The solution structure of the complex containing the isolated kringle 2 domain of human plasminogen (K2Pg) and VEK-30, a 30-amino acid residue internal peptide from a streptococcal M-like plasminogen (Pg) binding protein (PAM), has been determined by multinuclear high-resolution NMR. Complete backbone and side-chain assignments were obtained from triple-resonance experiments, after which structure calculations were performed and ultimately refined by restrained molecular simulation in water. We find that, in contrast with the dimer of complexes observed in the asymmetric unit of the crystal, global correlation times and buoyant molecular weight determinations of the complex and its individual components showed the monomeric nature of all species in solution. The NMR-derived structure of K2Pg in complex with VEK-30 presents a folding pattern typical of other kringle domains, while bound VEK-30 forms an end-to-end α-helix (residues 6–27) in the complex. Most of the VEK-30/K2Pg interactions in solution occur between a single face of the α-helix of VEK-30 and the lysine binding site (LBS) of K2Pg. The canonical LBS of K2Pg, consisting of Asp54, Asp56, Trp60, Arg69, and Trp70 (kringle numbering), interacts with an internal pseudo-lysine of VEK-30, comprising side-chains of Arg17, His18, and Glu20. Site-specific mutagenesis analysis confirmed that the electrostatic field formed by the N-terminal anionic residues of the VEK-30 α-helix, viz., Asp7, and the non-conserved cationic residues of K2Pg, viz., Lys43 and Arg55, play additional important roles in the docking of VEK-30 to K2Pg. Structural analysis and kringle sequence alignments revealed several important features related to exosite binding that provide a structural rationale for the high specificity and affinity of VEK-30 for K2Pg.  相似文献

Mitochondrial complex II (CII) has been recently identified as a novel target for anti-cancer drugs. Mitochondrially targeted vitamin E succinate (MitoVES) is modified so that it is preferentially localized to mitochondria, greatly enhancing its pro-apoptotic and anti-cancer activity. Using genetically manipulated cells, MitoVES caused apoptosis and generation of reactive oxygen species (ROS) in CII-proficient malignant cells but not their CII-dysfunctional counterparts. MitoVES inhibited the succinate dehydrogenase (SDH) activity of CII with IC(50) of 80 μM, whereas the electron transfer from CII to CIII was inhibited with IC(50) of 1.5 μM. The agent had no effect either on the enzymatic activity of CI or on electron transfer from CI to CIII. Over 24 h, MitoVES caused stabilization of the oxygen-dependent destruction domain of HIF1α fused to GFP, indicating promotion of the state of pseudohypoxia. Molecular modeling predicted the succinyl group anchored into the proximal CII ubiquinone (UbQ)-binding site and successively reduced interaction energies for serially shorter phytyl chain homologs of MitoVES correlated with their lower effects on apoptosis induction, ROS generation, and SDH activity. Mutation of the UbQ-binding Ser(68) within the proximal site of the CII SDHC subunit (S68A or S68L) suppressed both ROS generation and apoptosis induction by MitoVES. In vivo studies indicated that MitoVES also acts by causing pseudohypoxia in the context of tumor suppression. We propose that mitochondrial targeting of VES with an 11-carbon chain localizes the agent into an ideal position across the interface of the mitochondrial inner membrane and matrix, optimizing its biological effects as an anti-cancer drug.  相似文献

The genetic structure of red deer populations is under strong influence of human activities such as game management and habitat fragmentation. Using multilocus genotypes from 193 geo-referenced individuals, we evaluated the population genetic structure of three red deer populations in Croatia. The effect of habitat fragmentation on genetic structure was tested using Bayesian non-spatial and spatial clustering methods. Our results indicate levels of genetic diversity similar to the ones previously reported by other authors for stable and appropriately managed populations within all populations analyzed. The spatial clustering model was able to detect the effect of habitat fragmentation on population differentiation, supporting the use of spatially explicit methods in landscape genetics, and giving important guidelines for future road planning.  相似文献

Zusammenfassung Elektronenmikroskopische Replika-Präparate (Methylmethakrylat, Bedampfung mit Chrom-Beryllium) von nativen Schliffflächen von Dentin und mit HNO3, EDTA, KOH-Glyzerin, Äthylendiamin und Inzineration bei 300° C und 1100° C behandelte Präparate zeigen, daß sich die peritubuläre Zone von dem intertubulären Dentin in der Resistenz gegen die einwirkenden Mittel unterscheidet. Durch Inzineration bei 300° C wird sie nicht angegriffen. Säure, EDTA, KOH-Glyzerin, Äthylendiamin und Inzineration bei 1100° C greifen sie verschieden intensiv an. Die peritubuläre Zone unterscheidet sich dann häufig nicht mehr von der intertubulären Grundsubstanz. Dadurch, daß beim Polieren, bei teilweiser Inzineration und manchmal infolge Extraktion eine Fibrillenstruktur in Erscheinung tritt — wahrscheinlich durch Entfernen der Fibrillen und Darstellung des Mineralgerüstes, welches die Fibrillen einbettet — gewinnen wir einen Einblick in den Aufbau der peritubulären Zone. Anschließend wird der Strukturunterschied der peritubulären Zone dadurch bestimmt, daß die Fibrillen, die das Dentingerüst bilden, sich daselbst in feinere aufspalten, einen anderen Verlauf nehmen und von einer größeren Menge von homogener Kittsubstanz umgeben werden, die infolgedessen intensiver kalzifiziert. Obwohl die peritubuläre Zone der Dentingrundsubstanz angehört und nicht der periphere Anteil des Odontoblastenfortsatzes (Frank 1956) ist, scheint dieser bei ihrer Genese eine besondere Rolle zu spielen.  相似文献

A metabolic network consisting of 48 reactions was established to describe intracellular processes during growth and poly-3-hydroxybutyrate (PHB) production for Cupriavidus necator DSM 545. Glycerol acted as the sole carbon source during exponential, steady-state cultivation conditions. Elementary flux modes were obtained by the program Metatool and analyzed by using yield space analysis. Four sets of elementary modes were obtained, depending on whether the pair NAD/NADH or FAD/FADH2 contributes to the reaction of glycerol-3-phosphate dehydrogenase (GLY-3-P DH), and whether 6-phosphogluconate dehydrogenase (6-PG DH) is present or not. Established metabolic network and the related system of equations provide multiple solutions for the simultaneous synthesis of PHB and biomass; this number of solutions can be further increased if NAD/NADH or FAD/FADH2 were assumed to contribute in the reaction of GLY-3-P DH. As a major outcome, it was demonstrated that experimentally determined yields for biomass and PHB with respect to glycerol fit well to the values obtained in silico when the Entner–Doudoroff pathway (ED) dominates over the glycolytic pathway; this is also the case if the Embden–Meyerhof–Parnas pathway dominates over the ED.  相似文献

 We present an improved method for microphotometric measurement of enzyme activity in muscle fibres by determining maximum reaction rates using computer-assisted image analysis. The method was used to determine absolute and relative activities of succinate dehydrogenase (SDH) in 4801 whole-fibre cross-sections of rabbit tibialis anterior muscles stimulated at low frequency (10 Hz) for different time periods of up to 50 days. Measurements were performed on cross-sections of composite blocks from stimulated and contralateral control muscles. The validity of the method was checked by determining SDH activity in homogenates of the same muscles using a standard photometric assay. Both methods yielded similar results for the time-dependent increases of SDH activity in response to chronic low-frequency stimulation. Significant increases in catalytic activity were detected by the two methods only in muscles stimulated for longer than 8 days. According to homogenate measurements, overall SDH activity was 7.4-fold elevated in 50-day-stimulated total muscles. Depending on whether or not measurements were corrected for the so-called nothing-dehydrogenase activity, the average increase in microphotometrically determined SDH activity amounted to approximately 8-fold or 10-fold, respectively. Microphotometry revealed pronounced scattering of SDH activities within the fibre populations of both normal and stimulated muscles. The heterogeneity of the fibre population with regard to SDH activity increased in long-term stimulated muscles ranging between 5-fold and 15-fold elevations. Accepted: 2 September 1996  相似文献

The evolution of ten Robertsonian (Rb) races of the house mouse (Mus musculus domesticus) in the Rhaetian Alps of northern Italy and southern Switzerland is reconsidered. The mechanisms of centric fusion, zonal raciation and, for the first time, whole-arm reciprocal translocation (WART), are used in this non-mathematical approach to produce a phylogenetic tree (using chromosome fusions as characters) with the smallest number of steps. The shortest tree that we found (16 steps) is at least two to nine mutations shorter than previously published models. Three other trees (17 or 18 steps) are also considered, since they are geographically more sensible. In general, these four scenarios correspond more closely to the present distributions of the ten Rb races than previous trees. Our results suggest that zonal raciation and WARTs play an important role in the evolution of Rb races of the house mouse.  相似文献
