mutant StAR-pcDNA3.1 under control and (Bu)2-cAMP stimulation. The App. Vmax values refer to the rate of cholesterol transported and metabolized by the cytochrome P450scc enzyme present in the inner mitochondrial membrane. The App. Vmax was 1.61 ± 0.28 for control (Ctr) WT StAR and this value was significantly increased to 4.72 ± 0.09 for (Bu)2-cAMP stimulated preparations. App. Vmax of 5.53 (Ctr) and 4.82 ((Bu)2-cAMP) found for S194D StAR preparations were similar to that of the WT StAR stimulated preparations. At equal StAR quantity, an anti-phospho-(S/T) PKA substrate antibody revealed four times more phospho-(S/T) in (Bu)2-cAMP than in control preparations. The intensity of phosphorylated bands was decreased for the S55A, S56A and S194A mutants and it was completely abolished for the S55A/S56A/S194A mutant. StAR activity of control and stimulated preparations were diminished by 73 and 72% for the mutant S194A compared to 77 and 83% for the mutant S55A/S56A/S194A. The remaining activity appears to be independent of phosphorylation at PKA sites and could be due to the intrinsic activity of non-phosphorylated StAR or to an artefact due to the pharmacological quantity of StAR expressed in COS-1. In conclusion we have shown that (Bu)2-cAMP provokes an augmentation of both the quantity and activity of StAR, and that an enhancement in StAR phosphorylation increases its activity. The increased quantity of StAR upon (Bu)2-cAMP stimulation could be due to an augmentation of its mRNA or protein synthesis stability, or both; this is yet to be determined.  相似文献

A novel Arabidopsis thaliana gene (AtNADK-1) was identified based on its response to radiation and oxidative stress. Levels of AtNADK-1 mRNA increase eight-fold following exposure to ionising radiation and are enhanced three-fold by treatment with hydrogen peroxide. The gene also appears to be differentially regulated during compatible and incompatible plant-pathogen interactions in response to Pseudomonas syringae pv. tomato. The full-length AtNADK-1 cDNA encodes a 58-kDa protein that shows high sequence homology to the recently defined family of NAD(H) kinases. Recombinant AtNADK-1 utilises ATP to phosphorylate both NAD and NADH, showing a two-fold preference for NADH. Using reverse genetics, we demonstrate that AtNADK-1 deficient plants display enhanced sensitivity to gamma irradiation and to paraquat-induced oxidative stress. Our results indicate that this novel NAD(H) kinase may contribute to the maintenance of redox status in Arabidopsis thaliana.  相似文献
