Interspecific crosses among Kalmia species are reported for the first time. All possible crosses between K. angustifolia, K. cuneata, K. latifolia, K. hirsuta, K. polifolia var. rosmarinifolia and K. p. var. microphylla were attempted. Several crosses yielded no hybrids, many produced albino or yellow-green inviable seedlings, and six combinations produced at least a few seedlings with near-normal growth habit. Stylar inhibition of pollen tube growth prevented some species from hybridizing; reciprocal differences in crossability and pollen tube growth were noted. Kalmia species are highly heterozygous, but no evidence was found within a species for geographic variation of combining ability except for the two varieties of K. polifolia which had different crossing patterns with the other species. The anthocyaninless form of K. angustifolia, in contrast to the normal, has unique combining ability with K. latifolia. No genetic evidence was found to support the placement of K. hirsuta in another genus (Kalmiella) or to designate K. angustifolia var. caroliniana as a distinct species. Use of all the species to breed improved ornamentals is limited by genetic barriers and sterility of F1 hybrids.  相似文献

Megadromus guerinii, an endemic carabid beetle (Carabidae), is the most common carabid throughout its restricted range on Banks Peninsula, a formation of extinct volcanoes in Canterbury, New Zealand. This study characterises the small-scale phylogeographic patterns of M. guerinii across the formerly volcanically active Banks Peninsula using mitochondrial and ribosomal genes. Between the eastern and western areas of the peninsula, the mitochondrial, but not nuclear, DNA has a well-defined geographic distribution. Specifically, mitochondrial cytochrome c oxidase subunit I (CO1) identifies two distinct groups (> 6% divergence between eastern and western beetles) while ribosomal genes show no discernible pattern. Whether such a pattern represents male-biased dispersal, Wolbachia infection, a recent range expansion of a divergent lineage, or a deeper historic separation is explored. There is potential that male-biased dispersal could have occurred. Wolbachia infection was not detected. We conclude that historical processes have likely separated taxa in the eastern and western peninsula.  相似文献

DNA variation in human populations was studied by examining the last intron of the ZFX gene (about 1, 151 bp) with a worldwide sample of 29 individuals. Only one polymorphic site was found, which is located in an Alu sequence. This polymorphism is present at an intermediate frequency in all populations studied, and could be a shared polymorphism or due to migration among populations in Asia, Europe, and Africa. The nucleotide diversity is 0.04%, supporting the view that the level of nucleotide variation in nuclear DNA is very low in humans. From the sequence data, the age (T) of the most recent common ancestor of the sampled sequences is estimated: the mode of T is about 306,000 years, and the 95% confidence interval of T is 162,000-952,000 years. This mode estimate is considerably older than the estimates from Y- linked sequences.   相似文献

Background  

Seagrasses are a polyphyletic group of monocotyledonous angiosperms that have adapted to a completely submerged lifestyle in marine waters. Here, we exploit two collections of expressed sequence tags (ESTs) of two wide-spread and ecologically important seagrass species, the Mediterranean seagrass Posidonia oceanica (L.) Delile and the eelgrass Zostera marina L., which have independently evolved from aquatic ancestors. This replicated, yet independent evolutionary history facilitates the identification of traits that may have evolved in parallel and are possible instrumental candidates for adaptation to a marine habitat.  相似文献

Low-cost and simple technologies are needed to reduce watershed export of excess nitrogen to sensitive aquatic ecosystems. Denitrifying bioreactors are an approach where solid carbon substrates are added into the flow path of contaminated water. These carbon (C) substrates (often fragmented wood-products) act as a C and energy source to support denitrification; the conversion of nitrate (NO3?) to nitrogen gases. Here, we summarize the different designs of denitrifying bioreactors that use a solid C substrate, their hydrological connections, effectiveness, and factors that limit their performance. The main denitrifying bioreactors are: denitrification walls (intercepting shallow groundwater), denitrifying beds (intercepting concentrated discharges) and denitrifying layers (intercepting soil leachate). Both denitrifcation walls and beds have proven successful in appropriate field settings with NO3? removal rates generally ranging from 0.01 to 3.6 g N m?3 day?1 for walls and 2–22 g N m?3 day?1 for beds, with the lower rates often associated with nitrate-limitations. Nitrate removal is also limited by the rate of C supply from degrading substrate and removal is operationally zero-order with respect to NO3? concentration primarily because the inputs of NO3? into studied bioreactors have been generally high. In bioreactors where NO3? is not fully depleted, removal rates generally increase with increasing temperature. Nitrate removal has been supported for up to 15 years without further maintenance or C supplementation because wood chips degrade sufficiently slowly under anoxic conditions. There have been few field-based comparisons of alternative C substrates to increase NO3? removal rates but laboratory trials suggest that some alternatives could support greater rates of NO3? removal (e.g., corn cobs and wheat straw). Denitrifying bioreactors may have a number of adverse effects, such as production of nitrous oxide and leaching of dissolved organic matter (usually only for the first few months after construction and start-up). The relatively small amount of field data suggests that these problems can be adequately managed or minimized. An initial cost/benefit analysis demonstrates that denitrifying bioreactors are cost effective and complementary to other agricultural management practices aimed at decreasing nitrogen loads to surface waters. We conclude with recommendations for further research to enhance performance of denitrifying bioreactors.  相似文献

Bacteria and fungi drive the decomposition of dead plant biomass (litter), an important step in the terrestrial carbon cycle. Here we investigate the sensitivity of litter microbial communities to simulated global change (drought and nitrogen addition) in a California annual grassland. Using 16S and 28S rDNA amplicon pyrosequencing, we quantify the response of the bacterial and fungal communities to the treatments and compare these results to background, temporal (seasonal and interannual) variability of the communities. We found that the drought and nitrogen treatments both had significant effects on microbial community composition, explaining 2–6% of total compositional variation. However, microbial composition was even more strongly influenced by seasonal and annual variation (explaining 14–39%). The response of microbial composition to drought varied by season, while the effect of the nitrogen addition treatment was constant through time. These compositional responses were similar in magnitude to those seen in microbial enzyme activities and the surrounding plant community, but did not correspond to a consistent effect on leaf litter decomposition rate. Overall, these patterns indicate that, in this ecosystem, temporal variability in the composition of leaf litter microorganisms largely surpasses that expected in a short-term global change experiment. Thus, as for plant communities, future microbial communities will likely be determined by the interplay between rapid, local background variability and slower, global changes.  相似文献

The organisational principles of locomotor networks are less well understood than those of many sensory systems, where in-growing axon terminals form a central map of peripheral characteristics. Using the neuromuscular system of the Drosophila embryo as a model and retrograde tracing and genetic methods, we have uncovered principles underlying the organisation of the motor system. We find that dendritic arbors of motor neurons, rather than their cell bodies, are partitioned into domains to form a myotopic map, which represents centrally the distribution of body wall muscles peripherally. While muscles are segmental, the myotopic map is parasegmental in organisation. It forms by an active process of dendritic growth independent of the presence of target muscles, proper differentiation of glial cells, or (in its initial partitioning) competitive interactions between adjacent dendritic domains. The arrangement of motor neuron dendrites into a myotopic map represents a first layer of organisation in the motor system. This is likely to be mirrored, at least in part, by endings of higher-order neurons from central pattern-generating circuits, which converge onto the motor neuron dendrites. These findings will greatly simplify the task of understanding how a locomotor system is assembled. Our results suggest that the cues that organise the myotopic map may be laid down early in development as the embryo subdivides into parasegmental units.  相似文献

Summary The nucleotide sequence of cloned DNA corresponding to full-length mouse muscle creatine kinase mRNA has been determined. This 1415 base pair DNA sequence and the deduced 381 amino acid sequence of the protein have been compared to creatine kinase sequences from other vertebrate species and to invertebrate guanidino kinase sequences. These comparisons show that the vertebrate muscle creatine kinases constitute a remarkably conserved protein family with a unit evolutionary period of 30. The creatine kinases also retain marked sequence similarity with the more distantly related invertebrate guanidino kinases. A portion of the sequence, presumably part of the ATP binding site, shows similarity to other nucleotide binding proteins with diverse functions. Comparisons of the untranslated regions of the creatine kinase cDNA sequences show that the 5 untranslated regions are more highly conserved than are the 3 untranslated regions; this may point to some regulatory function in the 5 region.  相似文献
