An important adaptation to land habitats in terrestrial isopod crustaceans is development of embryos in a fluid-filled female brood pouch, marsupium. The study brings insight into the structure and protective role of egg envelopes and cuticle renewal during ontogenetic development of Porcellio embryos and marsupial mancas. Egg envelopes cover embryos, the outer chorion until late-stage embryo and the inner vitelline membrane throughout the whole embryonic development. Egg envelopes of Porcellio have relatively simple ultrastuctural architecture compared to Drosophila egg envelopes. Exoskeletal cuticle is produced in late embryonic development by hypodermal cells of the embryo and is renewed in further development in relation to growth of developing embryos and mancas. Cuticle structure and renewal in prehatching late-stage embryos and marsupial mancas exhibit main features of cuticle in adults. Epicuticle is thin and homogenous. The characteristic arrangement of chitin-protein fibers and the dense distal layer in exocuticle are hardly discernible in prehatching embryo and distinct in marsupial mancas. Endocuticle consists of alternating electron dense and electron lucent sublayers and is perforated by pore canals in both stages. Differences from adult cuticle are evident in cuticle thickness, ultrastructure and mineralization. Signs of cuticle renewal in prehatching embryo and marsupial mancas such as detachment of cuticle from hypodermis, partial disintegration of endocuticle and assembly of new cuticle are described.  相似文献

The study of host–parasite coevolution is one of the cornerstones of evolutionary biology. The majority of fish ectoparasites belonging to the genus Dactylogyrus (Monogenea) exhibit a high degree of host specificity. Therefore, it is expected that their evolutionary history is primarily linked with the evolutionary history of their cyprinoid fish hosts and the historical formation of the landmasses. In the present study, we used a cophylogenetic approach to investigate coevolutionary relationships between endemic Cyprinoidea (Cyprinidae and Leuciscidae) from selected regions in southern Europe and their respective Dactylogyrus species. A total of 49 Dactylogyrus species including endemic and non-endemic species were collected from 62 endemic cyprinoid species in the Balkan and Apennine Peninsulas. However, 21 morphologically identified Dactylogyrus species exhibited different genetic variants (ranging from 2 to 28 variants per species) and some of them were recognized as cryptic species on the basis of phylogenetic reconstruction. Phylogenetic analyses revealed several lineages of endemic and non-endemic Dactylogyrus species reflecting some morphological similarities or host affinities. Using distance-based and event-based cophylogenetic methods, we found a significant coevolutionary signal between the phylogenies of parasites and their hosts. In particular, statistically significant links were revealed between Dactylogyrus species of Barbini (Cyprinidae) and their hosts belonging to the genera Aulopyge, Barbus and Luciobarbus. Additionally, a strong coevolutionary link was found between the generalist parasites D. alatus, D. sphyrna, D. vistulae, and their hosts, and between Dactylogyrus species of Pachychilon (Leuciscidae) and their hosts. Cophylogenetic analyses suggest that host switching played an important role in the evolutionary history of Dactylogyrus parasitizing endemic cyprinoids in southern Europe. We propose that the high diversification of phylogenetically related cyprinoid species in the Mediterranean area is a process facilitating the host switching of specific parasites among highly diverse congeneric cyprinoids.  相似文献

The cave-dwelling shrimp, Troglocaris schmidtii, has a unique mode of lipid storage. The lipid lies extracellularly in specialized compartments of the hepatopancreas, named oleospheres. The lipid is synthesized in the R-cells of the hepatopancreatic epithelium and accumulates in lipid droplets which fuse to form bigger globules. Mature lipid globules display moderately electron dense centers probably comprising triglycerides, and a broad electron dense boundary presumably consisting of lipoproteins. The globules are discharged into the lumen of the hepatopancreatic tubules by a kind of apocrine secretion. There, they coalesce to form larger masses. Finally, these lipid masses are transported into the oleospheres through a valve-like structure. The continual accumulation of lipid results in a drastic expansion of the oleospheres up to 500 μm in diameter. The absence of food in the digestive tract and the inactivity of the digestive enzyme producing F-cells indicate that digestion is suspended in the period of oleosphere formation. The curious mode of lipid storage in T. schmidtii may represent an adaptation to the extreme environmental conditions of a cave.  相似文献

The influence of exposure to lead and cadmium on serum concentrations of calcium, zinc, and copper was examined in 299 healthy Croatian men 20-55 years of age, including 143 workers in a lead battery factory and 156 control subjects. The interrelationship of blood lead, activity of delta-aminolevulinic acid dehydratase, erythrocyte protoporphyrin, blood cadmium, age, smoking habits, and alcohol consumption with respect to serum calcium, zinc, and copper levels was calculated by forward stepwise multiple regression. The results showed that chronic moderate exposure to lead decreased serum zinc, calcium and, to a lesser extent, copper levels. Exposure to cadmium occurred mostly through smoking and seemed to contribute to a decrease of serum zinc. All changes in serum concentrations of these essential elements were significant but remained within the normal range. The results suggest possible mechanisms of lead- and/or cadmium-induced adverse health effects in humans.  相似文献

Background

Despite progress in adjuvant chemotherapy in the recent decades, pancreatic and colon cancers remain common causes of death worldwide. Bacterial toxins, which specifically bind to cell surface-exposed glycosphingolipids, are a potential novel therapy. We determined the expression of globotriaosylceramide (Gb3Cer/CD77), the Shiga toxin receptor, in human pancreatic and colon adenocarcinomas.

Methodology/Principal Findings

Tissue lipid extracts of matched pairs of cancerous and adjacent normal tissue from 21 pancreatic and 16 colon cancer patients were investigated with thin-layer chromatography overlay assay combined with a novel mass spectrometry approach. Gb3Cer/CD77 was localized by immunofluorescence microscopy of cryosections from malignant and corresponding healthy tissue samples. 62% of pancreatic and 81% of colon adenocarcinomas showed increased Gb3Cer/CD77 expression, whereas 38% and 19% of malignant pancreas and colon tissue, respectively, did not, indicating an association of this marker with neoplastic transformation. Also, Gb3Cer/CD77 was associated with poor differentiation (G>2) in pancreatic cancer (P = 0.039). Mass spectrometric analysis evidenced enhanced expression of Gb3Cer/CD77 with long (C24) and short chain fatty acids (C16) in malignant tissues and pointed to the presence of hydroxylated fatty acid lipoforms, which are proposed to be important for receptor targeting. They could be detected in 86% of pancreatic and about 19% of colon adenocarcinomas. Immunohistology of tissue cryosections indicated tumor-association of these receptors.

Conclusions/Significance

Enhanced expression of Gb3Cer/CD77 in most pancreatic and colon adenocarcinomas prompts consideration of Shiga toxin, its B-subunit or B-subunit-derivatives as novel therapeutic strategies for the treatment of these challenging malignancies.  相似文献

Freshwater mussels of the Order Unionida provide important ecosystem functions and services, yet many of their populations are in decline. We comprehensively review the status of the 16 currently recognized species in Europe, collating for the first time their life‐history traits, distribution, conservation status, habitat preferences, and main threats in order to suggest future management actions. In northern, central, and eastern Europe, a relatively homogeneous species composition is found in most basins. In southern Europe, despite the lower species richness, spatially restricted species make these basins a high conservation priority. Information on freshwater mussels in Europe is unevenly distributed with considerable differences in data quality and quantity among countries and species. To make conservation more effective in the future, we suggest greater international cooperation using standardized protocols and methods to monitor and manage European freshwater mussel diversity. Such an approach will not only help conserve this vulnerable group but also, through the protection of these important organisms, will offer wider benefits to freshwater ecosystems.  相似文献

Freshwater mussels of the order Unionida have been dramatically declining globally. Despite their ecological importance, conservation of these animals has been hindered by unresolved taxonomy and a lack of data on the distribution and status of populations, especially in southern Europe. Although the Italian Peninsula has been noted as a centre of endemism and one of the major refugia of the glacial ages for several taxa, few studies have been performed on the genetic diversity of Unionida. Most importantly, the taxonomic status of several freshwater mussel populations of the Italian Peninsula is still unresolved. Here we present the first comprehensive dataset for the Unionida of the region spanning Italy and the coastal Croatian region (west of the Dinaric Alps). In total, 191 specimens were collected (85 Anodonta, 64 Unio, 17 Microcondylaea bonellii and 25 Sinanodonta woodiana) from 34 sites across the Italian Peninsula and coastal Croatian river basins for molecular identification (COI, 16S and 28S). Genetic analyses were performed to understand major phylogenetic and phylogeographic patterns. Seven species were detected: three Anodonta species (A. anatina, A. cygnea and A. exulcerata), two Unio species (U. mancus and U. elongatulus), Microcondylaea bonellii, and the invasive Sinanodonta woodiana. The presence of three endemic species (A. exulcerata, U. elongatulus and M. bonellii) confirms the importance of the region as a centre of endemism for freshwater mussels. The Apennine Mountains act as an important biogeographic barrier.  相似文献

The antibacterial effect of cationic surfactants against the pure culture of phosphate (P)-accumulating bacterium Acinetobacter junii was investigated. The estimated EC50 values of the N-dodecylpyridinium chloride (DPC) for growth inhibition was 1.4±0.5 × 10−6 mol L−1 and for the inhibition of the P-uptake rates 7.3±2.6 × 10−5 mol L−1. The estimated EC50 values of the N-cetylpyridinium chloride (CPC) for growth inhibition was 4.9±1.3 × 10−7 mol L−1 and for the inhibition of the P-uptake rates 7.7±2.9 × 10−6 mol L−1. This suggests the importance of controlling the amounts of cationic surfactants in influent of the wastewater treatment systems in order to avoid the possible failure of the biological P removal from wastewaters.  相似文献
