Antibodies against Lubrol-solubilized Electrophorus electroplax (Na+ + K+)-ATPase (ATP phosphohydrolase, EC 3.6.1.3) and its 96 000-dalton polypeptide (P96) were raised in rabbits. The P96 antibody does not cross react with the (Na+ + K+)-ATPase from mammalian species and tissues, but it cross reacts with the (Na+ + K+)-ATPase from both Electrophorus electroplax and brain. The combination of enzyme with anti-P96 is found to inhibit phosphoryl enzyme formation to the same extent that it inhibits enzyme activity. The rate of K+-sensitive dephosphorylation of phosphoryl enzyme appears to be unchanged. These are also found to be true with the antibody against the whole enzyme. Upon tryptic digestion of the enzyme-anti-P96 complex only the large polypeptide of the enzyme is protected. In the case of enzyme-anti-Lubrol-solubilized enzyme complex, both the large and small polypeptides are protected, whereas preimmune sera are without any protecting effect. The data indicate that the phosphoryl acceptor polypeptide and the Lubrol-solubilized electroplax (Na+ + K+)-ATPase from which the polypeptide is derived are phylogenetically distinct from those of the mammalian (Na+ + K+)-ATPases. The selective tryptic resistance of the enzyme-anti-P96 complex indicates that the two polypeptides are spatially well separated, possibly on opposite sides of the membrane.  相似文献

Modes of aggregation fo alanine-, norvaline- and valine-contaiing dpepetides with the general formula R1? C1O1? N2H2? CHR2? C2O2? N3H3? R3 have been studied in CCl4 solution by using infrared and nuclear magnetic resonance spectroscopies. Solutions of the pure L isomer and of the racemic mixture do not give identical data. At a given concentration, the racemic mixtrue is more aggregated than the pure enantiomer, and the difference, negligible in the case of alanine derivative, increases wiht the bulkiness of the side cahin R2. The results show that a selective interaction takes place between enantiomeric molecules, resulting ina dimer associating tow inverse configurated C5 conformers. The stabilizaion of this dimer proceeds from two symmetrical and intermolecular H3 … O1 hydrogen bondings.  相似文献

Treatment of 2,5-anhydro-1-bromo-1-deoxy-2,3-O-isopropylidene-1-p-nitrophenylhydrazono-d-ribose with methyl acetylenecarboxylate gave methyl 3-(2,3-O-isopropylidene-β-d-erythrofuranosyl)-1-p-nitrophenylpyrazole- (8) and 5-carboxylate (9). Amidification at C-5 of 8 was easier than at C-4 of 9. Similarly, dimethyl 3-(2,3-O-isopropylidene-β-d-erythrofuranosyl)- 1 - p-nitrophenylpyrazole-4,5-dicarboxylate gave specifically a 5-carbamoyl derivative, the structure of which was established by comparison of the 13C-n.m.r.spectrum with those of a series of glycosylpyrazoles. The correlation between the experimental values of the chemical shifts of the carbon atoms of the pyrazole ring and the values calculated by addition of the contributions of the various groups linked to the ring was better (R 0.98) than the correlations obtained by calculation by the CNDO/2 method of the total electron population (R 0.92) or of the π-electron population of each carbon atom (R 0.85).  相似文献

Three forms of ferredoxin FdI, FdI′, and FdII have been isolated from Desulfovibrio gigas, a sulfate reducer. They are separated by a combination of DEAE-cellulose and gel filtration chromatographic procedures. FdI and FdI′ present a slight difference in isoelectric point which enables the separation of the two forms over DEAE-cellulose, while FdII is easily separated from the two other forms by gel filtration. The three forms have the same amino acid composition and are isolated in different aggregation states. Molecular weight determinations by gel filtration gave values of 18 000 for FdI and FdI′ and 24 000 for FdII, whereas a value of 6000 is determined when dissociation is accomplished with sodium dodecyl sulfate. The electronic spectra are different and their ultraviolet-visible absorbance rations are 0.77, 0.87 and 0.68 respectively for FdI, FdI′ and FdII. Despite these differences, the physiological activities of the three forms are similar as far as the reduction of sulfite by molecular hydrogen is concerned.  相似文献

Climbing fiber (CF)–-Purkinje cell (PC) relationships were studied electrophysiologically on the cerebellum of 8 to 15 day old rats. Some animals were rendered agranular by x-irradiation from birth; some others were treated with 3-acetyl pyridine 3 days before study to selectively destroy the CF. Unitary extracellular recordings in 8–9 day old normal rats revealed that more than 50μ of the PC units each exhibited either two types of all-or-none climbing fiber responses (CFR) or stepwise graded CFRs. The other PC units only presented one type of all-or-none CFR. These activities were entirely mediated via CF since they persisted at the same age in x-irradiated rats, but were absent in animals treated with 3-acetyl pyridine. Interaction experiments were performed between juxtafastigial and Inferior Olive stimulations on 49 PC units in 8–9 day old normal rats. Collisions between impulses set up in CFs were disclosed in 21 out of the 24 PCs which exhibited only one type of CFR. In the three others and in each of the 25 PCs that displayed two types of all-or-none CFRs, or CFRs graded by steps, no collision was detected. Moreover intracellular recordings of epsp's mediated via CFs in PCs of 8–9 day old normal rats revealed that they frequently fluctuated in stepwise fashion. These results demonstrate that in the immature rat more than 50μ of PCs are each innervated by at least two distinct CFs; later on, the disappearance of the supernumerary synapses between CF and PC leads, as early as day 15, to the one-to-one relationship between CF and PC.  相似文献

When three sheep were abruptly changed from a ration of 100% orchardgrass hay to 60% cracked corn-40% orchardgrass hay, fed at equal dry-matter intakes, significant increases in concentration were observed in the rumen microbial population. Bacterial numbers (colony counts) per gram of rumen contents did not appear to have stabilized within 21 days after the ration change; however, protozoan numbers per milliliter plateaued after 5 days. The concentration of cellulose-digesting bacteria varied considerably between animals and decreased in all animals with the change. Changes were observed in total and molar percentages of volatile fatty acids, which were typical for the two types of rations. Although the concentration of protozoa increased after the ration change, only minor differences were observed in their percent generic distribution. A significant decrease in rumen volume was measured in two of the three sheep with the change in ration; however, fluid turnover rates were not significantly affected. Rates of rumen dry-matter turnover were slower with the concentrate ration, although rumen dry-matter digestion was increased. Calculation of total bacterial numbers based on total rumen volume completely negated the effect of ration change in one animal, whereas total numbers in the other two animals were still significantly different between rations and very similar between animals. Adjustment of total protozoa numbers did not alter the trends seen previously with concentration values.  相似文献

Optimal conditions for amino acid incorporation into protein in vitro by isolated skeletal muscle mitochondria were established. Maximum incorporation rates were obtained when atractylate and glutamate were added to the incubation medium in the absence of any exogenous adenine nucleotides. Under these conditions, the rate of amino acid incorporation was more than 5-fold greater than that observed with glutamate and ADP and nearly 12-fold greater than that observed with ATP and an ATP-regenerating system consisting of phosphoenolpyruvate and pyruvate kinase. The optimal concentrations of adenine nucleotides, glutamate, cofactors and the substrate leucine were determined for all three energy-providing systems. The inhibitors of protein synthesis, puromycin and chloramphenicol, completely blocked amino acid incorporation by isolated skeletal muscle in mitochondria, while cycloheximide had no effect. Analysis of the labeled mitochondrial proteins by sodium dodecylsulfate polyacrylamide gel electrophoresis revealed five labeled bands of molecular weights ranging from 38,000 to 10,000.Amino acid incorporation by skeletal muscle mitochondria isolated from diabetic rats was decreased over 60% as compared to mitochondria from controls when measured in the presence of glutamate and atractylate, ADP and glutamate or the ATP regenerating system. By contrast, amino acid incorporation by liver mitochondria isolated from diabetic rats did not differ significantly from control values when measured with four different energy sources.  相似文献
