A 20-year-old woman had a cyst of the proximal part of the common bile duct and a cyst of the left hepatic duct; these lesions were diagnosed preoperatively by intravenous cholangiography and successfully operated upon. At the time of writing, she has been followed up for one year.Congenital defects in the biliary system are rare and, in a review of the literature, only two cases were found similar to this one. It is generally accepted that these lesions are congenital, but the exact pathogenesis is unknown.Alonso-Lej, Rever and Pessagno2 reviewed the literature in 1959 and found 403 authentic congenital cysts of the hepatic ducts. The most common congenital defect is a single choledochal cyst of the lower end of the common bile duct. Pain, jaundice and tumour are the main symptoms.Until the advent of intravenous cholangiography, these lesions were seldom recognized preoperatively. Electropotential differences between the cytoplasm and external medium have been compared in the mature R. pipiens occyte and the ovulated unfertilized egg as a function of [Na]o, [K]o, [Ca]o and [Cl]o. In solutions containing 1.0 mM Ca++ the oocyte behaved as though it were predominantly permeable to K+ and Cl?, i.e., like a KCl electrode. However, the steady potential decreased with decreasing [Ca]o and in 5 × 10?4 mM [Ca]o the oocyte membrane behaved like a NaCl electrode. Studies on the steady potential as a function of [Na]o, [K]o and [Cl]o in 1.0 mM Ca++ or Ca-free solutions suggest that Ca++ controls the passive permeability of the oocyte membrane to Na+ and Cl?. In the ovulated unfertilized egg the K+ selectivity of the cell membrane disappeared and the system behaved like a NaCl electrode. No effect of external Ca++ or K+ concentration changes on the steady potential was observed. These results indicate that the ion permeability properties of the ovulated egg are similar to that of the ovarian oocyte in Ca-deficient medium, and suggests that the mechanism of ovulation may involve the removal of Ca++ regulation of ion permeability of the egg cell membrane. 相似文献

The search for DNA biomarkers of oxidative stress has been hampered for several decades by the lack of relevant information on base oxidation products and the challenging issue of measuring low amounts of lesions, typically a few modifications within the range 106?C108 normal bases. In addition and this was ignored for a long time, there is a risk of artifactual oxidation of overwhelming nucleobases during DNA extraction and subsequent workup that has led to overestimation of some base damage up to 2?C3 orders of magnitude. The main aim of the survey is to critically review the available methods that have been developed for measuring oxidatively generated base damage in nuclear and mitochondrial DNA. Among the chromatographic methods, high-performance liquid chromatography associated with tandem mass spectrometry (HPLC?CMS/MS) is the most accurate and versatile approach whereas HPLC?Celectrochemical detection (ECD) is restricted to electrochemically active modifications. These methods allow measuring several single oxidized pyrimidine and purine bases, tandem base lesions and interstrand DNA cross-links in nuclear DNA. As complementary analytical tools, enzymatic methods that associate DNA repair enzymes with either the alkaline comet assay or the alkaline elution technique are suitable for assessing low variations in the level of different classes of oxidatively generated DNA lesions. Most of the immunoassays suffer from a lack of specificity due to the occurrence of cross-reactivity with overwhelming normal bases. One major exception concerns the immunodetection of 5-hydroxymethylcytosine, produced in a relatively high yield as an epigenetic DNA modification. HPLC?CMS/MS is now recognized as the gold standard for measuring oxidized bases and nucleosides in human fluids such as urine, saliva, and plasma. 相似文献

Both substance-P and vasoactive intestinal peptide (VIP) have previously been demonstrated to contract and relax, respectively, the isolated guinea pig trachea. In addition, substance-P and VIP have been localized within the pulmonary innervation of various species. In the present studies, substance-P was found to cause a concentration-related contraction of isolated lung parenchymal strips of the guinea pig, as well as isolated tracheal strips. VIP caused a significant concentration-related relaxation of the isolated tracheal strip, but not the lung parenchymal strip. Indomethacin, a prostaglandin synthetase inhibitor, potentiated the contractile response of the trachea to substance-P and inhibited the VIP- and isoproterenol-induced relaxation. These studies are potentially important in understanding the pathogenesis of bronchospastic disorders, since alterations in prostaglandin biosynthesis may result in hyperreactivity of airways to contractile agonists such as neurotransmitters, as well as an inhibition of relaxation induced by endogenous substances such as VIP or β agonists. 相似文献

Bacillus thuringiensis crystal toxin: Ultrastructural studies of its effect on silkworm midgut cells
Bacillus thuringiensis crystal toxin induced a cytoplasmic response in columnar cells within 1 min after ingestion although external symptoms were not exhibited by larvae until 15 min after ingestion. Microvilli became less consistently uniform in diameter; their organized internal microfilaments were disrupted and disappeared. The cisternae of rough endoplasmic reticulum were enlarged and denuded of ribosomes. By 5 min after ingestion, microvilli of some columnar cells disappeared entirely and gross ultrastructural changes were observed in other regions of the cells. Up to 5 min after ingestion there were few, if any, ultrastructural changes observed within goblet cells. Mitochondria in columnar cells were swollen but did not exhibit the condensed configuration reported by other workers. Both the buffer system used in the fixation medium and its osmolarity influenced the changes in the ultrastructure of midgut cells exposed to B. thuringiensis crystal toxin. 相似文献

Summary The lkyB gene of Escherichia coli K12 has been cloned from the Clarke and Carbon colony bank by selecting a ColE1 plasmid conferring cholic acid resistance to lkyB mutants. The lkyB gene was localized on hybrid plasmid pJC778 by analysis of mutated plasmids generated by Tn5 insertions. Restriction analysis and complementation studies indicated that plasmid pJC778 carried genes nadA, lkyB and sucA which mapped at min 16.5; the lkyB
+ allele was dominant over the lkyB207 mutant allele. Analysis of cell envelope proteins from strains carrying plasmids pJC778 (lkyB

The microstructure of two species of Cladochonusfrom Algerian Carboniferous, C. crassus (McCoy) and C. cf. tenuicollisMcCoy, has been studied by ultra-thin sections. Their walls consist of: 1. an external fibrous layer, 2. a median layer of undulating lamellae, 3. an internal zone of slender, elongated elements to which the name of «grundulae is given here. Abstract A novel procedure was used to purify a cytosolic chitinase from Candida albicans to electrophoretic homogeneity. The results represent the first demonstration of the purification of a fungal intracellular chitinase using the criterion of a single band detected following silver-staining of a polyacrylamide gel run under denaturing conditions. Purified chitinase had pH and temperature optima of 5.0 and 50°C, respectively. Inhibition of enzyme activity by allosamidin was pH-dependent occuring maximally at pH 8.0. Phospholipids had similar marked and highly specific effects on the activities of both the purified soluble enzyme and a solubilized microsomal chitinase from C. albicans . Administration of methamphetamine (METH) to animals causes loss of DA terminals in the brain. The manner by which METH causes these changes in neurotoxicity is not known. We have tested the effects of this drug in copper/zinc (CuZn)-superoxide dismutase transgenic (SOD Tg) mice, which express the human CuZnSOD gene. In nontransgenic (non-Tg) mice, acute METH administration causes significant decreases in DA and dihydroxyphenylacetic acid (DOPAC) in the striata of non-Tg mice. In contrast, there were no significant decreases in striatal DA in the SOD Tg mice. The effects of METH on DOPAC were also attenuated in SOD Tg mice. Chronic METH administration caused decreases in striatal DA and DOPAC in the non-Tg mice, but not in the SOD-Tg mice. Similar studies were carried out with 1-methyl-1,2,3,6-tetrahydropyridine (MPTP), which also causes striatal DA and DOPAC depletion. As in the case of METH, MPTP causes marked depletion of DA and DOPAC in the non-Tg mice, but not in the SOD Tg mice. These results suggest that the mechanisms of toxicity of both METH and MPTP involve superoxide radical formation. 相似文献
