Laboratory assays demonstrated the presence of a small positive geotaxis response to a 15° incline by Folsomia candida Willem (Collembola: Isotomidae). Negative phototaxis played an additive role to positive geotaxis when the experimental apparatus were exposed to light. The geotactic response was negatively affected by cold acclimation and decreasing surrounding temperature, but unaffected by food deprivation. The reduced mobility of springtails at low temperature did not seem to play a role in the corresponding decreased geotaxis. The low level of geotaxis and its further decrease with exposure to low temperature support an earlier suggestion that F. candida do not respond to cooling temperatures of fall by relocation to warmer deeper soil layers, but remain in the upper soil layers and increase their cold tolerance to continue foraging in the food‐rich upper soil layers. 相似文献

We studied the effect of sodium thiocyanate (NaSCN) in the determination of specific nuclear estrogen receptors from the hypothalamic-pituitary axis (HPA) of mice. We compared the results with those of the exchange assay using either suspensions of whole nuclei or KC1-nuclear extracts. Our findings demonstrated that 0.5 M NaSCN was more efficient than 0.5 M KC1 in extracting [3H] E2 nuclear content from HPA (91% vs 79.3%). Nuclear fractions extracted with 0.5 M NaSCN revealed the presence of a single class of low-capacity-high affinity binding sites that sedimented in the 4.0 S area. Nuclear binding was T° dependent and reached maximum levels of 450 ± 156 (S.E.) fmol/mg DNA after an overnight incubation at 4°C. Such levels were comparable to those observed in whole nuclei suspensions after 1 hour incubation at 37°C (618 ± 71 fmol/mg DNA, p > 0.05) but two-fold higher (p <0.01) than the concentration of binding sites measured in KC1-extracted nuclear fractions under similar experimental conditions. Plant-tissue-colonizing fungi fine-tune the deconstruction of plant-cell walls (PCW) using different sets of enzymes according to their lifestyle. However, some of these enzymes are conserved among fungi with dissimilar lifestyles. We identified genes from Glycoside Hydrolase family GH131 as commonly expressed during plant-tissue colonization by saprobic, pathogenic and symbiotic fungi. By searching all the publicly available genomes, we found that GH131-coding genes were widely distributed in the Dikarya subkingdom, except in Taphrinomycotina and Saccharomycotina, and in phytopathogenic Oomycetes, but neither other eukaryotes nor prokaryotes. The presence of GH131 in a species was correlated with its association with plants as symbiont, pathogen or saprobe. We propose that GH131-family expansions and horizontal-gene transfers contributed to this adaptation. We analysed the biochemical activities of GH131 enzymes whose genes were upregulated during plant-tissue colonization in a saprobe (Pycnoporus sanguineus), a plant symbiont (Laccaria bicolor) and three hemibiotrophic-plant pathogens (Colletotrichum higginsianum, C. graminicola, Zymoseptoria tritici). These enzymes were all active on substrates with β-1,4, β-1,3 and mixed β-1,4/1,3 glucosidic linkages. Combined with a cellobiohydrolase, GH131 enzymes enhanced cellulose degradation. We propose that secreted GH131 enzymes unlock the PCW barrier and allow further deconstruction by other enzymes during plant tissue colonization by symbionts, pathogens and saprobes. 相似文献

Design, synthesis and structure-activity relationships of benzimidazole derivatives as activators of the AMP-activated protein kinase (AMPK) are presented in this paper. AMPK is the central component of a protein kinase cascade that plays a key role in the regulation of energy balance. Once activated, AMPK initiates a series of responses that are aimed at restoring the energy balance of the cell and recent studies have indicated that AMPK plays an important role in regulation of the whole-body energy metabolism. Within populations, individual animals vary considerably in their behaviour, including mate choice and personality. There is mounting interest in the potential covariation between these two behaviours within individuals, such that personality would influence mate choice. We experimentally tested this proposition under controlled laboratory conditions using male Trinidadian guppies (Poecilia reticulata) as a model study system. We assayed repeatedly the mating preference of individual males for females based on their body size. Additionally, we assayed repeatedly two ecologically relevant personality traits in males, namely exploration of a novel environment and boldness under a simulated predation threat. Finally, we analysed whether male mating preference and personality traits were repeatable, and tested whether the personality of individual males was correlated (covaried) with their mating preference scores. Although all but one of the measures of exploration and boldness behaviour were repeatable over time, male mating preference scores were not repeatable. Measures of male exploration and boldness were not inter-correlated among individuals, suggesting the absence of a behavioural syndrome between exploration and boldness. Unexpectedly, males did not exhibit on average a significant mating preference for larger females over smaller ones; they chose randomly between the paired stimulus females. Overall, we found no compelling evidence for a relationship between individual personality traits and mating preference in male guppies, suggesting that personality does not predict mate choice, at least in our study population and under our experimental conditions. We discuss potential factors, other than male personality and body length, that might maintain inter-individual variation in male mating preferences in the guppy in the wild. 相似文献

Steroidogenic acute regulatory (StAR)—related lipid transfer proteins possess a START (steroidogenic acute regulatory-related lipid transfer) domain. START domains are conserved protein modules involved in the non-vesicular intracellular transport of lipids and cholesterol in mammals. Fifteen mammalian proteins, divided in five subfamilies, are reported to possess a START domain. Members of the STARD4 subfamily, i.e. STARD4, 5 and 6 are essentially single START domains and are thought to be involved in the intracellular transport of cholesterol. No structure of a cholesterol-bound START domain from this family has been resolved yet. The determination of the structure of such a complex would contribute to a better understanding of the mechanism of ligand binding and transport by START domains, two unresolved aspects of their structural biology. In this context, we have undertaken the structure determination of a ligand-bound form of STARD5 by NMR. The neotropical convict cichlid fish (Amatitlania siquia) is an important model species for the study of animal behaviour. It is therefore surprising that no genetic markers are
available for this species. Here, we have optimised four polymorphic microsatellite loci for use with this fish, where each
locus comprises between 9 and 28 alleles. We demonstrate how these markers can be used to genotype fry (young) and for the
analyses of genetic structure and prevalence of interfamilial mixing within biparental broods of wild convict cichlids. These
microsatellites will facilitate future research, hitherto not possible, on the molecular, behavioural and evolutionary ecology
