Soybean [Glycine max (L.) Merrill] plants that had been subjectedto 15 d of nitrogen deprivation were resupplied for 10 d with1.0 mol m3 nitrogen provided as NO3, NH4+, orNH4++NO3 in flowing hydroponic culture. Plants in a fourthhydroponic system received 1.0 mol m3 NO3 duringboth stress and resupply periods. Concentrations of solublecarbohydrates and organic acids in roots increased 210 and 370%,respectively, during stress. For the first day of resupply,however, specific uptake rates of nitrogen, determined by ionchromatography as depletion from solution, were lower for stressedthan for non-stressed plants by 43% for NO3- resupply, by 32%for NH4+ + NO3 resupply, and 86% for NH4+ resupply. Whenspecific uptake of nitrogen for stressed plants recovered torates for non-stressed plants at 6 to 8 d after nitrogen resupply,carbohydrates and organic acids in their roots had declinedto concentrations lower than those of non-stressed plants. Recoveryof nitrogen uptake capacity of roots thus does not appear tobe regulated simply by the content of soluble carbon compoundswithin roots. Solution concentrations of NH4+ and NO3 were monitoredat 62.5 min intervals during the first 3 d of resupply. Intermittenthourly intervals of net influx and net effluxoccurred. Rates of uptake during influx intervals were greaterfor the NH4+ -resupplied than for the NO3 -resuppliedplants. For NH4+ -resupplied plants, however, the hourly intervalsof efflux were more numerous than for NO3 -resuppliedplants. It thus is possible that, instead of repressing NH4+influx, increased accumulation of amino acids and NH4+ in NH4+-resupplled plants inhibited net uptake by stimulation of effluxof NH4+ absorbed in excess of availability of carbon skeletonsfor assimilation. Entry of NH4+ into root cytoplasm appearedto be less restricted than translocation of amino acids fromthe cytoplasm into the xylem. The nucleotide (nt) sequence encoding the ovine homologue of interleukin-8 (IL-8) was determined. The mRNA is 1494-nt long with an ORF of 101 codons. The long 3' non-coding element contains several ATTTA repeats implicated in the swift turnover of other chemokine mRNAs. The encoded protein of 11 kDa before processing, and 9 kDa as mature protein, contains the Cys-Xaa-Cys motif common to -chemokines, and has conserved amino acids (aa) at positions identified as receptor contact sites for IL-8. Identities with other published IL-8 aa sequences are: dog, 91%; pig, 87%; rabbit, 84%; human, 78%; guinea pig, 69%. A 49% aa identity is also found with a chicken embryo fibroblast protein. 相似文献

Abstract: Recent data have indicated that the long-lasting increase in tyrosine hydroxylase (TH) protein could be differently expressed in the anterior and posterior locus coeruleus (LC) after a single intraperitoneal injection of RU24722, which has been proposed as a potent activator of catecholaminergic systems. In the present study, we have evaluated the dose and time course responses and the effect of a repeated treatment with RU24722 at 3-day intervals on TH protein level in the anterior and posterior rat LC. The results showed that RU24722 induces a long-lasting increase of TH protein level in the anterior and posterior LC that was maximal 3 days following a single injection of 30 mg/kg. The increase in TH protein was maintained at a constant level after repeated administrations of RU24722 at 3-day intervals. Furthermore, we have investigated whether the effect of the drug on TH protein could be modulated via several hormonal systems. The long-term increase of TH steady-state content after RU24722 was still observed 15 days after castration, adrenalectomy, hypophysectomy, and thyroidectomy. The initial steady-state TH protein level was significantly higher in the anterior LC of thyroid- or hypophysectomized and in the posterior LC of hypophysectomized rats. Whereas large-sized species of oligotrichs, some of whichwere mixotrophic, dominated at the surface, haptorids were bestrepresented in deep waters. The effect of allelochemicals from its host, the larva of the cabbage root fly, Delia radicum (L.) (Diptera: Anthomyiidae), and the host's food plant on the ovipositor probing response of the parasitoid Trybliographa rapae (Westw.) (Hymenoptera: Cynipidae) were investigated. Trybliographa rapae probed both cabbage root fly infested and uninfested swede (Brassica napus var. napobrassica), although significantly more wasps responded to infested swede. Antennal sensilla are likely to be the mediators of this response. The synomones and kairomones involved are extractable in water, diethyl ether and methanol. No response was observed to washed, starved cabbage root fly larvae. Wasps spent significantly longer searching infested swede than uninfested, although probing frequency remained constant. It is suggested that the initiation of probing in T. rapae is dependent on a threshold concentration of general synomones or host related synomones and kairomones, whereas time spent searching a particular area is dependent on the environment perceived by sensilla on the ovipositor. 相似文献

We used partial sequences of three mitochondrial genes (12S rRNA, cytochrome b, and cytochrome c oxidase subunit I) to reconstruct the phylogeny of European seed beetles (Bruchidae) belonging to the genera Bruchus Linnaeus and Bruchidius Schilsky. Adult beetles examined in this study were obtained from larvae bred from seeds directly collected in the field. Parsimony, maximum likelihood, and Bayesian inference were used to infer phylogenetic relationships among species. Both genera, Bruchidius and Bruchus, formed monophyletic groups in all analyses. Our results were partially in discrepancy with existing taxonomic groups (Borowiec, 1987). Critical analysis of relationships among taxa, and exhaustive review of host-plants data highlight the very high level of specialization of these seed beetles. Phylogenetically related insects were associated with host-plants belonging to the same botanical tribes. 相似文献

Comparison of the extent of leaf senescence depending on the genetic background of different recombinant inbred lines (RILs) of Arabidopsis (Arabidopsis thaliana) is described. Five RILs of the Bay-0 x Shahdara population showing differential leaf senescence phenotypes (from early senescing to late senescing) were selected to determine metabolic markers to discriminate Arabidopsis lines on the basis of senescence-dependent changes in metabolism. The proportion of gamma-aminobutyric acid, leucine, isoleucine, aspartate, and glutamate correlated with (1) the age and (2) the senescence phenotype of the RILs. Differences were observed in the glycine/serine ratio even before any senescence symptoms could be detected in the rosettes. This could be used as predictive indicator for plant senescence behavior. Surprisingly, late-senescing lines appeared to mobilize glutamine, asparagine, and sulfate more efficiently than early-senescing lines. The physiological basis of the relationship between leaf senescence and flowering time was analyzed. 相似文献

Partial cDNA clones encoding approximately the carboxy terminal half of Pleurodeles fibronectin (FN) were isolated. They account for 4.7 Kbp of the 3' region of the FN mRNA. The cDNA nucleotide sequence comprises all three alternatively spliced segments designated EIIIA, EIIIB and V-segment, respectively. All three segments are included in FN mRNA synthesized during early embryogenesis whereas, from the tailbud stage onward the V-region was partially excluded. The isolation of Pleurodeles cDNA clones including the three different spliced EIIIA, EIIIB and V segment raises new possibilities for the study of the precise role of specific regions of FN in early amphibian development. 相似文献

The South African population faces many of the global concerns relating to micronutrient deficiency and the impact this has on health and well-being. Moreover, there is a high prevalence of HIV infection, compounded by a high level of co-infection with TB.This pilot study evaluates the impact of a fortified supplementary food on the health and well-being of a cohort of crèche children, aged 3 to 6, and adult TB patients drawn from the Presidential Node of Alexandra, Johannesburg, South Africa. A further aim of this study was to evaluate the sensitivity and validity of non-invasive indicators of nutritional status in a field-based population sample.The investigational product, e’Pap, is supported by extensive anecdotal evidence that whole grain cereals with food-style nutrients constitute an effective supplementary food for those suffering from the effects of food insecurity, poor health and well-being, and coping with TB and HIV infection.The results indicate a beneficial effect of e’Pap for both study populations, and particularly for adult TB patients, whose baseline data reflected severe food insecurity and malnutrition in the majority of cases. There is evidence to suggest statistically significant improvements in key micronutrient levels, well-being and energy, hand-grip strength, the Bioelectrical Impedance Analysis (BIA) Illness Marker, and certain clinical indicators. Although Body Mass Index (BMI) and Mid Upper Arm Circumference (MUAC) are frequently used as standard measures to evaluate the efficacy of nutritional interventions, these indicators were not sufficiently sensitive in this study. Nor does weight gain necessarily indicate improved nutritional status. Hand-grip strength, lean body mass, and the BIA Illness Marker seem to be more useful indicators of change in nutritional status. 相似文献

Retroviral integrases (INs) catalyse the integration of the reverse transcribed viral DNA into the host cell genome. This process is selective, and chromatin has been proposed to be a major factor regulating this step in the viral life cycle. However, the precise underlying mechanisms are still under investigation. We have developed a new in vitro integration assay using physiologically-relevant, reconstituted genomic acceptor chromatin and high-throughput determination of nucleosome positions and integration sites, in parallel. A quantitative analysis of the resulting data reveals a chromatin-dependent redistribution of the integration sites and establishes a link between integration sites and nucleosome positions. The co-activator LEDGF/p75 enhanced integration but did not modify the integration sites under these conditions. We also conducted an in cellulo genome-wide comparative study of nucleosome positions and human immunodeficiency virus type-1 (HIV-1) integration sites identified experimentally in vivo. These studies confirm a preferential integration in nucleosome-covered regions. Using a DNA mechanical energy model, we show that the physical properties of DNA probed by IN binding are important in determining IN selectivity. These novel in vitro and in vivo approaches confirm that IN has a preference for integration into a nucleosome, and suggest the existence of two levels of IN selectivity. The first depends on the physical properties of the target DNA and notably, the energy required to fit DNA into the IN catalytic pocket. The second depends on the DNA deformation associated with DNA wrapping around a nucleosome. Taken together, these results indicate that HIV-1 IN is a shape-readout DNA binding protein. 相似文献
