The development of the Neotropical predatory pentatomid Brontocoris tabidus on an artificial diet based on beef meat and liver was evaluated. The predator showed significantly longer nymphal development and lower adult weights on this diet than when reared on pupae of the mealworm Tenebrio molitor. The survival of nymphs fed exclusively on the artificial diet was somewhat lower compared with feeding on T. molitor pupae. When B. tabidus was bred on this artificial diet during part of its nymphal period i.e. during the second; second and third; and second, third and fourth instars , and was subsequently returned to T. molitor pupae, the predator nymphs completed the nymphal stage with a developmental rate similar to that of nymphs fed on live prey throughout. The adults attained after switching from the artificial diet to live prey from the third and fourth instar onwards had similar weights to those in the control. Considering the relatively good results obtained with B. tabidus and other members of the pentatomid subfamily Asopinae, this meat-based diet may be a valuable alternative for use in the mass production of predatory pentatomids. 相似文献

In a pot trial growth and transpiration of 3-year-old Douglas-fir seedlings on an acid, sandy soil was examined at a deficient

1) and an excessive level (120 kg N ha –

1) of NH4 application. Dissolved ammonium sulphate was applied to the pots weekly for two growing seasons. In half of the pots a complete
set of other nutrients was applied in optimal proportions to the applied nitrogen. Water supply was optimal and transpiration
was recorded. At the end of the second treatment season irrigation was stopped for 2 weeks during dry and sunny weather. Both
high application of NH4 and additional nutrients increased shoot growth and transpiration demand in the first treatment year. The root system was
smaller at higher N level and this reduced water uptake accordingly. In the second year the combination of high NH4
+ and additional nutrients affected root functioning predominantly due to salinity effects and this seriously decreased water
uptake capacity and shoot water potentials, finally resulting in tree death. Without addition of other nutrients the high
NH4
+ application resulted in a high degree of soil acidification, which damaged the roots, that showed a decrease in water uptake
capacity. At the low NH4 supply level soil acidification was lower, and root functioning was not affected, and the trees recovered quickly from the
imposed drought. Higher needle K and P status depressed transpiration rates at the low NH4 application rate.

Relatively subtle forms of exploitation of human subjects may arise from the inefficiency or incompetence of a researcher, from the existence of a power imbalance between principal and subject, or from the uneven distribution of research risks among various segments of the population. A powerful and knowledgeable person (or institution) may perpetrate the exploitation of an unempowered and ignorant individual even without intending to. There is an ethical burden on the former to protect the interests of the vulnerable. Excessive or insufficient compensation may be exploitative. However, genuine economic imperatives motivating needy volunteers have to be considered. These forms of exploitation should be appreciated in the context of social and cultural factors suggesting that the relationship between researcher and subject cannot properly be appraised as a contractual undertaking. While compliance with pertinent codes and regulations minimises the exploitative potential, they cannot be enforced in a way that does not recognize a society's peculiar characteristics. The experience with some Filipino cultural traits illustrates this point. 相似文献

The role of colony-stimulating factors (CSFs) in regulating the synthesis of thrombospondin 1 (TSP1) by cultured human macrophages is investigated. Macrophage (M)-CSF is shown rapidly and transiently to induce two predominant species of TSP1 mRNA. One of these species was 3.2 kb in size and appeared to be specific to M-CSF-stimulated macrophages. Adherent M-CSF-treated macrophages are also shown to express abundant surface cell-associated TSP rapidly when examined by indirect immunofluorescence staining. Granulocyte-macrophage (GM)-CSF induced TSP1 mRNA at a later time point, and this was attributable to the effects of endogenous M-CSF induced by the GM-CSF; the GM-CSF-treated cells did not display surface-associated TSP after 3 hr of treatment. Analysis of the TSP1 protein synthesised by the M-CSF-treated macrophages revealed the expected trimeric form of the molecule. In addition, an unidentified 95-kDa protein was found to be covalently associated with immunoreactive TSP1, and this appeared to be specific to the macrophages as it was not found in TSP1 precipitated from other cell types. It is suggested that the induction of TSP1 by M-CSF may play an important role in the major physiological functions of macrophages. © 1995 Wiley-Liss, Inc. 相似文献

Summary In the present study we report a novel histochemical method which, by sequential pre-incubations in alkaline and acidic media,
selectively differentiates muscle fibres expressing myosin heavy chain IIX, on the basis of a specific profile for myofibrillar
actomyosin ATPase (mATPase) activity. The enzyme reactions were tested for specificity by means of anti-myosin heavy chain
monoclonal antibodies, which were characterized on Western blots of muscle homogenates. Enzyme histochemical reactions with
the traditional pH buffers were compared to those of the new method and, in conjunction with the immunoreactions, used to
confirm the relationship between MyHC expression and the distinct profiles for mATPase. Imrnunohistochemical reactions demonstrated
that the new method only differentiates those fibres expressing myosin heavy chain IIX. The method revealed a continuum in
which the intermediate staining intensities corresponded to hybrid fibres expressing myosin heavy chain IIX in combination
with either the IIA or IIB forms. Quantitative histochemistry and immunohistochemistry (by image analysis), used to examine
the relationship between staining intensities for mATPase and amounts of myosin heavy chain IIX expression, revealed that
the new method discriminates well between hybrid fibres expressing variable amounts of the IIX isoform (r2 = 0.93). 相似文献

Interferons (IFNs) are generally recognized as the most important therapeutic agent in some infectious diseases such as chronic
hepatitis B and C. Since the early clinical trials it was documented that the therapeutic use of IFNs could be complicated
by the development of antibodies able to neutralize or to bind to the IFN molecule.
After several years of research it is now widely accepted that the presence of circulating anti-IFN antibodies may affect
the response to IFN.
Here we summarize what is currently know on the clinical significance of antibodies to IFN in IFN-treated viral diseases patients. 相似文献

Teratocarcinoma-derived growth factor-1 (Tdgf1), a member of the ``EGF family' of growth factors, is expressed during mouse gastrulation in the forming mesoderm and later
in the truncus arteriosus of the developing heart. In humans, TDGF1 is highly expressed in germ cell tumors and in colon and
mammary carcinomas. In mouse, one gene (Tdgf1) and two pseudogenes (Tdgf1-ps1 and Tdgf1-ps2) have been isolated and characterized. Tdgf1 corresponds to the gene expressed in F9 teratocarcinoma cells. Tdgf1-ps1 and Tdgf1-ps2 are two intronless sequences with all the characteristics of retroposons. In the present paper, we assign the chromosomal
location for Tdgf1, Tdgf1-ps1, and Tdgf1-ps2 sequences to Chromosomes (Chrs) 9, 16, and 17, respectively. Two previously described mouse mutants, scant hair (sch) and fur deficient (fd), map near the Tdgf1 gene. Analysis of their DNA coding region provided no evidence that Tdgf1 could be the responsible gene for these phenotypes. Finally, analysis of the DNA from several Mus musculus strains and from Mus spretus mice revealed a highly variable restriction pattern and the absence of the Tdgf1-ps1 genomic sequence from the Mus spretus genome.

The six group B coxsackieviruses (CVBs) are highly prevalent human pathogens that cause viremia followed by involvement of different organs. Clinical and experimental evidence suggests that CVBs can induce kidney injury, but the susceptibility of human renal cells to these viruses is unknown. By using pure cultures of human glomerular and tubular cells, we demonstrated that all CVBs are capable of productively infecting renal cells of three different histotypes. Distinct pathogenic effects were observed. Proximal tubular epithelial cells and, to a lesser extent, glomerular podocytes were highly susceptible to CVBs; in both cases, infection led to cytolysis. In contrast, glomerular mesangial cells supported the replication of the six CVBs but failed to develop overt cytopathologic changes. Mesangial cells continued to produce infectious progeny for numerous serial subcultures (i.e., more than 50 days), especially with type 1, 3, 4, and 5 viruses. In the above cells, persistent infection induced the de novo synthesis of platelet-derived growth factor A/B and enhanced the release of transforming growth factor beta1/2. These two factors are important mediators of progression from glomerular inflammation to glomerulosclerosis. CVB replication appeared also to impair the phagocytic and contractile activity of mesangial cells. Loss of these properties--which are important in glomerular physiopathology--may contribute to the development of progressive nephropathy. The results show that CVBs induce distinct effects in different types of cultured renal cells and suggest that CVB infections may be associated with both acute and progressive renal injury. 相似文献
