Summary The fermentation profiles ofSporotrichum pulverulentum andDichomitus squalens showed distinct differences.D. squalens digested the substrate more slowly thanSporotrichum pulverulentum. The relative degradation rates of total organic matter and lignin also differed considerably.Whereas withS. pulverulentum the ratio was about the same throughout the whole observation period, withDichomitus squalens it altered in favour of lignin degradation.WithSporotrichum pulverulentum an optimum digestion in vitro of 40%–50% was achieved after 20 days of incubation. WithDichomitus squalens the best value (about 60%) was reached after 30 days of incubation. Increasing incubation temperatures enhanced the degradation of the substrate.As found with wheat straw, all other substrates tested (straw of rape and barley, glumes of rice) were degraded more slowly byDichomitus squalens than bySporotrichum pulverulentum. The degradation rates for oak, spruce and beech sawdust were very low compared to those for straw.Small amounts of ammonium nitrate stimulated the degradation of straw byS. pulverulentum whereas higher concentrations had an inhibitory effect. The optimum water content of the substrate, measured by decomposition of total organic matter and lignin and by in vitro digestibility, was between 50 and 100 ml of water/25 g substrate. Higher and lower water contents had an unfavourable effect.Varying the pore size of the substrate by using milled straw of defined particle size had no influence on the 6 parameters tested under the given experimental conditions.The best method to supress potential competitors was to heat the substrate to 90°C for 24 h.  相似文献

The cyclodipeptides cyclo(L -alanyl-L -phenylalanyl) and cyclo(D -alanyl-L -phenylalanyl) were synthesized with various atoms substituted by the isotopes 15N and 13C. Thus, the coupling constants 15N–1H, 13C–1H, and 13C–15N could be obtained, in addition to the commonly used 1H–1H constants. The applicability of these coupling constants for obtaining conformational information on side chains and substituted 2,5-piperazinedione rings is discussed.  相似文献

Loss of genome stability leads to reduced fitness, fertility and a high mutation rate. Therefore, the genome is guarded by the pathways monitoring its integrity and neutralizing DNA lesions. To analyze the mechanism of DNA damage induction by cytidine analog zebularine, we performed a forward-directed suppressor genetic screen in the background of Arabidopsis thaliana zebularine-hypersensitive structural maintenance of chromosomes 6b (smc6b) mutant. We show that smc6b hypersensitivity was suppressed by the mutations in EQUILIBRATIVE NUCLEOSIDE TRANSPORTER 3 (ENT3), DNA METHYLTRANSFERASE 1 (MET1) and DECREASE IN DNA METHYLATION 1 (DDM1). Superior resistance of ent3 plants to zebularine indicated that ENT3 is likely necessary for the import of the drug to the cells. Identification of MET1 and DDM1 suggested that zebularine induces DNA damage by interference with the maintenance of CG DNA methylation. The same holds for structurally similar compounds 5-azacytidine and 2-deoxy-5-azacytidine. Based on our genetic and biochemical data, we propose that zebularine induces enzymatic DNA–protein crosslinks (DPCs) of MET1 and zebularine-containing DNA in Arabidopsis, which was confirmed by native chromatin immunoprecipitation experiments. Moreover, zebularine-induced DPCs accumulate preferentially in 45S rDNA chromocenters in a DDM1-dependent manner. These findings open a new avenue for studying genome stability and DPC repair in plants.  相似文献

Apamin, an 18 amino acid peptide with two disulfide bonds, elicits specific T cell proliferative responses in H-2d and H-2b mouse strains. We evaluated the processing requirement of this compact peptide by accessory cells for presentation to apamin-reactive T hybridoma cells (THC) by analyzing the IL-2 responses of 16 THC from apamin-primed BALB/c or C57BL/6 mice, to various forms of either native or chemically synthesized apamin analogs. These included: unfolded peptides (whose four sulfhydryl groups were blocked by acetamidomethyl residues), N-and/or C-truncated peptides, and an analog with a single amino acid substitution at position 10. Assessment of the Ag-specific THC responses in the presence of either live or formaldehyde-prefixed APC indicated the following: 1) all THC stringently required Ag processing; 2) in 8 of 16 cases, the simple unfolding of apamin was sufficient to eliminate the need for Ag processing, or even induced increased THC IL-2 responses (other cells required further antigenic alterations in addition to unfolding, or rare processing steps dependent on the integrity of the two disulfide bonds); and 3) for most THC, the Leu10 and the N terminus arm of apamin were shown to be critical for expression of the epitopes involved in T cell recognition. These data indicate that apamin, a natural peptide having an appropriate size for T cell triggering, acquires its antigenic conformation after a processing by APC which primarily involves an alteration of a disulfide bond-dependent peptide folding.  相似文献

Several studies focusing on brain irradiation are in progress. Reflecting updates of relevant outcomes in palliative treatment of patients suffering from brain metastases, the primary objective of these studies is the evaluation of neurocognitive function and quality of life. Improvements of technology in radiation oncology allows us to spare the hippocampal region while appropriately irradiating other parts of brain tissue. Irradiation of the hippocampus region is likely to lead to manifestations of adverse events with a subsequent impact on patient''s quality of life, which is in fact an improper approach in palliative medicine. Ongoing studies evaluate results of hippocampus avoiding radiotherapy compared to standard whole brain radiotherapy. Incorporation of neurocognitive function assessment may result in the confirmation of superiority of sparing the region of hippocampus and thus change current style of providing brain irradiation.  相似文献

Understanding how animals respond to disturbance by investigators is essential for a fair assessment of the presence of bias in routinely used research protocols. It is also an essential prerequisite for anyone interested in animal welfare and ethically sound research. Here, we utilize an automatic logging system to monitor nest box use by PIT‐tagged edible dormice, Glis glis, after standard handling procedures applied during a regular nest‐box monitoring programme. The edible dormouse is an arboreal nocturnal rodent that relies on tree hollows as daytime den sites. We assessed the effect of disturbance on dormice in two ways: whether it affected the decision of an individual to stay in the same den site for a subsequent day and whether it affected the timing of the individual's nocturnal emergence from the den site. We found handling had a strong negative effect on short‐term den use. In addition, females and sexually active individuals were more likely to spend the following day in the nest box. Individuals that had left the den site after our handling returned to them after an average of 4 d. Handling did not have a significant effect on the period of absence, but reproductively active animals returned on average after 3 d, while reproductively quiescent animals returned after more than 5 d. Manipulation did not have a significant effect on the initiation of nocturnal activity. Our study suggests that disturbance by investigators may modify certain aspects of animal behaviour, but this effect is likely to be short term and does not appear to impair the efficacy of routinely practiced capture‐mark‐recapture field protocols.  相似文献
