Fat metabolism in higher plants. Properties of a soluble stearyl-acyl carrier protein desaturase from maturing Carthamus tinctorius 总被引:10,自引:0,他引:10
A soluble extract from maturing safflower seeds (Carthamus tinctorius) synthesized [14C]oleic acid from [14C]malonate, or [14C]stearyl-acyl carrier protein. Stearyl-acyl carrier protein was generated from [14C]malonate by the seed extract. The desaturase had only a trace of activity when stearyl-CoA was the substrate. The stearyl-acyl carrier protein desaturase had a specific requirement for ferredoxin which was only partially replaced by flavodoxin. While NADPH was an effective reductant, NADH was ineffective. However, the most effective reductant was a system composed of ferredoxin, grana lamellae, ascorbic acid, dichlorophenolindophenol, and light. No NADPH requirement was observed when this reducing system was employed. Stearylacyl carrier protein desaturase activity was enhanced by dithiothreitol and reduced glutathione, but was partially inhibited by β-mercaptoethanol. The desaturase activity was inhibited by 1 mm potassium cyanide but insensitive to carbon monoxide. No lipid micelle requirement could be demonstrated. 相似文献

In a 6-year cover crop sequence study, nematode population densities varied with different cover crops. Millet favored rapid increase of Belonolaimus longicaudatus and supported relatively large numbers of Pratylenchus brachyurus. Beggarweed and ''Coastal'' bermudagrass favored rapid increase of B. Iongicaudatus and to a lesser extent P. brachyurus and Trichodorus christiei. Hairy indigo and Crotalaria supported relatively large numbers of P. brachyurus but suppressed B. longicaudatus. Hairy indigo also supported increases of T. christiei and Xiphinema americanum. Marigold did not favor development of any parasitic nematode species present. Tomato transplant yield was inversely related to nematode population, particularly to B. Iongicaudatus. Largest yields were obtained from plots with smallest numbers of B. longicaudatus and smallest yields were from plots with largest numbers of B. longicaudatus. 相似文献

Stearyl-acyl carrier protein desaturase (EC 1.14.99.6), present in the stroma fraction of spinach (Spinacia oleracea) chloroplasts, rapidly desaturated enzymatically prepared stearyl-acyl carrier protein to oleic acid. No other substrates were desaturated. In addition to stearyl-acyl carrier protein, reduced ferredoxin was an essential component of the system. The electron donor systems were either ascorbate, dichlorophenolindophenol, photosystem I and light, or NADPH and ferredoxin-NADP reductase. The desaturase was more active in extracts prepared from chloroplasts obtained from immature spinach leaves than from mature leaves. Stearyl-acyl carrier protein desaturase also occurs in soluble extracts of avocado (Persea americana Mill.) mesocarp and of developing safflower (Carthamus tinctorius) seeds. 相似文献

Total DNA was extracted from vegatative mycelia of the water mold Achlya bisexualis. Fractionation of the DNA in CsCl gradients resulted in two components: a major component with a buoyant density of 1.697 g cm?3 and a minor component with a density of 1.685 g cm?3. The minor component has been identified as mitochondrial DNA based on extractions from isolated mitochondria and Triton X-100 washed nuclei. Detergent washing of the nuclei yielded DNA in which the mitochondrial DNA component was absent, while the isolated mitochondrial preparations contained DNA enriched in the 1.685 g cm?3 component. Hybridization studies of A. bisexualis DNA to rRNA show that the ribosomal cistrons have a buoyant density coincident with that obtained with the nuclear DNA. In addition, preliminary evidence indicates that the mitochondrial DNA does not hybridize to the cytoplasmic RNA under the conditions used for this study. Benzothiadiazines, potent oral diuretics, also inhibit carbonic anhydrase, although to a lesser degree than acetazolamide (Diamox). The latter has little effect on urinary calcium but causes hypocitriuria which predisposes to nephrolithiasis. The effect of benzothiadiazines on urinary calcium/citrate balance is less well known. In this study, the fall of urinary calcium and citrate was roughly 50% and 30%, respectively, in normal and hypertensive subjects during the prolonged administration of hydrochlorothiazide, a member of the benzothiadiazine family. Hence, in contradistinction to acetazolamide, this drug produces hypocalciuria but depresses the urinary citrate less. Consequently, the benzothiadiazines would not be expected to predispose to nephrolithiasis. In addition, hydrochlorothiazide was found to produce a 10-20% fall in creatinine clearance and an initial rise of roughly 0.7 mg./100 ml. in serum calcium concentration. In search for new drugs lowering arterial blood pressure, which could be applied in anti‐hypertensive therapy, research concerning agents blocking of renin‐angiotensin‐aldosteron system has been conducted. Despite many years of research conducted at many research centers around the world, aliskiren is the only one renin inhibitor, which is used up to now. Four novel potential renin inhibitors, having structure based on the peptide fragment 8–13 of human angiotensinogen, a natural substrate for renin, were designed and synthesized. All these inhibitors contain unnatural moieties that are derivatives of N‐methylleucyl‐β‐hydroxy‐γ‐amino acids at the P2‐P1' position: 4‐[N‐(N‐methylleucyl)‐amino]‐3‐hydroxy‐7‐(3‐nitroguanidino)‐heptanoic acid (AHGHA), 4‐[N‐(N‐methylleucyl)‐amino]‐3‐hydroxy‐5‐phenyl‐pentanoic acid (AHPPA) or 4‐[N‐(N‐methylleucyl)‐amino]‐8‐benzyloxycarbonylamino‐3‐hydroxyoctanoic acid (AAHOA). The previously listed synthetic β‐hydroxy‐γ‐amino acids constitute pseudodipeptidic units that correspond to the P1‐P1' position of the inhibitor molecule. An unnatural amino acid, 4‐methoxyphenylalanin (Phe(4‐OMe)), was introduced at the P3 position of the obtained compounds. Three of these compounds contain isoamylamide of 6‐aminohexanoic acid (ε‐Ahx‐Iaa) at the P2'‐P3' position. The proposed modifications of the selected human angiotensinogen fragment are intended to increase bioactivity, bioavailability, and stability of the inhibitor molecule in body fluids and tissues. The inhibitor Boc‐Phe(4‐OMe)‐MeLeu‐AHGHA‐OEt was obtained in the form of an ethyl ester. The hydrophobicity coefficient, expressed as log P varied between 3.95 and 8.17. In vitro renin inhibitory activity of all obtained compounds was contained within the range 10?6‐10?9 M. The compound Boc‐Phe(4‐OMe)‐MeLeu‐AHPPA‐Ahx‐Iaa proved to be the most active (IC50 = 1.05 × 10?9 M). The compounds Boc‐Phe(4‐OMe)‐MeLeu‐AHGHA‐Ahx‐Iaa and Boc‐Phe(4‐OMe)‐MeLeu‐AHPPA‐Ahx‐Iaa are resistant to chymotrypsin. Copyright © 2016 European Peptide Society and John Wiley & Sons, Ltd. 相似文献

From the culture broth of the mold Trichoderma viride NRRL 5243 a mixture of polypeptides, named trichovirins (TV), could be isolated and purified by chromatography on XAD-2 adsorber resin and Sephadex LH-20 gel. Chromatography on silica gel using chloroform/methanol 8:2 as eluent provided a mixture of peptides named TV I. Subsequent elution with chloroform/methanol 1:1 yielded a second group of peptides named TV II. That group could be separated into individual components by repetitive HPLC on an octadecylsilyl and a fluorocarbon stationary phase. The sequences of 12 peptides of TV II could be determined by electrospray ionization tandem mass spectrometry of isolated peptides and gas chromatography-mass spectrometry of methanolysates. The N-termini of the 18-mer peptides are acetylated and the C-termini consist of leucinol. Owing to the presence of alpha-aminoisobutyric acid (Aib) residues and the bactericidal and hemolytic activity, the peptides belong to the family of peptaibol antibiotics. 相似文献

Fournier gangrene is a rare, rapidly progressive, life-threatening condition. We report a 23-day-old boy with pulmonary atresia and ventricular septal defect treated surgically, who developed Fournier gangrene. Emergency surgery was performed with tissue sampling for microbiological examination. Candida albicans was confirmed; caspofungin followed by fluconazole was administered with excellent results. 相似文献
