Strontium-90, strontium-89 and S.U. values were determined in human milk before and after the resumption of atmospheric nuclear testings in 1961, and the levels were compared to cows'' milk values reported during the same time. S.U.90 levels in human milk were approximately one-fifth of those found in cows'' milk. Assuming an average dietary intake of 11-13 S.U.90 during the period tested, the mean strontium/calcium ratio of 1.78 found in human milk represents an Observed Ratio milk-diet of approximately 0.14-0.16. Although strontium-89 was present in cows'' milk already in September 1961, it did not appear in human milk until November 1961. It seems, therefore, that there was a two-month lag period between the appearance of fresh fallout in cows'' milk and human milk. Calcium-supplement mineral preparations used by pregnant and lactating women were tested to find their strontium-89, strontium-90 and S.U. levels, because strontium isotopes, if present in these products, will be transferred to the fetus and to breast-fed infants. The degree of phosphorylation and phosphoethanolaminylation of lipid A on neisserial lipooligosaccharide (LOS), a major cell-surface antigen, can be correlated with inflammatory potential and the ability to induce immune tolerance in vitro. On the oligosaccharide of the LOS, the presence of phosphoethanolamine and sialic acid substituents can be correlated with in vitro serum resistance. In this study, we analyzed the structure of the LOS from 40 invasive isolates and 25 isolates from carriers of Neisseria meningitidis without disease. Invasive strains were classified as groups 1–3 that caused meningitis, septicemia without meningitis, and septicemia with meningitis, respectively. Intact LOS was analyzed by high resolution matrix-assisted laser desorption/ionization time-of-flight mass spectrometry. Prominent peaks for lipid A fragment ions with three phosphates and one phosphoethanolamine were detected in all LOS analyzed. LOS from groups 2 and 3 had less abundant ions for highly phosphorylated lipid A forms and induced less TNF-α in THP-1 monocytic cells compared with LOS from group 1. Lipid A from all invasive strains was hexaacylated, whereas lipid A of 6/25 carrier strains was pentaacylated. There were fewer O-acetyl groups and more phosphoethanolamine and sialic acid substitutions on the oligosaccharide from invasive compared with carrier isolates. Bioinformatic and genomic analysis of LOS biosynthetic genes indicated significant skewing to specific alleles, dependent on the disease outcome. Our results suggest that variable LOS structures have multifaceted effects on homeostatic innate immune responses that have critical impact on the pathophysiology of meningococcal infections. 相似文献

Organic-walled dinoflagellate cyst (dinocyst) and geochemical records across the Cenomanian–Turonian boundary (CTB) are presented for the NW European reference section at Eastbourne, England. Dinocyst and nannofossil fertility indexes indicate that an upwelling-driven productivity pulse accompanied a eustatic sea-level fall that preceded, by at least 40 kyr, the global rise in δ13C values that marks the onset of Oceanic Anoxic Event 2 (OAE2) and the deposition of black shales in the deep ocean. A marine productivity collapse in the latest Cenomanian is evidenced by the falling absolute and relative abundance of peridinioid dinocysts, believed to be the product of heterotrophic dinoflagellates. This biotic change accompanied transgression and sharply rising sea-surface temperatures, following an Atlantic-wide episode of short-lived cooling. Geochemical tracers provide evidence of Caribbean–Colombian plateau volcanism. The increase in water depth caused by the latest Cenomanian transgression resulted in less eutrophic waters in epicontinental seas, where CTB biotic turnover was driven largely by water-mass changes rather than oxygen depletion.
