Purpose: Deficiency of the mannan-binding lectin (MBL) pathway of innate immunity is associated with increased susceptibility to infections. In patients with colorectal cancer (CRC), postoperative infection is associated with poor prognosis. The aim of the present study was to evaluate (1) the relation between the MBL pathway and postoperative infectious complications and survival of patients resected for CRC, and (2) the role of MBL in acute phase response compared to C-reactive protein (CRP). Methods: Preoperative MBL concentration, MBL-associated serine protease (MBL/MASP) activity and CRP were determined in serum from 611 patients and 150 healthy controls. The patients were observed for 8 years. Postoperative infections, recurrence and survival were recorded. Results: The MBL pathway components were increased in the patients compared with the healthy controls (p<0.0001). Low MBL levels were predictive of pneumonia (p=0.01), and pneumonia (n=87) was associated with poor survival (p=0.003; HR=1.5; 95% CI, 1.1 to 1.9). MBL and MBL/MASP activity showed no correlation with CRP (Spearmans =0.02; 95% CI, –0.06 to 0.10). Conclusion: Low preoperative MBL levels are predictive of pneumonia, which is associated with poorer survival. MBL concentration and MBL/MASP activity was not predictive of other postoperative infections or long-term prognosis, and showed no correlation with CRP.  相似文献

Campylobacter jejuni is a major cause of bacterial food-borne infection in the industrial world. There is evidence that C. jejuni is present in eggs and hatchery fluff, opening the possibility for vertical transmission from hens to progeny. Poultry operations in Iceland provide an excellent opportunity to study this possibility, since breeding flocks are established solely from eggs imported from grandparent flocks in Sweden. This leaves limited opportunity for grandparents and their progeny to share isolates through horizontal transmission. While Campylobacter was not detected in all grandparent flocks, 13 of the 16 egg import lots consisted of eggs gathered from one or more Campylobacter-positive grandparent flocks. No evidence of Campylobacter was found by PCR in any of the 10 relevant quarantine hatchery fluff samples examined, and no Campylobacter was isolated from the parent birds through 8 weeks, while they were still in quarantine rearing facilities. After the birds were moved to less biosecure rearing facilities, Campylobacter was isolated, and 29 alleles were observed among the 224 isolates studied. While three alleles were found in both Sweden and Iceland, in no case was the same allele found both in a particular grandparent flock and in its progeny. We could find no evidence for vertical transmission of Campylobacter to the approximately 60,000 progeny parent breeders that were hatched from eggs coming from Campylobacter-positive grandparent flocks. If vertical transmission is occurring, it is not a significant source for the contamination of chicken flocks with Campylobacter spp.  相似文献

Reactive oxygen intermediates induce cell injury in a variety of pathophysiological conditions. Human umbilical cord vein endothelial cell (HUVEC) cultures were exposed to 1 or 200 microM H2O2 for 15 min, and observed after 15 min, or 1, 4, 24, or 120 h. Factor VIII and the cytoskeletal proteins vimentin and tubulin were visualized immunocytochemically. Release of lactate dehydrogenase (indices of cell membrane injury) did not increase after H2O2 exposure; nor was cellular expression of factor VIII affected. 200 microM H2O2 induced cell contraction after 15 min which disappeared after 1 and 4 h, but was evident again after 24 h. Immediately after exposure, the filamentous structure of vimentin and tubulin disappeared, but normalized after 1 h. After 120 h, the cytoskeleton filaments were coarsened and disorganized, and an abundance of multinucleated giant cells were observed. Catalase (150 U/ml) abolished all effects of H2O2. One microM H2O2 did not induce any changes in HUVEC. Thus, the present concentrations of H2O2 did not induce cell necrosis or altered expression of factor VIII. Early, reversible cell contraction and depolymerization of cytoskeletal proteins were observed, followed by a delayed contraction and cell atypia after 200 microM H2O2.  相似文献

While it has been widely proven that many lichens are extremely freeze-tolerant in the dry state, little is known about how moist lichens respond to freezing under oxic and anoxic conditions. In circumpolar areas where lichens are an important component of boreal and Arctic ecosystems, winter climate is changing, leading to increased frequency of winter thaw and ground-icing events. It is imperative to elucidate in further detail how northern vegetation responds to being encapsulated in ice. A winter icing simulation experiment was therefore undertaken, encapsulating two reindeer lichens (Cladonia stellaris and Cladonia rangiferina) and two epiphytic lichens (Parmeliopsis ambigua and Melanohalea olivacea) in ice at temperatures just below freezing for a maximum period of 98 d. Photosynthetic and chlorophyll fluorescence rates decreased strongly, clearly suggesting that the algal partner of the lichens was dying. Low but detectable respiration rates indicate that the fungal partner maintained some physiological activity. Ethanol accumulated in the lichens during ice encapsulation as a result of anaerobic respiration. The algae probably were dying from a combination of depletion of carbon reserves and toxic levels of ethanol and other stress metabolites. This experiment shows that boreal and Arctic-alpine lichens are sensitive to a warmer and more fluctuating winter climate. Increasing frequency of winter icing events may therefore have extensive and hitherto unknown effects on lichen-dominated ecosystems, their herbivores and the indigenous peoples whose livelihoods largely depend on them.  相似文献

Altered gene expression of the proto-oncogenes c-fos and c-myc is associated with differentiation of the human promyelocytic leukemia (HL-60) cells. We studied changes of cyclic AMP levels and c-fos and c-myc mRNA levels after stimulation with theophylline and theophylline plus isoproterenol. Reduced c-fos and c-myc mRNA levels were detected, but the reduction could not, however, be related to the observed alternations in cyclic AMP concentrations. Expression of c-jun and c-Ha-ras was not affected under these conditions.  相似文献

Climate change impacts are not uniform across the Arctic region because interacting factors causes large variations in local ecosystem change. Extreme climatic events and population cycles of herbivores occur simultaneously against a background of gradual climate warming trends and can redirect ecosystem change along routes that are difficult to predict. Here, we present the results from sub‐Arctic heath vegetation and its belowground micro‐arthropod community in response to the two main drivers of vegetation damage in this region: extreme winter warming events and subsequent outbreaks of the defoliating autumnal moth caterpillar (Epirrita autumnata). Evergreen dwarf shrub biomass decreased (30%) following extreme winter warming events and again by moth caterpillar grazing. Deciduous shrubs that were previously exposed to an extreme winter warming event were not affected by the moth caterpillar grazing, while those that were not exposed to warming events (control plots) showed reduced (23%) biomass from grazing. Cryptogam cover increased irrespective of grazing or winter warming events. Micro‐arthropods declined (46%) following winter warming but did not respond to changes in plant community. Extreme winter warming and caterpillar grazing suppressed the CO2 fluxes of the ecosystem. Evergreen dwarf shrubs are disadvantaged in a future sub‐Arctic with more stochastic climatic and biotic events. Given that summer warming may further benefit deciduous over evergreen shrubs, event and trend climate change may both act against evergreen shrubs and the ecosystem functions they provide. This is of particular concern given that Arctic heath vegetation is typically dominated by evergreen shrubs. Other components of the vegetation showed variable responses to abiotic and biotic events, and their interaction indicates that sub‐Arctic vegetation response to multiple pressures is not easy to predict from single‐factor responses. Therefore, while biotic and climatic events may have clear impacts, more work is needed to understand their net effect on Arctic ecosystems.  相似文献

The genetic diversity of metapopulations is influenced not only by the effective sizes (N e ) of individual subpopulations, but also by the total effective size of the metapopulation (meta-N e ). We estimated meta-N e of four neighbouring Atlantic salmon populations connected by gene flow using genetic estimates of subpopulation N e s and migration rates derived from capture–recapture data. The meta-[^(N)]e meta{\hbox{-}}\hat{N}_{e} was lower than the sum of [^(N)]e \hat{N}_{e} s of the subpopulations, suggesting that genetic diversity harboured by the four river salmon metapopulation is lower than what would have been expected by viewing individual subpopulations separately. In addition, meta-[^(N)]e meta{\hbox{-}}\hat{N}_{e} was found to be sensitive to changes in [^(N)]e \hat{N}_{e} of the subpopulation from which net emigration rate was largest, so as that the genetic diversity of the metapopulation would be best preserved by avoiding any reductions in N e of this subpopulation. Yet, this subpopulation is the one that has historically—and still is—experiencing the highest exploitation rate in the metapopulation system.  相似文献
