Mature?mammalian?oocytes?are?poised?for?completing?meiosis?II (MII) on fertilization by positioning the spindle close to an actomyosin-rich cortical cap. Here, we show that the Arp2/3 complex localizes to the cortical cap in a Ran-GTPase-dependent manner and nucleates actin filaments in the cortical cap and a cytoplasmic actin network. Inhibition of Arp2/3 activity leads to rapid dissociation of the spindle from the cortex. Live-cell imaging and spatiotemporal image correlation spectroscopy analysis reveal that actin filaments flow continuously away from the Arp2/3-rich cortex, driving a cytoplasmic streaming expected to exert a net pushing force on the spindle towards the cortex. Arp2/3 inhibition not only diminishes this actin flow and cytoplasmic streaming but also enables a reverse streaming driven by myosin-II-based cortical contraction, moving the spindle away from the cortex. Thus, the asymmetric MII spindle position is dynamically maintained as a result of balanced forces governed by the Arp2/3 complex. 相似文献

Food consumption, digesta retention time, and food preference were measured for captive whooping cranes fed pelleted diets. The basal commercial diet was compared to four mixtures containing 70% basal and 30% of one of four important winter foods for the whooping crane: blue crab (Callinectes sapidus), wolfberry fruit (Lycium carolinianum), live oak acorn (Quercus virginiana), or common Rangia clam (Rangia cuneata). Because captive birds would not eat whole foods, we were prevented from direct food preference tests. Food passed through the gut rapidly, with almost complete elimination within 7 hr. There was some indication that retention time was shorter for the low fiber and high ash and calcium clam diet. Cranes ate less wolfberry feed (g/day) than the other feeds, and all birds ate less wolfberry feed on the day it was first fed, compared to basal diet the previous day. Birds ate more low energy feed than high energy feed. Due to combined effects of low energy content, lower metabolizable energy coefficients, and reduced feed consumption, less energy was assimilated for study diets than basal diet. Apparent shorter retention times for some diets containing whooping crane foods may partly explain lower digestibilities and metabolizable energy of winter whooping crane foods compared to commercial crane diet. Many disease states have associated blood viscosity changes. Molecular rotors, fluorescent molecules with viscosity sensitive quantum yields, have recently been investigated as a new method for biofluid viscosity measurement. Current viscometer measurements are complicated by proteins adhering to surfaces and forming air-surface layers. It is unknown at this time what effects proteins may have on biofluid viscosity measurements using molecular rotors. To answer this question, binding affinities to blood plasma proteins were investigated by equilibrium dialysis for four hydrophilic molecular rotors. Aqueous solutions of 9-[(2-cyano-2-hydroxy-carbonyl)vinyl]julolidine (CCVJ) and three derivatives were prepared and dialyzed against solutions of bovine source albumin, fibrinogen and immunoglobulin G approximating normal physiologic concentrations and fresh-frozen human plasma. After equilibration, dye concentration on each side of the dialysis membrane was assessed by spectrophotometry. The relative binding affinity of the four dyes to the proteins and to the plasma was compared. Affinity of all dyes was highest for albumin. The bound dye fraction showed little change in relation to protein concentration in the physiological concentration range. Diol, the most hydrophilic molecular rotor tested showed the lowest affinity for albumin. Efforts to further optimize the clinical candidate razaxaban have led to a new series of pyrazole-based factor Xa (fXa) inhibitors. Designed to prevent the potential formation of primary aniline metabolites in vivo, the nitrogen of the carboxamido linker between the pyrazole and proximal phenyl moiety of the razaxaban scaffold was replaced with a methylene group. The resulting ketones demonstrated excellent potency and selectivity for fXa but initially had poor oral bioavailability. Optimization by conversion from a P1 aminobenzisoxazole to a P1 p-methoxyphenyl residue, replacing the 3-trifluoromethylpyrazole with a 3-amidopyrazole, and employing a pyridone P4 group provided a fXa inhibitor with a potency and pharmacokinetic profile equivalent to that of razaxaban and improved selectivity over thrombin. 相似文献

Glioblastoma multiforme (GBM) is a high-grade primary brain cancer with a median survival of only 14.6 months in humans despite standard tri-modality treatment consisting of surgical resection, post-operative radiation therapy and temozolomide chemotherapy 1. New therapeutic approaches are clearly needed to improve patient survival and quality of life. The development of more effective treatment strategies would be aided by animal models of GBM that recapitulate human disease yet allow serial imaging to monitor tumor growth and treatment response. In this paper, we describe our technique for the precise stereotactic implantation of bio-imageable GBM cancer cells into the brains of nude mice resulting in tumor xenografts that recapitulate key clinical features of GBM 2. This method yields tumors that are reproducible and are located in precise anatomic locations while allowing in vivo bioluminescent imaging to serially monitor intracranial xenograft growth and response to treatments 3-5. The bacterial symbiont Cardinium (Bacteroidetes) was previously implicated in the thelytokous reproduction of the parasitoid Plagiomerus diaspidis Crawford (Hymenoptera: Encyrtidae). Horizontal transmission of the symbiont among the cactus scale Diaspis echinocacti Bouché (Homoptera: Diaspididae) and its hymenopteran parasitoids has been suggested. In this study, the bacteria associated with D. echinocacti, its parasitoids P. diaspidis and Aphytis sp. (Hymenoptera: Aphelinidae), and the hyperparasitoid Marietta leopardina Motschulsky (Hymenoptera: Aphelinidae) were characterized using molecular fingerprinting techniques, and the localization of Cardinium in P. diaspidis was studied using fluorescence in situ hybridizations (FISH). Cardinium was the only bacterium found in P. diaspidis, but it could not be detected in any of the other insects tested. The symbiont was specifically located in the reproductive tissues of its P. diaspidis host. 相似文献
