It is unknown whether nutritional deficiencies affect the morphology and function of structural cells, such as epithelial cells, and modify the susceptibility to viral infections. We developed an in vitro system of differentiated human bronchial epithelial cells (BEC) grown either under selenium-adequate (Se+) or selenium-deficient (Se–) conditions, to determine whether selenium deficiency impairs host defense responses at the level of the epithelium. Se– BECs had normal SOD activity, but decreased activity of the selenium-dependent enzyme GPX1. Interestingly, catalase activity was also decreased in Se– BECs. Both Se– and Se+ BECs differentiated into a mucociliary epithelium; however, Se– BEC demonstrated increased mucus production and increased Muc5AC mRNA levels. This effect was also seen in Se+ BEC treated with 3-aminotriazole, an inhibitor of catalase activity, suggesting an association between catalase activity and mucus production. Both Se– and Se+ were infected with influenza A/Bangkok/1/79 and examined 24 h postinfection. Influenza-induced IL-6 production was greater while influenza-induced IP-10 production was lower in Se– BECs. In addition, influenza-induced apoptosis was greater in Se– BEC as compared to the Se+ BECs. These data demonstrate that selenium deficiency has a significant impact on the morphology and influenza-induced host defense responses in human airway epithelial cells.  相似文献

Xeroderma pigmentosum (XP) and Cockayne syndrome (CS) are two rare inherited disorders with a clinical and cellular hypersensitivity to the UV component of the sunlight spectrum. Although the two traits are generally considered as clinically and genetically distinct entities, on the biochemical level a defect in the nucleotide excision-repair (NER) pathway is involved in both. Classical CS patients are primarily deficient in the preferential repair of DNA damage in actively transcribed genes, whereas in most XP patients the genetic defect affects both "preferential" and "overall" NER modalities. Here we report a genetic study of two unrelated, severely affected patients with the clinical characteristics of CS but with a biochemical defect typical of XP. By complementation analysis, using somatic cell fusion and nuclear microinjection of cloned repair genes, we assign these two patients to XP complementation group G, which previously was not associated with CS. This observation extends the earlier identification of two patients with a rare combined XP/CS phenotype within XP complementation groups B and D, respectively. It indicates that some mutations in at least three of the seven genes known to be involved in XP also can result in a picture of partial or even full-blown CS. We conclude that the syndromes XP and CS are biochemically closely related and may be part of a broader clinical disease spectrum. We suggest, as a possible molecular mechanism underlying this relation, that the XPGC repair gene has an additional vital function, as shown for some other NER genes.  相似文献

To follow stomatal responses to ozone (O3) in different Arabidopsis lines, we constructed a rapid-response O3 exposure/gas-exchange measurement device consisting of eight through-flow whole-rosette cuvettes. To separate rosette from roots and growth substrate, plant is grown through an agar-filled hole in a polished glass plate fixed on the pot. Following insertion of the plant, the plate forms air-tight bottom surface of the cuvette; thus the rosette is enclosed without touching it during any phase of the insertion of the plant to the cuvette. The device allows monitoring rapid responses in the stomatal function. For example, an acute exposure to 150 ppb O3 decreased stomatal conductance to 60–70% of its initial value within 9–12 min. Thereafter, the conductance regained its preexposure value within further 30–40 min in spite of the continuing O3 exposure. The transient decrease was absent in the abscisic acid-insensitive mutant abi2 defective in a class 2C protein phosphatase. This provides an in vivo confirmation that the early transient decrease in stomatal conductance is not a result of physical damage by the reactive oxygen species (ROS) formed from O3 breakdown but reflects the biological action of ROS, transduced through a signalling cascade. Thus, the apparatus will be helpful in specifying complex molecular and genetic interactions in rapid responses in guard cells in vivo.  相似文献

Background

Modified function of immune cells in nasal secretions may play a role in the enhanced susceptibility to respiratory viruses that is seen in smokers. Innate immune cells in nasal secretions have largely been characterized by cellular differentials using morphologic criteria alone, which have successfully identified neutrophils as a significant cell population within nasal lavage fluid (NLF) cells. However, flow cytometry may be a superior method to fully characterize NLF immune cells. We therefore characterized immune cells in NLF by flow cytometry, determined the effects of live attenuated influenza virus (LAIV) on NLF and peripheral blood immune cells, and compared responses in samples obtained from smokers and nonsmokers.

Methods

In a prospective observational study, we characterized immune cells in NLF of nonsmokers at baseline using flow cytometry and immunohistochemistry. Nonsmokers and smokers were inoculated with LAIV on day 0 and serial nasal lavages were collected on days 1-4 and day 9 post-LAIV. LAIV-induced changes of NLF cells were characterized using flow cytometry. Cell-free NLF was analyzed for immune mediators by bioassay. Peripheral blood natural killer (NK) cells from nonsmokers and smokers at baseline were stimulated in vitro with LAIV followed by flow cytometric and mediator analyses.

Results

CD45(+)CD56(-)CD16(+) neutrophils and CD45(+)CD56(+) NK cells comprised median 4.62% (range 0.33-14.52) and 23.27% (18.29-33.97), respectively, of non-squamous NLF cells in nonsmokers at baseline. LAIV did not induce changes in total NK cell or neutrophil percentages in either nonsmokers or smokers. Following LAIV inoculation, CD16(+) NK cell percentages and granzyme B levels increased in nonsmokers, and these effects were suppressed in smokers. LAIV inoculation enhanced expression of activating receptor NKG2D and chemokine receptor CXCR3 on peripheral blood NK cells from both nonsmokers and smokers in vitro but did not induce changes in CD16(+) NK cells or granzyme B activity in either group.

Conclusions

These data are the first to identify NK cells as a major immune cell type in the NLF cell population and demonstrate that mucosal NK cell cytotoxic function is suppressed in smokers following LAIV. Altered NK cell function in smokers suggests a potential mechanism that may enhance susceptibility to respiratory viruses.  相似文献

Polar bears are heavily dependent on sea ice for hunting sufficient prey to meet their energetic needs. When the bears are left fasting, it may cause a rise in the levels of the stress hormone cortisol. Cortisol is the major corticosteroid hormone in most mammals, including polar bears. Production and regulation of this stress hormone are vital for the body as it is part of a myriad of processes, including in relation to metabolism, growth, development, reproduction, and immune function. In the present study, we examined the correlation between East Greenland polar bear hair cortisol concentration (HCC), a matrix that reflects longer-term hormone levels, and the fluctuations of the North Atlantic Oscillation (NAO) index, a large-scale climate phenomenon applied as a proxy for sea ice extent in the Greenland Sea along the coast of East Greenland. In doing so, a significant positive correlation (r = 0.88; p = 0.0004) was found between polar bear hair cortisol and the NAO, explaining 77 % of the variation in HCC observed between years over the period 1989–2009. This result indicates that interannual fluctuations in climate and ice cover have a substantial influence on longer-term cortisol levels in East Greenland polar bears. Further research into the implications and consequences inherent in this correlation are recommended, preferably across multiple polar bear populations.  相似文献

V(D)J rearrangement in lymphoid cells involves repair of double-strand breaks (DSBs) through non-homologous end joining (NHEJ). Defects in this process lead to increased radiosensitivity and severe combined immunodeficiency (RS-SCID). Here, a SCID patient, M3, is described with a T?B+NK+ phenotype but without causative mutations in CD3δ, ?, ζ or IL7Rα, genes specifically involved in T cell development. Clonogenic survival of M3 fibroblasts showed an increased sensitivity to the DSB-inducing agents ionizing radiation and bleomycin, as well as the crosslinking compound, mitomycin C. We did not observe inactivating mutations in known NHEJ genes and results of various DSB-repair assays in G1 M3 cells were indistinguishable from those obtained with normal cells. However, we found increased chromosomal radiosensitivity at the G2 phase of the cell cycle. Checkpoint analysis indicated functional G1/S and intra-S checkpoints after irradiation but impaired activation of the “early” G2/M checkpoint. Together these results indicate a novel class of RS-SCID patients characterized by the specific absence of T lymphocytes and associated with defects in G2-specific DSB repair. The pronounced G2/M radiosensitivity of the RS-SCID patient described here, suggests a defect in a putative novel and uncharacterized factor involved in cellular DNA damage responses and T cell development.  相似文献

The value of the diffusion coefficient for oxygen in muscle is uncertain. The diffusion coefficient is important because it is a determinant of the extracellular oxygen tension at which the core of muscle fibers becomes anoxic (Po(2crit)). Anoxic cores in muscle fibers impair muscular function and may limit adaptation of muscle cells to increased load and/or activity. We used Hill's diffusion equations to determine Krogh's diffusion coefficient (Dalpha) for oxygen in single skeletal muscle fibers from Xenopus laevis at 20 degrees C (n = 6) and in myocardial trabeculae from the rat at 37 degrees C (n = 9). The trabeculae were dissected from the right ventricular myocardium of control (n = 4) and monocrotaline-treated, pulmonary hypertensive rats (n = 5). The cross-sectional area of the preparations, the maximum rate of oxygen consumption (Vo(2 max)), and Po(2crit) were determined. Dalpha increased in the following order: Xenopus muscle fibers Dalpha = 1.23 nM.mm(2).mmHg(-1).s(-1) (SD 0.12), control rat trabeculae Dalpha = 2.29 nM.mm(2).mmHg(-1).s(-1) (SD 0.24) (P = 0.0012 vs. Xenopus), and hypertrophied rat trabeculae Dalpha = 6.0 nM.mm(2).mmHg(-1).s(-1) (SD 2.8) (P = 0.039 vs. control rat trabeculae). Dalpha increased with extracellular space in the preparation (Spearman's rank correlation coefficient = 0.92, P < 0.001). The values for Dalpha indicate that Xenopus muscle fibers cannot reach Vo(2 max) in vivo because Po(2crit) can be higher than arterial Po(2) and that hypertrophied rat cardiomyocytes can become hypoxic at the maximum heart rate.  相似文献
